Article Citation - WoS: 0Citation - Scopus: 1Activity of nanosized copper-boron alloys against Phytophthora species(Springer Science and Business Media Deutschland GmbH, 2023) Derviş, Sibel; Türkkan, Muharrem; İlhan, Hasan; Şimşek, Tuncay; Güler, Ömer; Derviş, Sibel; Department of Organic Agriculture / Organik Tarım BölümüThis study aimed to evaluate the antifungal activity of copper-boron (Cu-B) nanoalloys against a range of Phytophthora species, including P. capsici, P. citrophthora, P. palmivora, P. cinnamomi, P. nicotianae, P. cactorum, P. plurivora, P. inundata, and P. megasperma. The nanoalloys were synthesized via mechanical alloying under an argon atmosphere, resulting in the formation of nanocrystalline Cu-B nanoalloys with irregular morphology and particle sizes ranging from 50 to 240 nm. At a concentration of 250 µg mL−1, the Cu-B nanoalloys demonstrated complete inhibition of mycelial growth, sporangium production, and zoospore germination in all tested Phytophthora species. The EC50 values for mycelial growth ranged from 28.02 to 120.17 µg mL−1, while for sporangium production and zoospore germination, they were below 10 µg mL−1. Furthermore, the nanoalloys exhibited fungicidal activity against specific Phytophthora species, such as P. capsici, P. citrophthora, P. inundata, and P. megasperma, at concentrations of 100, 250, 250, and 250 µg mL−1, respectively. Notably, the Cu-B nanoalloys displayed significant protective and curative effects on tuber rot severity in P. nicotianae-inoculated potatoes, resulting in reductions of 94.13% and 92.61% compared to the control, respectively, at a concentration of 10 µg mL−1 (P < 0.05). These findings highlight the potential of Cu-B nanoalloys as a promising fungicide for the management of plant diseases caused by Phytophthora spp.Article Citation - WoS: 8Citation - Scopus: 10Allelic variations of glutenin subunits and their association with quality traits in bread wheat genotypes(2017) Aktaş, Hüsnü; Baloch, Faheem Shehzad; Department of Seed Production / Tohumculuk Teknolojisi BölümüThe present study was conducted to evaluate the genotype × environment interaction of the yield and quality traits for five bread wheat varieties commonly grown in the Southeastern Anatolia Region of Turkey and 20 advanced lines developed within the framework of the International Winter Wheat Improvement Project. We also determined the allelic pattern of the Glu-1 and Glu-3 loci of these genotypes and examined whether these loci had an effect on the quality traits. There was a significant variation among the genotypes and environments in terms of grain yield, protein content, sedimentation volume (SV), and the extensograph dough energy value (EDEV). The results of the study indicated that genotypic effect was more influential on SV and EDEV than environmental effect; thus, both traits could be used in breeding programs to develop elite cultivars with better quality. Twelve different high-molecularweight (HMW) glutenin alleles were identified at the Glu-A1, Glu-B1, and Glu-D1 loci, resulting in 14 allelic combinations, and 17 different alleles were observed in 19 combinations for low-molecular-weight (LMW) subunits. Furthermore, among all the genotypes, 13 + 16 and 13 + 19 alleles at Glu-B1 and 5 + 12 at Glu-D1 were observed to have the lowest frequency. Our study indicated that the combinations of HMW glutenin alleles with 2* at Glu-A1, 17 + 18 and 13 + 16 at Glu-B1, and 5 + 10 at Glu-D1, as well as the combinations of LMW alleles with subunits c and d at Glu-A3; subunits d, b, c, and g at Glu-B3; and subunits a and b at Glu-D3 had positive effects on the quality traits.Article Citation - WoS: 11Citation - Scopus: 11Anoxybacillus sp. SO B1-immobilized Amberlite XAD-16 was developed. The functional groups of Anoxybacillus sp. SO B1 immobilized on Amberlite XAD-16 were characterized in KBr tablets by Fourier transform infrared (FT-IR) spectrometry. Various parameters such as pH, amount of the adsorbent, eluent type and volume, and flow rate of the sample solution were studied. The optimum pH values of quantitative sorption for Cu(II) and Pb(II) were found to be pH 7.0 and 5.0 and Cu(II) and Pb(II) ions could be quantitatively eluted with 5.0ml of 1.0mol L-1 HCI and 10.0ml of 0.25mol L-1 HNO3, respectively. Recoveries of Cu(II) and Pb(II) were found to be 100.9 +/- 1.57% and 100.3 +/- 0.49% (N = 5), the limits of detection of Cu(II) and Pb(II) in the determination by FAAS (3s, N = 10) were found to be 0.8 and 1.6 mu g L-1, respectively. The proposed enrichment method was applied for metal ion determination from water samples such as two parts of Tigris River water in Diyarbakir and Elazig, Lake of Hazar in Elazig, and tap water in Diyarbakir. Tests used are as follows: reducing power, free radical