Mardin Artuklu University / Mardin Artuklu ÜniversitesiThis study aimed to evaluate the antifungal activity of copper-boron (Cu-B) nanoalloys against a range of Phytophthora species, including P. capsici, P. citrophthora, P. palmivora, P. cinnamomi, P. nicotianae, P. cactorum, P. plurivora, P. inundata, and P. megasperma. The nanoalloys were synthesized via mechanical alloying under an argon atmosphere, resulting in the formation of nanocrystalline Cu-B nanoalloys with irregular morphology and particle sizes ranging from 50 to 240 nm. At a concentration of 250 µg mL−1, the Cu-B nanoalloys demonstrated complete inhibition of mycelial growth, sporangium production, and zoospore germination in all tested Phytophthora species. The EC50 values for mycelial growth ranged from 28.02 to 120.17 µg mL−1, while for sporangium production and zoospore germination, they were below 10 µg mL−1. Furthermore, the nanoalloys exhibited fungicidal activity against specific Phytophthora species, such as P. capsici, P. citrophthora, P. inundata, and P. megasperma, at concentrations of 100, 250, 250, and 250 µg mL−1, respectively. Notably, the Cu-B nanoalloys displayed significant protective and curative effects on tuber rot severity in P. nicotianae-inoculated potatoes, resulting in reductions of 94.13% and 92.61% compared to the control, respectively, at a concentration of 10 µg mL−1 (P < 0.05). Mardin Artuklu University / Mardin Artuklu ÜniversitesiThe present study was conducted to evaluate the genotype × environment interaction of the yield and quality traits for five bread wheat varieties commonly grown in the Southeastern Anatolia Region of Turkey and 20 advanced lines developed within the framework of the International Winter Wheat Improvement Project. We also determined the allelic pattern of the Glu-1 and Glu-3 loci of these genotypes and examined whether these loci had an effect on the quality traits. There was a significant variation among the genotypes and environments in terms of grain yield, protein content, sedimentation volume (SV), and the extensograph dough energy value (EDEV). The results of the study indicated that genotypic effect was more influential on SV and EDEV than environmental effect; thus, both traits could be used in breeding programs to develop elite cultivars with better quality. Twelve different high-molecularweight (HMW) glutenin alleles were identified at the Glu-A1, Glu-B1, and Glu-D1 loci, resulting in 14 allelic combinations, and 17 different alleles were observed in 19 combinations for low-molecular-weight (LMW) subunits. Furthermore, among all the genotypes, 13 + 16 and 13 + 19 alleles at Glu-B1 and 5 + 12 at Glu-D1 were observed to have the lowest frequency. Our study indicated that the combinations of HMW glutenin alleles with 2* at Glu-A1, 17 + 18 and 13 + 16 at Glu-B1, and 5 + 10 at Glu-D1, as well as the combinations of LMW alleles with subunits c and d at Glu-A3; subunits d, b, c, and g at Glu-B3; and subunits a and b at Glu-D3 had positive effects on the quality traits.Article Citation - WoS: 11Citation - Scopus: 11Anoxybacillus sp. Mardin Artuklu University / Mardin Artuklu ÜniversitesiA new method for the determination of Cu(II) and Pb(II) by flame atomic absorption spectrometry (FAAS) after preconcentrating on a column containing Anoxybacillus sp. SO B1-immobilized Amberlite XAD-16 was developed. The functional groups of Anoxybacillus sp. SO B1 immobilized on Amberlite XAD-16 were characterized in KBr tablets by Fourier transform infrared (FT-IR) spectrometry. Various parameters such as pH, amount of the adsorbent, eluent type and volume, and flow rate of the sample solution were studied. The optimum pH values of quantitative sorption for Cu(II) and Pb(II) were found to be pH 7.0 and 5.0 and Cu(II) and Pb(II) ions could be quantitatively eluted with 5.0ml of 1.0mol L-1 HCI and 10.0ml of 0.25mol L-1 HNO3, respectively. Recoveries of Cu(II) and Pb(II) were found to be 100.9 +/- 1.57% and 100.3 +/- 0.49% (N = 5), the limits of detection of Cu(II) and Pb(II) in the determination by FAAS (3s, N = 10) were found to be 0.8 and 1.6 mu g L-1, respectively. The proposed enrichment method was applied for metal ion determination from water samples such as two parts of Tigris River water in Diyarbakir and Elazig, Lake of Hazar in Elazig, and tap water in Diyarbakir. Tests used are as follows: reducing power, free radical scavenging, superoxide anion radical scavenging, total antioxidant activity, metal chelating activitiy, etc.; total phenolic content was determined. The percentage inhibition of P. ostreatus, P. eryngii, P. florida, and