For this study, the biological functions of Verbascum thapsus (VT) methanol extract were determined in vitro. The plant's methanol extract was created through the maceration process. The phytochemical composition of plant extracts was investigated using liquid chromatography-electrospray ionization tandem mass spectrometry. The antioxidant capacity of the extract was determined using the 2,2-diphenyl-1-picrylhydrazil (DPPH radical) and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS radical). Cell lines Caco-2 (human colorectal adenocarcinoma cells), LNCaP (Lymph Node Carcinoma of Prostate), and HEK293 (Human embryonic kidney 293 cells) were used to model colon, prostate, and non-cancerous cells. The cytotoxic activity of the plant extract on the proliferation of these cells was determined by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) assay protocol. VT extract showed moderate DPPH and ABTS radical scavenging activities at 30 mg/ml concentration. With this, VT extract was determined to inhibit acetylcholinesterase (AChE) enzyme and had strong cytotoxic activity on cancerous cell lines. In addition, our findings clearly showed that the plant extract had greater cytotoxic activity on the viability of cancerous cells compared to non-cancerous (Human embryonic kidney cells; HEK293) cells. The current findings showed that V. thapsus might be a promising anti-cancer medication candidate for the treatment of human colorectal adenocarcinoma and colon cancer, as well as a good source antioxidans. To the extent of our knowledge, no previous study in literature has reported acute MP intoxication in children. Aim: Our aim was to determine the clinical effects and treatment strategies of MP poisoning in children.Materials and methods: We retrospectively reviewed the medical records of children <18 years of age with MP intoxication who were followed up in the Intensive Care Unit between January 2016 and April 2018 in our center. Results: Forty-one patients (M/F= 25/16) were included in this study. The mean age was 13.2±22.4 months (age range: 7 to 30 months). The patients presented with vomiting (n=23, 56%), cough (n=17, 41.5%), loss of consciousness (n=11, 26.8%), respiratory distress (n=6, 14.6%), convulsion (n=7, 17%), cyanosis (n=1, 2.4%), and abdominal pain (n=1, 2.4%) following oral ingestion of the substance. On their physical examination all patients with convulsion were in a comatose state. Thirty-two patients (78%) had tachycardia; 15 patients (36.5%) had pharyngeal hyperemia; and three (7.3%) had hiccups. Although the female patients had a lesser rate of symptoms than male patients, the difference was not statistically significant. Conclusions: Our aim was to inform the doctors about the clinical picture that develops after taking this substance and contribute to the understanding of the treatment approach.Article Assessment of renal function in obese and overweight children with ngal and kim-1 biomarkers(Nutricion Hospitalaria, 2020) Unsal, Velid; Gül, Ali; Yılmaz, Resul; Özmen, Zeliha Cansel; Demir, Osman; Gümüşer, RüveydaAim and background:the incidence of obesity has increased among children, and obesity has been considered an independent risk factor for chronic kidney disease. We aimed to determine the degree of kidney function impairment by evaluating urine neutrophil gelatinase-associated lipocalin (NGAL) and kidney injury molecule-1 (KIM-1) levels. Materials and methods: in total, 15 obese, 26 overweight, and 26 control adolescents aged 10 to 16 years were enrolled into the study. Urine samples were evaluated for NGAL and KIM-1 levels using enzyme-linked immunosorbent assay kits. We investigated the association between obesity and related comorbidities with urinary NGAL and KIM-1 excretion. Results: no significant differences were noted between the obese, overweight, and control groups in urinary NGAL and KIM-1 excretion (p = 0.327 and p = 0.917, respectively). In the obese and overweight groups urinary NGAL levels were 50.39 [30.88-74.22] in females and 26.67 [23.24-45.59] in males (p = 0.013). Also, urinary NGAL levels were increased in obese and overweight adolescents with LDL dyslipidemia at 64.12 [30.98-114.32] as compared to those without LDL dyslipidemia: 39.51 [25.59.56.37] (p = 0.024). Furthermore, a correlation was observed between insulin and homeostasis model assessment of insulin resistance levels with the NGAL/creatinine ratio in the overweight group (r = 0.515; p = 0.008, and r = 0.483; p = 0.014, respectively). Such correlation was not found in the obese group. Conclusion: the effect of obesity on renal function could not be determined in children. A longer exposure may be required for obesity-induced disruption of renal function in children. Renal function may be disrupted by dyslipidemia