scavenging, superoxide anion radical scavenging, total antioxidant activity, metal chelating activitiy, etc.; total phenolic content was determined. The percentage inhibition of P. ostreatus, P. eryngii, P. florida, and P. sajor-caju at 20 mg/mL concentration on peroxidation in a -carotenelinoleic acid system was 57.19, 60.68, 62.12, and 58.81%, respectively. The reducing power of P. eryngii was higher than the other samples, and its value was 0.86 at 10 mg/mL concentration. P. ostreatus and P. sajor-caju proved to be better at scavenging superoxide anion radicals than the P. eryngii and P. florida. Several polymorphisms in leptin gene (LEP), which encodes for leptin, have been described. However, its association with obesity is still controversial. Therefore, in the present study, we aimed to investigate whether LEP c.-2548 G > A polymorphism was associated with serum leptin levels, lipid parameters, and body mass index in Turkish obese patients. Forty-seven obese patients and 48 healthy individuals were included in the study. Blood samples were collected for DNA extraction. LEP c.-2548 G > A polymorphism were detected using polymerase chain reaction-restriction fragment length polymorphism technique. Serum leptin levels and lipid parameters were measured by ELISA and enzyme colorimetric assay techniques, respectively. GA or AA genotypes and A allele carrier frequencies of the c.-2548 G > A polymorphism in the LEP were higher in obese (38.3, 34.0 and 72.3 %) when compared with controls (14.6, 12.5, and 27.1 %; p = 0.011, 0.016, and 0.002, respectively). On the other hand, AA or AG genotypes were also related to increased serum leptin levels (p < 0.001) and body mass index (p < 0.0001). The main fatty acids were linoleic (LA), palmitic (PALM), oleic (OLA) and linolenic (ALA) acids in the fruits, resins and in both in vivo and in vitro grown root, leaf and stem sections of male or female tree. The major fatty acids were represented by polyunsaturated fatty acids (PUFA) accounting for 56.94 %, 64.44 % and 55.57 % in root, leaf and stem part of male tree grown in vivo, respectively. The prominent class of fatty acid composition of different male organs of P. lentiscus L. regenerated in vitro was represented by PUFA accounting for 63.24 %. It is based on the batch magnetic solid phase extraction of Bi ions by fullerene C-60 modified maghemite (gamma-Fe2O3 superparamagnetic iron oxide nanoparticles - SPION) nanoparticles. The various factors that influence the preconcentration of Bi such as pH, the amount of magnetic nanoparticles, initial Bi concentration, sample volume and the effects of possible interfering ions were investigated. A linear analytical curve was achieved in the concentration range of 0.25-10 ng mL(-1) with a limit of detection of 0.06 ng mL(-1). The precision expressed as a RSD was 7.5% for a concentration of 0.25 ng mL(-1). The method was validated by the analysis of a standard reference material (NIST SRM 1643e). In addition, 96.6-98.7% recoveries were achieved for the spiked samples. Bi concentrations in ten milk samples were successfully determined after the developed method was applied. The change of THD(I)was examined in relation to the ambient temperature (T) and solar irradiance (G). The developed model is being used to extract parameters for a given THD(I)as a function of temperature and solar radiation. This study outlines the working principle of photovoltaic (PV) panel as well as PV array. Off-grid PV system is modeled by using Matlab/Simulink program, and detailed analytical study has been carried out in this work. The design, modeling and simulation of this study are performed from 50 up to 988 W/m(2)for solar irradiance. Harmonic components have negative effects on the steady-voltage stability of the PV system. Therefore, analytical expression is needed for steady-state stability analysis in order to reduce negative effects. Hence, two analytical expressions of THD(I)were obtained by two new different methods which are statistical package for the social sciences program and genetic expression programming. Eventually, two different methods have been verified by the Matlab/Simulink program in order to find out THD(I)and demonstrated the effectiveness of the proposed strategy. As a result of this study, it is observed that input current THDI of nonlinear load is too high at low irradiance. Thermophilic bacteria Geobacillus vulcani 2Cx, Bacillus licheniformis 3CA, Paenibacillus macerans 3CA1, Anoxybacillus ayderensis FMB1, and Bacillus paralicheniformis FMB2-1 were selected, and their MIC and MBC values were assessed by treatment with AgNPs in a range of 62.5-1500 mu g mL(-1). The growth inhibition curves showed that the G. vulcani 2Cx, and B. paralicheniformis FMB2-1 strains were more sensitive to AgNPs, demonstrating