P. sajor-caju at 20 mg/mL concentration on peroxidation in a -carotenelinoleic acid system was 57.19, 60.68, 62.12, and 58.81%, respectively. The reducing power of P. eryngii was higher than the other samples, and its value was 0.86 at 10 mg/mL concentration. P. ostreatus and P. sajor-caju proved to be better at scavenging superoxide anion radicals than the P. eryngii and P. florida. Mardin Artuklu University / Mardin Artuklu ÜniversitesiLeptin is a protein hormone which plays a critical role in the regulation of both body-weight through reducing food intake and stimulating energy expenditure. Several polymorphisms in leptin gene (LEP), which encodes for leptin, have been described. However, its association with obesity is still controversial. Therefore, in the present study, we aimed to investigate whether LEP c.-2548 G > A polymorphism was associated with serum leptin levels, lipid parameters, and body mass index in Turkish obese patients. Forty-seven obese patients and 48 healthy individuals were included in the study. Blood samples were collected for DNA extraction. LEP c.-2548 G > A polymorphism were detected using polymerase chain reaction-restriction fragment length polymorphism technique. Serum leptin levels and lipid parameters were measured by ELISA and enzyme colorimetric assay techniques, respectively. GA or AA genotypes and A allele carrier frequencies of the c.-2548 G > A polymorphism in the LEP were higher in obese (38.3, 34.0 and 72.3 %) when compared with controls (14.6, 12.5, and 27.1 %; p = 0.011, 0.016, and 0.002, respectively). On the other hand, AA or AG genotypes were also related to increased serum leptin levels (p < 0.001) and body mass index (p < 0.0001). The main fatty acids were linoleic (LA), palmitic (PALM), oleic (OLA) and linolenic (ALA) acids in the fruits, resins and in both in vivo and in vitro grown root, leaf and stem sections of male or female tree. The major fatty acids were represented by polyunsaturated fatty acids (PUFA) accounting for 56.94 %, 64.44 % and 55.57 % in root, leaf and stem part of male tree grown in vivo, respectively. The prominent class of fatty acid composition of different male organs of P. lentiscus L. regenerated in vitro was represented by PUFA accounting for 63.24 %. Mardin Artuklu University / Mardin Artuklu ÜniversitesiIn this research, a procedure for the preconcentration of Bi at trace levels in milk samples prior to inductively coupled plasma optical emission spectrometry (ICP-OES) is proposed. It is based on the batch magnetic solid phase extraction of Bi ions by fullerene C-60 modified maghemite (gamma-Fe2O3 superparamagnetic iron oxide nanoparticles - SPION) nanoparticles. The various factors that influence the preconcentration of Bi such as pH, the amount of magnetic nanoparticles, initial Bi concentration, sample volume and the effects of possible interfering ions were investigated. A linear analytical curve was achieved in the concentration range of 0.25-10 ng mL(-1) with a limit of detection of 0.06 ng mL(-1). The precision expressed as a RSD was 7.5% for a concentration of 0.25 ng mL(-1). The method was validated by the analysis of a standard reference material (NIST SRM 1643e). In addition, 96.6-98.7% recoveries were achieved for the spiked samples. Bi concentrations in ten milk samples were successfully determined after the developed method was applied. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThe purpose of this study is to evaluate the experimental performance of a solar assisted vertical ground source heat pump system (VGSHP) for the winter climatic conditions of Mardin, which is in the South-Eastern Anatolia region of Turkey. For this aim, an experimental analysis was performed on solar assisted VGSHP system, which was designed to meet the heating needs of an experimental room, during the heating season (10.01.2013/03.31.2014). The experimentally obtained results were used to calculate energy, exergy and exergoeconomic analyses of the system and its components. The energy efficiency, exergy efficiency and exergoeconomic factors of the entire system were 67.36 %, 27.40 % and 60.51 %, respectively. In this study, the system was proposed for disseminating the use of alternative technologies supported by renewable energy systems and it has been tested for the first time in Mardin to meet its heating needs with convectional systems. The experimental results showed that the proposed solar assisted VGSHP system can be used for residential heating in Mardin and similar regions. From this perspective, hybride nanostructure composed from gamma-Fe2O3 magnetic nanoparticle and