Murat; Çelik, Mustafa; Uçar, Edip; Güneşaçar, RamazanLeptin is a protein hormone which plays a critical role in the regulation of both body-weight through reducing food intake and stimulating energy expenditure. Several polymorphisms in leptin gene (LEP), which encodes for leptin, have been described. However, its association with obesity is still controversial. Therefore, in the present study, we aimed to investigate whether LEP c.-2548 G > A polymorphism was associated with serum leptin levels, lipid parameters, and body mass index in Turkish obese patients. Forty-seven obese patients and 48 healthy individuals were included in the study. Blood samples were collected for DNA extraction. LEP c.-2548 G > A polymorphism were detected using polymerase chain reaction-restriction fragment length polymorphism technique. Serum leptin levels and lipid parameters were measured by ELISA and enzyme colorimetric assay techniques, respectively. GA or AA genotypes and A allele carrier frequencies of the c.-2548 G > A polymorphism in the LEP were higher in obese (38.3, 34.0 and 72.3 %) when compared with controls (14.6, 12.5, and 27.1 %; p = 0.011, 0.016, and 0.002, respectively). On the other hand, AA or AG genotypes were also related to increased serum leptin levels (p < 0.001) and body mass index (p < 0.0001). All these consequences showed that LEP -2548 AA or AG genotypes are important predictors for increased levels of leptin and BMI in Turkish obese patients and it may be a useful marker for obesity risk in our population.Article Bazı fakültatif buğday genotiplerinin farklı çevrelere tepkileri(Ege Üniversitesi Ziraat Fakültesi Dergisi, 2021) Erdemci, İrfan; Aktaş, Hüsnü; Karaman, MehmetAmaç: Bu çalışmanın amacı fakültatif gelişme tabiatına sahip ekmeklik buğdaygenotiplerin farklı çevrelerde verim ve kalite özelliklerin belirlenmesidir.Materyal ve Yöntem: Bu araştırma, 2014-2015 yetiştirme sezonunda yağışa dayalışartlarda 25 ekmeklik buğday genotipi (20 ileri hat ve 5 standart çeşit) ile Diyarbakırve Muş lokasyonlarında Tesadüf Blokları Deneme Desenine göre 3 tekrarlamalıolarak yürütülmüştür. Heywood subsp. orientale Grierson and T. nitens (Boiss. & Noë) Grierson together with T. polycephalum (L.) Sch.Bip subsp. argyrophyllum (K. Koch) Podlech, and T. parthenium (L.) Sch.Bip. The fatty acids were determined by using gas chromatography, while phenolics, lipid soluble vitamins and sterols were determined by using HPLC and radical scavenging activities, total phenolics, and FRAP were determined spectrophotometrically. It was found that Tanacetum taxa have palmitic acid (C16:0), and stearic acid (C18:0) as major saturated fatty acids and linoleic acid (C18:2 n6), α-linolenic acid (C18:3 n3) and oleic acid (C18:1 n9) as principal unsaturated fatty acids. It was found that Tanacetum taxa had more total unsaturated fatty acid contents (60.24±0.3%- 70.54±0.29%) than saturated fatty acids and it was found that T. parthenium had the highest total essential fatty acid composition (58.65%±0.59%). It was also reported that the omega6/omega3 ratio of T. cadmium subsp. orientale (8.22) differed from other taxa in this study. Also, the present study showed that Tanacetum had the lowest amount of lipid soluble vitamins. On the other hand, catechin was found to be the main polyphenolic compound in this study and it was determined that T. parthenium had the highest catechin (4479.1±5.71 µg/mg) and total phenolic contents (324.91±2.01 µgGAE/mg) in this study. Rutin was only determined in two endemic taxa T. cadmeum subsp. orientale (23±0.91 µg/mg), and T. nitens (5.7±0.27 µg/mg). Also, the naringenin, vanillic acid and caffeic acid amounts of the endemic T. cadmeum subsp. orientale and T. nitens were higher than other taxa in the study. In addition, it was determined that Tanacetum taxa had a high stigmasterol content. However, T. parthenium had a higher ergosterol content (271±2.36 µg/mg). It was also found that T. parthenium has highest D2, α-tocopherol, retinol acetate, ergosterol, and stigmasterol contents among the studied taxa. In addition, the study showed that Tanacetum taxa have strong DPPH and ABTS radical scavenging activities. It was concluded that Tanacetum taxa have potent antioxidant capacity.Article Citation - WoS: 8Citation - Scopus: 3Cardioprotective Effects of Hypericum Triquetrifolium Turra. Against Cyclophosphamide Related Cardiotoxicity in Rats(Marmara Univ, 2018) Cetik Yildiz, Songul; Keskin, Cumali; Sahinturk, Varol; Ayhanci, AdnanCyclophosphamide (CYP) is commonly used as anticancer agent but its usage is limited by cardiotoxic side effects such as dose-dependent cardiac damage, morphologically defined necrosis and bleeding. Hypericum triquetrifolium Turra. (HT) shows anti-oxidative and anticarciogenic properties with its rich phenolic contents. The current study was designed to investigate the possible protective effect of HT on CYP-induced cardiotoxicity. Albino rats were randomly divided into 9 groups, each included 7 animals. Serum creatine kinase-MB (CK-MB), malondialdehyde (MDA), aspartate transaminase (AST), glutathione (GSH), total antioxidant (TAC) and total oxidant capacity (TOC) levels were investigated. Furthermore, the cardiac tissue samples were investigated histopatologically. While the levels of serum CK-MB, MDA, AST and TOC were high, the levels of serum GSH and TAC levels were low in the CYP groups. It was also observed that CYP-induced cardiotoxicity was dose dependent. In the treatment with CYP plus HT doses there was observed an essential decrease in the CYP cardiotoxicity; decreased cell damage and oxidative stress parameters and also increased GSH and TAC levels. Based on our findings, it can be proposed that HT seed methanol extract was a strong candidate in preventing the CYP-induced cardiotoxicity.Article Citation - WoS: 2Citation - Scopus: 4Characterization of Durum Wheat (triticum durum L.) Knowing the total, phospholipid (PL), and triacylglycerol (TAG) fatty acid content of L. sericata fly, which is important for health, in its different stages is important both in terms of taxonomy and physiology. After L. sericata samples used in the study were obtained commercially, they were bred under laboratory conditions and then the fatty acids in different stages were analyzed by gas chromatography. Sixteen fatty acids are determined as a result of the analysis. When individual fatty acids are considered, Palmitic acid (16:0), Palmitoleic Acid (16:1n-7), oleic acid (18:1n-9) and Linoleic Acid (18:2n-6) were found to be major ones, while the others are detected in trace amounts. It is determined that, out of total, PL and TAG, saturated fatty acids (SFA) are found at a high percentage in the eggs, monounsaturated fatty acids (MUFA) are the highest in the larvae, and polyunsaturated fatty acids (PUFA) are the highest in fly and pupa. Furthermore, out of total, PL and TAG, ∑PUFA is at a low percentage in the egg and the larval stages. Different results are found at different stages in this study. This may be because fatty acid percentages that change during metamorphosis meet different physiological needs at different phases. © 2021, ABADER (Adıyaman Bilimsel Arastırmalar Dernegi). In this context, the plants pose a danger to human health when they are eaten by the public, consumed as herbal tea or used as medicinal plants especially the toxic metals in their composition. For this reason, determining the macro, micro and toxic element content in the plants is important for health. In this study; the metal contents of root and aerial parts of nine different Salvia species were determined and the chemometric evaluation of the obtained results was made. In the principal component analysis (PCA) made with 20 elements of Salvia species, it was determined that the first two principal components explained 62.60% of the variance and the first six principal components explained 92.70%. When PCA and hierarchical cluster analysis (HCA) results are evaluated; the parts of Salvia species root and aerial parts were determined that were not clearly separated and there was no regional grouping. In addition, Salvia species prepared as herbal tea was left to different simulated body fluid medium conditions, and the changes in metal contents were examined. It was determined that were taken up Cr, Cd, Pb, Cu, Zn metals in the simulated saliva fluid (SSF), Ni, Mn metals in the simulated gastric fluid (SGF), and Na, K metals in the simulated intestinal fluid (SIF). It was determined that Fe metal in almost all Salvia species was not taken from three simulated body fluid mediums. Thus, elements determined which were taken up in body fluid mediums.Article Citation - Scopus: 4Determination of Multi-Elemental Analysis and Antioxidant Activities of Helichrysum arenarium (L.) Moench Species(Hacettepe University, 2023) Umaz, Adil; Umaz, Kader; Aydın, Fırat; Aydın, IşılIn this study, multi-elemental analysis and antioxidant activity of samples belonging to two different