a reduction in population by 71.1% and 31.7% at 62.5 mu g mL(-1) and by 82.9% and 72.8% at 250 mu g mL(-1), respectively. TEM and FT-IR analysis revealed that AgNPs caused structural damage, cytoplasmic leakage, and disruption of cellular integrity. Furthermore, cell viability showed a significant decrease alongside an increase in superoxide radical (SOR; O-2(-)) production. beta-galactosidase biosynthesis decreased to 28.8% level at 500 mu g mL(-1) AgNPs for G. vulcani 2Cx, 32.2% at 250 mu g mL(-1) for A. ayderensis FMB1, and 38.8% only at 62.5 mu g mL(-1), but it was completely inhibited at 500 mu g mL(-1) for B. licheniformis 3CA. Moreover, B. paralicheniformis FMB2-1 showed a significant decrease to 11.2% at 125 mu g mL(-1). For this aim, an experimental analysis was performed on solar assisted VGSHP system, which was designed to meet the heating needs of an experimental room, during the heating season (10.01.2013/03.31.2014). The experimentally obtained results were used to calculate energy, exergy and exergoeconomic analyses of the system and its components. The energy efficiency, exergy efficiency and exergoeconomic factors of the entire system were 67.36 %, 27.40 % and 60.51 %, respectively. In this study, the system was proposed for disseminating the use of alternative technologies supported by renewable energy systems and it has been tested for the first time in Mardin to meet its heating needs with convectional systems. The experimental results showed that the proposed solar assisted VGSHP system can be used for residential heating in Mardin and similar regions. Through Application of Selenium(Pakistan Botanical Society, 2020) Ahmad, Z.; Kızılgeçi, Ferhat; Waraich, E.A.; Barutçular, C.; Alharby, H.; Bamagoos, A.; Kizilgeci, F.; Sabagh, A.E.; Department of Seed Production / Tohumculuk Teknolojisi BölümüConsidering the burning issue the present study was undertaken in pot culture at the Cholistan Institute of Desert Studies (CIDS), The Islamia University of Bahawalpur, Pakistan for enhancing drought tolerance in two oilseed crops (OC) crops camelina (Camelina sativa L.) and canola (Canola napus L.) through improving physiological, biochemical, and antioxidants activity by foliar application of selenium (Se) under drought stress. Two Camelina (i.e.] ‘Australian Camelina’ and ‘Canadian Camelina’) and canola genotypes (i.e.] ‘AARI Canola’ and ‘UAF Canola’) were used as plant materials during the growing season of 2016. Both Camelina and Canola genotypes were grown under normal (100% FC) and water deficit (drought stress) (40% FC) conditions. Four levels of Se: such as seeds priming with 75μM Se, foliar application of Se at 7.06 μM and foliar Se in combination with seeds priming (7.06 μM & 75μM) along with control were applied at the vegetative stage of both OC crops for screening drought tolerant genotypes. All treatments were arranged three times in a randomized complete block design. Both OC crops were grown upto the maturity and data on physiochemical, antioxidants and yield components were recorded during this study. Results of the present study indicated that the physio-biochemical parameters such as WP (water potential), OP (osmotic potential), TP (turgor pressure), proline, TSS (total soluble sugar), TFAA (total free amino acids), TPr (total proteins) and TS (total sugars); [and total chlorophyll contents were improved by foliar application Se along with seed priming by Se in both OC crops in both drought stress and non-stress (control) conditions. Similarly, osmoprotectants such as GB (Glycinebetaine), anthocyanin, TPC (total phenolic contents) and flavonoids; [as well as antioxidants such as APX (ascorbate peroxidase), SOD (superoxide dismutase), POD (peroxidase) and CAT (catalase) were also showed better enhancement in both OC crops through foliar application in combination with seed priming with Se (7.06 μM & 75μM) under normal as well as water deficit (drought) conditions. Yield and its components i.e.] branches plant-1 (no.), 1000-seed weight (g), seed and biological yield (t ha-1) of both OC crops were increased through foliar application in combination with seed priming by Se (7.06 μM & 75μM) under drought and non-drought stress conditions. Both camelina and Canola genotypes categorized based on all the above-mentioned parameters under the water deficit (drought stress) condition and foliar application of Se, the genotype ‘Canadian Camelina’ maintained the highest values for all these attributes. Therefore, it is revealed that foliar application in combination with seed priming by Se helps to improve drought tolerance of OC crops and also leads to an increase in the productivity of crops under drought stress. Among the genotypes, ‘Canadian Camelina’ performed the best when seeds of the genotypes were primed with