carboxylated multi walled carbon nanotube was synthesized and characterized by FT-IR, VSM, SEM, HR-TEM and ICP-OES. Microscopy images showed that magnetic nanoparticles were nearly spherical structure that arranged on the axis of carboxylated MWCNT. Particle size was found lower than 10 nm. VSM results showed that the obtained magnetic nanoparticles presented superparamagnetic properties at room temperature. The magnetic saturation value was determined as 35.2 emu/g. It was used for the adsorption and controlled release of harmane, a potent tremor-producing neurotoxin. Maximum adsorption capacity was calculated as 151.5 mg/g from Langmuir isotherm. Concentration of harmane was determined by HPLC with fluorescence detection. The antimicrobial activity of synthesized magnetic nanoparticle was investigated against gram-negative and gram-positive bacteria. However, no activity was observed. (C) 2015 Elsevier B.V. Non-volatile elements such as Mo, Zr, W and Ta were tried as coating materials. It was observed that W-coated SQT gave the best sensitivity for the determination of Bi for SQT-FAAS and SQT-AT-FMS. The parameters for W-coated SQT-FAAS and W-coated SQT-AT-FAAS were optimized. Sensitivity of FAAS for Bi was improved as 4.0 fold by W-coated SQT-FAAS while 613 fold enhancement in sensitivity was achieved by W-coated SQT-AT-FAAS using 5.0 min trapping with respect to conventional FAAS. MIBK was selected as organic solvent for the re-atomization of Bi from the trapping surface. Limit of detection values for W-coated SQT-FAAS and W-coated SQT-AT-FAAS was obtained as 0.14 mu g mL(-1) and 0.51 ng mL(-1), respectively. Linear calibration plot was obtained in the range of 2.5-25.0 ng mL(-1) for W-coated SQT-AT-FAAS. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThe present study was carried out to assess the frequency of methicillin-resistant staphylococci (MRS) among racehorses (n=209) and veterinary personnel (n=13) as well as environmental surfaces (n=14) at an equine hospital in Adana, Turkey. In addition, species distribution, antimicrobial susceptibility, resistance genes, staphylococcal chromosomal cassette mec (SCCmec) type and clonality of these isolates were also investigated. MRS were identified by 16S rRNA sequencing, and typed by pulsed-field gel electrophoresis (PFGE). As a result, MRS was isolated in horses (48.3%), clinic staff (92.3%) and environmental samples (71.4%). Of the 123 MRS isolates, 118 isolates were identified as Staphylococcus lentus, and the remaining ones were found to be S. sciuri (n=3), S. intermedius (n=1) and S. fleuretti (n=1). All isolates were found to be susceptible against vancomycin, quinupristin-dalfopristin and rifampicin. Additionally, single or various combinations of resistance genes were detected among MRS isolates. SCCmec type II was identified in all isolates. Similar PFGE patterns were observed among MRS isolated from horses, humans, and environmental samples. Since MRS were concurrently isolated from horses and humans it is suggested that cross-transmission of MRS between horses and humans might occur. Bacillus subtilis-immobilized Amberlite XAD-16 was used as biosorbent. Effects of critical parameters such as pH, flow rate of samples, amount of Amberlite XAD-16 and biosorbent, sample volume, eluent type, and volume and concentration of eluent on column preconcentration of Cu2+ and Ni2+ were optimized. Applicability of the method was validated through the analysis of the certified reference tea sample (NCS ZC73014). Sensitivity of ICP-OES was improved by 36.4-fold for Cu2+ and 38.0-fold for Ni2+ by SPE-ICP-OES method. Limit of quantitation (LOQ) was found to be 0.7 and 1.1ng/ml for Cu2+ and Ni2+, respectively. Concentrations of Cu2+ and Ni2+ were determined by ICP-OES after application of developed method. Relative standard deviations (RSDs) were lower than 4.9% for Cu2+ and 7.9% for Ni2+. The Tigris River that irrigates a large agricultural part of Southeast Turkey is polluted by domestic and industrial wastes. Some Schiff metal complexes of the new Schiff base were prepared by the reaction of some metal salts and the Schiff base. The complexes are non-electrolytes as shown by their molar conductivities (Lambda M). The structures of metal complexes are proposed from elemental analysis, FT-IR, UV-vis, magnetic susceptibility measurements, molar conductivity measurements, mass spectra and thermal gravimetric analysis. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThis study was concerned with isolation and identification of thermophilic bacteria from hot spring in Afyonkarahisar (Gecek) and optimization of a-amylase production, partial purification of alpha-amylase, and characterization of extracellular enzyme from isolated thermophilic strain 19. To characterize and identify the thermophilic isolated bacteria, morphological analysis and biochemistry tests were studied. Besides, for classification 16S rRNA gene, G-C content and DNA-DNA hybridization analysis were performed. These results indicated that strain 19 is a novel species, Anoxybacillus flavithermus sp. nov. The effects of different fermentation conditions, such as incubation time, temperature, and pH, different carbon and nitrogen sources, and surfactants on a-amylase production were investigated. Various parameters such as temperature and temperature stability, pH and pH stability, detergents and surfactants, different starches, and metal ions on influence of enzyme characterization were assayed. About 93, 87, and 81% of the activities were retained after heating the partially purified enzyme solution at 50, 60, and 70 for 240 min, respectively. Enzyme was excessively inhibited by Hg2+ (6%). The enzyme was activated by Co2+ (212%) and Mg2+ (142%). Enzyme degradated 82% of starch content in apple juice at 70 degrees C in 30 min. Experimental groups of the rats were randomly divided into 13 groups, each including seven animals: Group 1 (control) treated with saline; groups 2, 3, and 4 treated with 50, 100, or 150 mg/kg of CP, respectively; group 5 treated with 0.5 mL olive oil; groups 6 and 7 treated with 5.0 and 10 mg/kg of Car, respectively; groups 8, 9, or 10 treated with respective CP plus 5.0 mg/kg of Car; and groups 11, 12, or 13 treated with respective CP plus 10 mg/kg of Car. Serum alanine transaminase (ALT), aspartat transaminase (AST), lactate dehydrogenase (LDH), malondialdehyde (MDA), creatine kinase-MB (CK-MB), total oxidant state (TOS), oxidative stress index (OSI), and levels were high only in the CP groups. There was a dose-dependence on the CP-induced cardiotoxicity. Hemorrhage, inflammatory cell infiltration and the separation of the muscle fibers in the heart tissue supported the biochemical data. With 5.0 and 10 mg/kg Car, there was an important decrease in the CP toxicity and this was related to the oxidative and nitrosative stress in the CP-induced cardiotoxicity. Reduced inflammation and lipid peroxidation in the heart tissue and increase of serum glutathione (GSH) and total antioxidant capacity (TAS) levels were found when carvacrol was applied. The minimum inhibition concentration of (MIC) value of U(VI) was found as 85 mg/L and 15 mg/L in liquid and solid medium, respectively. Furthermore, the effect of various U(VI) concentrations on the growth of bacteria and bioaccumulation on B. vallismortis was examined in the liquid culture media. The growth was not significantly affected in the presence of 1.0, 2.5 and 5.0 mg/L U(VI) up to 72 h. The highest bioaccumulation value at 1 mg/L U(VI) concentration was detected at the 72nd hour (10 mg/g metal/dry bacteria), while the maximum bioaccumulation value at 5 mg/L U(VI) concentration was determined at the 48th hour (50 mg metal/dry bacteria). In addition to these, various concentration of U(VI) on alpha-amylase production was studied. The alpha-amylase activities at 0, 1, 2.5 and 5 mg/L U(VI) were found as 3313.2, 3845.2, 3687.1 and 3060.8 U/mg, respectively at 48th. Besides, uranium (VI) ions were preconcentrated with immobilized B. vallismortis onto multiwalled carbon nanotube (MWCNT) and were determined by UV-vis spectrophotometry. The surface macro structure and functionalities of B. vallismortis immobilized onto multiwalled carbon nanotube with and without U(VI) were examined by FT-IR and SEM. The optimum pH and flow rate for the biosorption of U(VI) were 4.0-5.0 and 1.0 mL/min, respectively. The quantitative elution occurred with 5.0 mL of 1 mol/L HCI. The loading capacity of immobilized B. vallismortis was determined as 23.6 mg/g. The certified reference sample was employed for the validation of developed solid phase extraction method. UV–vis and fluorescence spectra, aggregation, and fluorescence parameters of the zinc phthalocyanine compound were investigated. As biological properties, DPPH radical capture, antidiabetic, DNA cutting, antimicrobial, photodynamic antimicrobial, anti-biofilm activities, and microbial cell viability parameters of the compounds were determined. The highest antioxidant activity was found as 62.80% at 100 mg/L concentration with compound 4, and compound 4 also showed the best antidiabetic activity as 65.17% at 400 mg/L concentration. In DNA cutting activity, it was determined that 3 and 