in obese adolescents. Furthermore, obesity impaired renal function in female adolescents. The normalization of these urinary markers as related to urine creatinine should be discussed.Presentation Assessment Of The Antimicrobial, Antioxidant And Cytotoxic Activities Of The Wild Edible Mushroom Pleurotus Eryngii From Hakkari, Turkey(2017) Acay, Hilal; Dündar, Abdurrahman; Keskin, Cumali; Kaçar, SemraAbstract: Mushroom is an important source of natural compounds with acknowledged bioactivity. Pleurotus eryngii, in particular, is widely recognized for its organoleptic quality and favorable healthy effects, being commercially produced in great extent. The present study is just for evaluating the antioxidant capacity (scavenging DPPH radicals, reducing power, Total antioxidant activity by β-carotene-linoleic acid metod, Chelating effect on ferrous ions, superoxide anion radical scavenging activity ), antimicrobial effects (by using the disc diffusion method) and insufficiently studied cytotoxic activity (using PC-3 cell lines) of Pleurotus eryngii by using various solvent systems. For this reason; obtained extracts of the mushroom applying hexane, ethylacetate and methanol respectively were tested with in its biologic activities. As a result, The highest total antioxidant activity by β -carotene-linoleic acid metod was identified in methanol extract (63.62 mg /ml). The highest DPPH scavenging activity was obtained from ethyl acetate extract with 88.81 mg/ml. EDTA is used as standard in methal chelating activity. The highest metal chelating activity of Pleurotus eryngii identified in hexane (88,1 mg /ml) and ethylacetate (88.84 mg /ml) extracts. Additionally, in most of studied tests antioxidant activity of the mushroom increased with increasing concentration. On the other hand, the highest antimicrobial activity was obtained against Pseudomonas aeruginosa ATCC 27853 (11 mm) in ethyl acetate extract. However, the best antimicrobial activities were observed in hexane and methanol extracts for other tested microorganism. It was not determined any antimicrobial activity against Escherichia coli ATCC 25922. It has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Ethyl acetate extract showed significant inhibitory value at the concentrations of 530 µg/ml (86.20 %) against PC-3 cell lines. Using methanol extract, the maximun inhibition was observed at concentration of 680 µg/ml with value of 51.86 %. Overall, the mushroom used as a daily nutrient could be a source for new drug developments and a kind of treatment in cancer therapies, and also, organic extracts of Pleurotus eryngii may contain substances that stimulate bioactivityPresentation Atatürk Baraj Gölü’nden Dört Tatlısu Balığının Yağ Asidi İçeriklerinin Tespiti.(2012) Kaçar, Semra; Başhan, Mehmet; Oymak, AhmetBu çalışmada Eylül 2008 tarihinde Atatürk Baraj Gölü'nden toplanan Capoeta trutta, Liza abu, Cyprinion macrostomum ve Achanthobrama marmid'in kas dokusu yağ asidi kompozisyonu araştırılmıştır. In particular, some components in certain foods can directly affect human health by showing biological activity (bioactive) in the body. In this context, extra virgin olive oil is a food that has recently gained importance due to its positive effects on health. In this study, phenolic compound profiles (oleuropein, hydroxytyrosol, tyrosol, catechin, pinoresinol, luteolin, apigenin, caffeic acid, vanillic acid, t-ferulic acid, and p-coumaric acid), total phenolic content (TFC) and total antioxidant capacity (TAC) of 5 different extra virgin olive oils obtained from different olive varieties (Domat, Uslu, Arbequina, Taggiasca, Koroneiki) were investigated. The amounts of the individual phenolic compound in the oil samples were determined by a high-pressure liquid chromatography system (HPLC), while TFC and TAC were determined by using spectrophotometric methods. Hydroxytyrosol, as the major phenolic compound, was found at the highest content (129.375 mg/kg) in Koroneiki olive oil, whereas its lowest (14.485 mg/kg) content was found in Domat olive oil (P<0.05). Koroneiki olive oil also showed the highest content for tyrosol, while Uslu and Arbequina olive oils had the lowest contents for this compound (P<0.05). For pinoresinol, the maximum contents were found in Domat (16,817 mg/kg) and Koroneiki (15,160 mg/kg) samples, whereas the Uslu oils (5,260 mg/kg) exhibited the minimum content. Phenolic acids were found at minor levels in all samples. According to the spectrophotometric results, although the TFC level in Koroneiki olive oil was the highest (294.822 mg gallic acid equivalent (GAE)/kg), this value was not statistically different from the values in Domat (259.950 mg GAE/kg) and Uslu (284.267 mg GAE/kg) oils (P>0.05). Similarly, the difference between the TFC of Arbequina (208.962 mg GAE/kg) and Taggiasca (219.526 mg GAE/kg) oils were not significant (P>0.05). The results of TAC demonstrated that Koroneiki oil showed the highest (110.396 mg trolox equivalent (TE)/kg) value for antioksidan aktivity, while Taggiasca oil showed the lowest (58.465 mg TE/kg) value (P<0.05). The results of this study show that the investigated bioactive properties can be used as a distinguishing feature for olive oil from different olive varieties.Article Biosynthesis of silver nanoparticles from Arum dioscoridis plant leaf aqueous extract: anticancer and antimicrobial properties(Gültekin ÖZDEMİR, 2023) İpek, Polat; Baran, Mehmet Fırta; Yıldız, Reşit; Hatipoğlu, Abdulkerim; Yıldız, ReşitThis study was carried out to synthesize silver nanoparticles (AgNPs) from Arum dioscoridis (AD) leaf extract and to investigate the cytotoxic and antipathogenic effects of them. The plant material had a reducing and stabilizing effect on the synthesized nanomaterial. During the plant-mediated synthesis of nanomaterials, no substances that would cause environmental pollution were used. For the structural characterization of AD-AgNPs, Ultraviolet-visible (UV-vis) Spectroscopy, Field Emission Scanning Electron Microscopy (FE-SEM), Electron Dispersive X-ray (EDX) Spectroscopy, Fourier Transform Infrared (FT-IR) Spectroscopy, Transmission Electron Microscopy (TEM), X-ray Diffractometer (XRD), Atomic Force Microscopy (AFM) and Zetasizer analyses were performed. The produced AgNPs showed maximum surface plasmon resonance at 431.67 nm and had mostly spherical morphology. The zeta potential value of the nanomaterial was -9.76 mV and the average powder crystal size was 31.48 nm. The minimum inhibitory concentration (MIC) values (mg/L) of AD-AgNPs on Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans were 0.25, 2.00, 0.125, 4.00, and 1.00, respectively. After 24 and 48 hours of application by MTT [3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazolium bromid] assay, the half-maximal inhibitory concentrations (IC50: μg/mL) of AD-AgNPs on human colon adenocarcinoma cell (CACO-2), human breast cancer cell (MCF-7), glioblastoma multiforme cell (T98-G), and healthy human umbilical vein endothelial cell (HUVEC) lines were determined as 2.977, 2.801, 5.694, 4.392; 2.115, 2.300, 2.612, 4.091, respectively.Article Changes in volatile compounds, sugars and organic acids of different spices of peppers (Capsicum annuum L.) during storage(FOOD CHEMISTRY, 2020) Korkmaz, Aziz; Atasoy, Ahmet Ferit; Hayaloglu, Ali AdnanChanges in sugars, organic acids and volatile compounds (VC) of red pepper flakes (RPF), traditional (TRI), and industrial (INI) isot peppers were evaluated during one year storage at the room condition. The changes in the flavor components were significantly affected by the production methods and storage time. Glucose content decreased gradually along storage and reduced by about 21.23, 47.22 and 56.65% for TRI, INI and RPF, respectively. However, fructose decreased significantly only in RPF (11.29%). Citric and succinic acids exhibited slight changes, but malic acid showed an increasing trend, especially in RPF (4-fold). Most of the VC in all samples decreased or disappeared after storage. The major quantitative losses in these compounds were found in TRI during the first 3 months as 81.76%. The storage was found to be caused deterioration flavor properties in red pepper spices and revealed the importance of appropriate storage conditions.Article Characterization and Comparison of Extra Virgin Olive Oils of Turkish Olive Cultivars(Molecules, 2023) Korkmaz, AzizExtra virgin olive oils (EVOOs) obtained from five Turkish olive cultivars widely produced in the Aegean and Marmara regions were investigated based on their total antioxidant capacity (TAC), total phenolic content (TPC), pigment contents, fatty acid (FA) profiles, phenolic compounds (PC), volatile compounds (VC), and sensory properties. The results showed that all properties of EVOO samples were significantly affected by the olive cultivar used. The pigment contents in Ayvalık (9.90 mg·kg−1) and Uslu (9.00 mg·kg−1) oils were higher than the others (p < 0.05). The greatest values for oleic acid (74.13%) and TPC (350.6 mg·kg−1) were observed in Gemlik and Domat oils, respectively (p < 0.05). Edincik oil showed the maximum hydroxytyrosol content (48.022 mg·kg−1) and TAC value (515.36 mg TE·kg−1) (p < 0.05). The Edincik, Domat, and Uslu oils were significantly not different for the total content of C6 compounds derived by lipoxygenase, which are the main volatiles responsible for the typical aroma of EVOOs (p > 0.05). Domat oil also exhibited the highest scores for bitterness and pungency perceptions (p < 0.05). The fruitiness scores of the oil samples (except for Ayvalık