Se in combination with foliar application of Se at the vegetative stage. © 2020, Pakistan Botanical Society. From this perspective, hybride nanostructure composed from gamma-Fe2O3 magnetic nanoparticle and carboxylated multi walled carbon nanotube was synthesized and characterized by FT-IR, VSM, SEM, HR-TEM and ICP-OES. Microscopy images showed that magnetic nanoparticles were nearly spherical structure that arranged on the axis of carboxylated MWCNT. Particle size was found lower than 10 nm. VSM results showed that the obtained magnetic nanoparticles presented superparamagnetic properties at room temperature. The magnetic saturation value was determined as 35.2 emu/g. It was used for the adsorption and controlled release of harmane, a potent tremor-producing neurotoxin. Maximum adsorption capacity was calculated as 151.5 mg/g from Langmuir isotherm. Concentration of harmane was determined by HPLC with fluorescence detection. The antimicrobial activity of synthesized magnetic nanoparticle was investigated against gram-negative and gram-positive bacteria. However, no activity was observed. (C) 2015 Elsevier B.V. In order to investigate in vivo protective effects of the HT extract on rat tissues, the rats were divided into nine groups. The toxic effects of CP and the protective effects of HT extract on nucleated cells that are produced by bone marrow, serum alanine transaminase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH) and oxidative stress index (OSI) levels were investigated biochemically. Additionally, liver tissue samples were examined for histopathological changes and apoptosis by Bcl-2, Bax and caspase-3 immunohistochemistry. The results of this study show that HT seed methanol extract has high total phenolic content (179.52 mu g GAE/mg) and antioxidant activity (87.48% in 500 mu g/mL concentration). CP administration caused hepatotoxicity, myelotoxicity and hematotoxicity in the rats. Non-volatile elements such as Mo, Zr, W and Ta were tried as coating materials. It was observed that W-coated SQT gave the best sensitivity for the determination of Bi for SQT-FAAS and SQT-AT-FMS. The parameters for W-coated SQT-FAAS and W-coated SQT-AT-FAAS were optimized. Sensitivity of FAAS for Bi was improved as 4.0 fold by W-coated SQT-FAAS while 613 fold enhancement in sensitivity was achieved by W-coated SQT-AT-FAAS using 5.0 min trapping with respect to conventional FAAS. MIBK was selected as organic solvent for the re-atomization of Bi from the trapping surface. Limit of detection values for W-coated SQT-FAAS and W-coated SQT-AT-FAAS was obtained as 0.14 mu g mL(-1) and 0.51 ng mL(-1), respectively. Linear calibration plot was obtained in the range of 2.5-25.0 ng mL(-1) for W-coated SQT-AT-FAAS. Accuracy of the W-coated SQT-AT-FAAS system was checked by analyzing a standard reference material, NIST 1643e. (C) 2013 Elsevier B.V. In addition, species distribution, antimicrobial susceptibility, resistance genes, staphylococcal chromosomal cassette mec (SCCmec) type and clonality of these isolates were also investigated. MRS were identified by 16S rRNA sequencing, and typed by pulsed-field gel electrophoresis (PFGE). As a result, MRS was isolated in horses (48.3%), clinic staff (92.3%) and environmental samples (71.4%). Of the 123 MRS isolates, 118 isolates were identified as Staphylococcus lentus, and the remaining ones were found to be S. sciuri (n=3), S. intermedius (n=1) and S. fleuretti (n=1). All isolates were found to be susceptible against vancomycin, quinupristin-dalfopristin and rifampicin. Additionally, single or various combinations of resistance genes were detected among MRS isolates. SCCmec type II was identified in all isolates. Similar PFGE patterns were observed among MRS isolated from horses, humans, and environmental samples. Since MRS were concurrently isolated from horses and humans it is suggested that cross-transmission of MRS between horses and humans might occur. Aman rice). The late sown mustard is often exposed to high-temperature stress during the reproductive stage, which causes a significant decrease in seed yield. Most of the farmers plant a low yielding local mustard variety ‘Tori-7’, which is sensitive to high-temperature stress. The Oil Seeds Research Centre of Bangladesh Agricultural Research Institute has recently released short duration and high-yielding mustard varieties for the rice-based cropping system of northern Bangladesh to tackle this issue. However, the performance of these varieties in the target cropping system under late sown conditions (together with exposure to heat stress) has not been evaluated yet. Therefore, this two-year field experiment was conducted to assess the performance of newly released mustard varieties ‘BARI Sarisha-14’ and ‘BARI Sarisha-15’ in comparison with the