4 cut DNA at all concentrations. For compounds 3–4, the minimum inhibitory concentration (MIC) values were found to be 32 and 16 mg/L for Enterococcus hirae and Enterococcus feacalis, respectively. The strongest MIC value of photodynamic antimicrobial activity was achieved as 4 mg/L for E. feacalis with 4. It was observed that both compounds inhibited the microbial viability activity of Escherichia coli by 100% at concentrations of 100 mg/L. In this study, 12 spring wheat genotypes were artificially inoculated and preserved in two locations, Diyarbakr and Adyaman, during the 2011-2012 season to investigate loss in yield and yield components. Genotypes were evaluated at the adult plant stage using two partial resistance parameters: final disease severity and area under the disease progress curve (AUDPC). AUDPC ranged from 14.8 to 860 in Diyarbakr, and 74 to 760 in Adyaman. Yield loss ranged from 0.6 to 68.5% in Diyarbakr and 9.8 to 56.8% in Adyaman. Genotypes G1, G5, G7 and G8 were found to lose less yield, while higher yield loss was observed in G3, G4 (Nurkent), G5 and G9 (Karacada-98). The highest loss in thousand kernel weight was observed in a susceptible cultivar Karacada-98 in Diyarbakr followed by 43.4 and 24.4% in Adyaman. Test weight loss reached 8.89% in Diyarbakr and 20.8% in Adyaman. Yield loss and AUDPC had a positive significant relationship. Based on the values of AUDPC, final disease severity and yield loss, three major clusters were formed for 12 wheat genotypes. Chemical control of Pst is not a sustainable or economic strategy in these countries. Therefore, a suitable alternative to these pesticides, such as the development of resistant wheat cultivars, is needed. During the present investigation, a large set of wheat germplasms was characterized to determine the presence of the broad-spectrum resistance Yr15 gene using Y15K1_F2/uhw30_1R primers. This collection contained 360 accessions including 233 durum and bread wheat collected from KASIB cultivars and advanced lines that were developed by eight breeding programs within the Kazakhstan-Siberia Wheat Improvement Network and Turkey, and 127 wild emmer collected from Turkey only. The positive control was wild emmer IDs WEW 83-1, WEW 84-1, and WEW-85, which previously showed positive amplification of the Yr15 gene with these primers. Yr15, a broad-spectrum R-gene derived from wild emmer wheat encoding a putative kinase-pseudokinase protein designated as wheat tandem kinase 1, comprises a unique R-gene structure in wheat against the yellow (stripe) rust. None of the tested cultivated wheat germplasms from 174 durum wheat of the KASIB Network or 59 of bread wheat from Turkey showed positive amplification, but 89 of 127 (70.07%) wild Triticum dicoccoides showed positive amplification. The collection from the Karacadağ regions showed the maximum number of genotypes (46) with positive amplification for Yr15, followed by Eruh (10), Şırnak (5), Malazgir-Tunceli (4), Tunceli (4), Şırnak-Uludere yol (3), Tunceli-Pülümür (3), and Karacadağ/Kerteş Köyü –Siverek (3). The crystallographic analysis showed the square pyramid structures of the complexes. Electronic spectra analysis was performed to determine the nature of the interaction between the complexes and calf thymus DNA (CT-DNA). DNA cleavage activities of the complexes were examined by gel electrophoresis (pBR322 DNA). The cytotoxicity of the complexes was determined against human cervical carcinoma (HeLa) and human colorectal adenocarcinoma (HT-29) cell lines by MTT assay. The results indicated that complex Fe1 is bound to CT-DNA via the intercalation mode, while complexes Mn1 and Mn2 are bound to CT-DNA via groove binding and/or electrostatic interactions rather than the intercalation mode. In addition, they showed good binding activity, which followed the order of Fe1>Mn2>Mn1. Complexes were found to promote the cleavage of DNA from supercoiled form (SC, Form I) to nicked circular form (NC, Form II) without concurrent formation of Form III, revealing the single-strand DNA cleavage. No significant cleavage was found in the presence of Mn1 and Mn2; however, it was observed at 2000 and 3000 mu M concentrations of Fe1. The ability of Fe1 to cleave DNA was greater than that of other complexes and these results are in conformity with their DNA-binding affinities. Cytotoxicity determination tests revealed that the complex Fe1 on HeLa and HT-29 cells exhibited a higher anti-proliferative effect than Mn1 and Mn2 (Fe1>Mn2>Mn1). These studies suggested that the complex Fe1 could be a good candidate as a chemotherapeutic drug targeting DNA. [GRAPHICS]