oil) were close to each other, even if they were statistically different (p < 0.05). Principal component analysis (PCA) indicated that the Ayvalık oil was separated from the others due to its poor-quality characteristics. As a result, it can be stated that Domat olive oil has better quality than the othersArticle Clinical and Cytokine Profile of Children With COVID-19: A Report From Turkey(Cureus, 2023) Unsal, Velid; Tahir Dalkıran, Emine M Kara, Velid Ünsal, Sadık Yurttutan, Sevcan İpek, Besra Dağoğlu, Yaşar KandurBackground We aimed to analyze the expression of infection-related biomarkers and inflammatory cytokines in laboratory-confirmed cases and compare the differences between clinically severe and non-severe ones. Method We randomly selected 35 patients who were hospitalized with the diagnosis of coronavirus disease 2019 (COVID-19). Blood serum was obtained at the time of admission to the hospital, on the third to the fifth day, and at the time of discharge. Result The median age of our patients was 56.5±69.7 months (range: 1-205 months). The mean pro-B-type natriuretic peptide (pro-BNP) was significantly higher at the time of admission than on the third to the fifth day of illness. The mean pro-B-type natriuretic peptide levels at three time points were significantly higher in patients with severe cases than in mild-moderate cases. However, there was no significant difference between the clinical severity with regard to the cytokine levels at disease onset and recovery. Conclusion In the study, it was shown that cytokines play an important role in the pathogenesis of COVID-19. Therefore, it may be beneficial to use agents such as tocilizumab in the treatment.Article Clinical Relevance of Preoperative Neutrophil to Lymphocyte and Platelet to Lymphocyte Ratio in Renal Cell Carcinoma(Journal of Urological Surgery, 2018) Kölükçü, Engin; Kılıç, Şahin; Atılgan, Doğan; Parlaktaş, Bekir; Ceylan, Salih; Unsal, Velid; Etikan, İlkerObjective: The association of preoperative neutrophil-to-lymphocyte ratio (NLR) and platelet-to-lymphocyte ratio (PLR) with postoperative tumor stage and Fuhrman nuclear grade was investigated in patients with renal cell carcinoma (RCC). Materials and Methods: Data of 123 patients, who were operated due to RCC, in our clinic was analysed. NLR and PLR were evaluated in patients who were classified according to tumor stage (T1 and T2 low stage, T3 and T4 high stage) and Fuhrman nuclear grade (grade 1 and 2 low-grade, grade 3 and 4 high-grade). NLR and PLR were compared using Levene’s test between the groups. Results: Sixty four patients were female (52.1%) and 59 were male (47.9%). All haematological parameters were expressed as 103 /μL. Mean age, blood neutrophil, lymphocyte and platelet counts, and NLR and PLR values of the patients were 62.49±12.43 years, 6.27±2.8, 2.05±0.83, 263.72±89.03, 4.01±3.93, and 149.73±82.1, respectively. In this study, Cyclotrichium niveum is used as a potential green corrosion inhibitor to investigate the corrosion behavior of mild steel (MS) in chloride solution using electrochemical, morphological, structural and quantum chemical methods. Fifty-three flavonoids are detected in methanol extract of cyclotrichium niveum by LC/ESI-MS/MS. While icorr values decreased, Rp and inhibition efficiencies values derived from electrochemical impedance spectroscopy (EIS) and linear polarization resistance (LPR) increased in increasing inhibitor concentrations. Maximum inhibition efficiency is found to be 97.3%, that is the almost highest value, when compared to previous studies and plant extract is classified as a mixed type inhibitor with respect to the potentiodynamic polarization (PDP). The model that best fits the experimental values is the Langmuir isotherm. The temperature effect is also studied in the range of 25–55 ○C. Higher activation energy is obtained in the presence of inhibitor, suggesting that it acts as an efficient inhibitor by forming a physical barrier to the charge and mass transfer reaction, leading to reduction in corrosion rate. Scanning electron and atomic force microscopies showed the more uniform, crack and pits free structure, confirming that there is a protective film over the MS surface for inhibited solution. XPS analysis showed the existence of C, O, and Fe atoms on the MS surface. Quantum chemical calculation manifested the adsorption mechanism associated with the electronic structure of the molecules. The findings of this work can be applicable in chemical cleaning process with acid treatment.Presentation Corrosion Inhibition Performance of Pyrazinecarboxylic acid on Mild Steel in 0.5 M HCl(4rd Internatıonal Turkıc World Conference on Chemıcal Scıences and Technologıes (ITWCCST2018),, 2018) Keleşoğlu, Ayşen; Yıldız, Reşit; Dehri, İlyas; Yıldız, Reşit…