local check variety ‘Tori-7’ sown at five planting dates (viz. October 21, November 1, November 10, November 20, and November 30). The delay in sowing shortened time (days) to flowering and maturity, regardless of the variety. Similarly, plant height, branches/plant and total dry matter accumulation of the tested varieties were also reduced upon delay in planting. The greatest seed yield was observed in varieties ‘BARI Sarisha-15’ and ‘BARI Sarisha-14’ when sown on October 21. These two varieties produced fair seed yield when sown at November 20. However, the local variety ‘Tori7’ yielded the least across all the planting dates. Therefore, the varieties ‘BARI Sarisha-14’ and ‘BARI Sarisha-15’ may be planted in the rice-based cropping system of northern Bangladesh after harvest of T. Bacillus subtilis-immobilized Amberlite XAD-16 was used as biosorbent. Effects of critical parameters such as pH, flow rate of samples, amount of Amberlite XAD-16 and biosorbent, sample volume, eluent type, and volume and concentration of eluent on column preconcentration of Cu2+ and Ni2+ were optimized. Applicability of the method was validated through the analysis of the certified reference tea sample (NCS ZC73014). Sensitivity of ICP-OES was improved by 36.4-fold for Cu2+ and 38.0-fold for Ni2+ by SPE-ICP-OES method. Limit of quantitation (LOQ) was found to be 0.7 and 1.1ng/ml for Cu2+ and Ni2+, respectively. Concentrations of Cu2+ and Ni2+ were determined by ICP-OES after application of developed method. Relative standard deviations (RSDs) were lower than 4.9% for Cu2+ and 7.9% for Ni2+. The Tigris River that irrigates a large agricultural part of Southeast Turkey is polluted by domestic and industrial wastes. Some Schiff metal complexes of the new Schiff base were prepared by the reaction of some metal salts and the Schiff base. The complexes are non-electrolytes as shown by their molar conductivities (Lambda M). The structures of metal complexes are proposed from elemental analysis, FT-IR, UV-vis, magnetic susceptibility measurements, molar conductivity measurements, mass spectra and thermal gravimetric analysis. In addition theoretical H-1 NMR, HOMO-LUMO studies of the ligand; antimicrobial and cyclic voltammetric studies of the compounds were also carried out. To characterize and identify the thermophilic isolated bacteria, morphological analysis and biochemistry tests were studied. Besides, for classification 16S rRNA gene, G-C content and DNA-DNA hybridization analysis were performed. These results indicated that strain 19 is a novel species, Anoxybacillus flavithermus sp. nov. The effects of different fermentation conditions, such as incubation time, temperature, and pH, different carbon and nitrogen sources, and surfactants on a-amylase production were investigated. Various parameters such as temperature and temperature stability, pH and pH stability, detergents and surfactants, different starches, and metal ions on influence of enzyme characterization were assayed. About 93, 87, and 81% of the activities were retained after heating the partially purified enzyme solution at 50, 60, and 70 for 240 min, respectively. Enzyme was excessively inhibited by Hg2+ (6%). The enzyme was activated by Co2+ (212%) and Mg2+ (142%). Enzyme degradated 82% of starch content in apple juice at 70 degrees C in 30 min. Experimental groups of the rats were randomly divided into 13 groups, each including seven animals: Group 1 (control) treated with saline; groups 2, 3, and 4 treated with 50, 100, or 150 mg/kg of CP, respectively; group 5 treated with 0.5 mL olive oil; groups 6 and 7 treated with 5.0 and 10 mg/kg of Car, respectively; groups 8, 9, or 10 treated with respective CP plus 5.0 mg/kg of Car; and groups 11, 12, or 13 treated with respective CP plus 10 mg/kg of Car. Serum alanine transaminase (ALT), aspartat transaminase (AST), lactate dehydrogenase (LDH), malondialdehyde (MDA), creatine kinase-MB (CK-MB), total oxidant state (TOS), oxidative stress index (OSI), and levels were high only in the CP groups. There was a dose-dependence on the CP-induced cardiotoxicity. Hemorrhage, inflammatory cell infiltration and the separation of the muscle fibers in the heart tissue supported the biochemical data. With 5.0 and 10 mg/kg Car, there was an important decrease in the CP toxicity and this was related to the oxidative and nitrosative stress in the CP-induced cardiotoxicity. Reduced inflammation and lipid peroxidation in the heart tissue and increase of serum glutathione (GSH) and total antioxidant capacity (TAS) levels were found when carvacrol was applied. Based on these findings, it could be proposed that Car was a strong candidate in preventing the CP-induced cardiotoxicity but further clinical studies should be done in order to verify its application on humans.