populations of Helichrysum arenarium (L.) Fifteen bread wheat varieties were used for field trails during 2012-13 and 2013-14 under irrigated conditions of Diyarbakir and Mardin locations, Turkey. We investigated the quality of varieties that have same HMW-GS (High molecular weight subunits) and different LMW-GS (Low molecular weight subunits) or vice versa. Results indicated that GluA3-c > d > e, GluB3-g > b' = i, and GluD3-b = c > a for LMW-GS, and GluB1-7+8 > GluB1-17+18; GluD1-5+10 > GluD1-2+12 for HMW-GS have higher extensograph dough energy, extensibility, resistance, also higher farinograph stability time, gluten index and grain hardness values. The overall evaluation of the results obtained from this study demonstrated that GluA1-1 or 2*, GluB1-7+8 and GluD1-5+10 for HMW glutenin alleles and GluA3-d, GluB3-g and b, GluD3-c for LMW glutenin alleles are associated with a strong gluten structure and accordingly selection of lines harboring these alleles can contribute to the development of genotypes with high quality in wheat breeding programs.Article Effect of Lambda-cyhalothrin on the Gill Phospholipid (PL) Subclass of Oreochromis niloticus(ABADER (Adıyaman Bilimsel Arastırmalar Dernegi), 2023) Yolcu, Murat; Satar, Elif İpek; Başhan, Mehmet; Kızmaz, VeysiFatty acids have a crucial role in providing energy and performing essential functions in living organisms. Moreover, these substances exhibit the most significant alterations in their structure based on ecotoxicological parameters when viewed from a biochemical perspective. These bioactive chemicals are present in the cellular architecture. The study of these fatty acids, crucial for maintaining the integrity and permeability of cell membranes, holds great significance for all living organisms. Consequently, doing fatty acid analysis specifically at the phospholipid level holds significant importance. The impact of lambda cyhalothrin on the fatty acid content of several phospholipid subclasses (phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS) in the gill tissue of O. niloticus (Perciformes: Cichlidae) was assessed using gas chromatography. The alterations in the fatty acid composition was analyzed 21 days after exposure. Following the complete extraction of lipids from gill tissue, the tissue was subsequently separated into different subclasses of phospholipids using thin layer chromatography. The samples were subjected to methylation and then evaluated using Gas Chromatography to determine the percentage of the fatty acid. After doing the analysis, a grand total of 16 fatty acids were identified. The research revealed that the primary fatty acids were 16:0 and 18:0 of saturated fatty acids, monounsaturated 18:1n-9, and polyunsaturated 18:2n-6, 20:4n-6, and 22:6n-6. Upon analyzing the distribution of fatty acids, it was observed that PC, PE, and PI included 16:0, PE contained 18:1, PE and PS contained C18:2n-6 and 20:4n-6, and significant alterations in C22:6n-3 were detected in PE. Our investigation revealed that the n-3/n-6 ratio of fish in the PE subclass was the lowest when compared to PC, PI, and PS.Article Citation - WoS: 2Citation - Scopus: 3Effects of chemical, organic and microbial fertilization on agronomical growth parameters, seed yield and chemical composition of chickpea(POLISH SOCIETY MAGNESIUM RESEARCH, 2023) Doğan, Serap; Çığ, FatihBecause of its valuable nutritional content, chickpea is expected to become the most important crop for the increasingly larger global population. Therefore, this research was carried out in 2018 and 2019 to investigate the effects of microbial (Bacillus-GC group, Pseudomonas tetraodonis and Brevibacillus choshinensis), organic (vermicompost and chicken manure) and chemical (DAP/2 ve DAP) fertilizer applications on yield and nutritient content of two different chickpea cultivars (Arda and Azkan). The experiment was laid out according to a randomized complete split-block design with three replications. The results expressed as the average values of two-year experiments projected that the application of chicken manure significantly improved the morphological traits of chickpea plants compared to the other treatments, while the highest phosphorus content was recorded after the application of farm manure. Additionally, the highest grain yield from both cultivars was obtained owing to the application of chicken manure. Apart from this effect, other microbial applications also played a positive role in plant growth and production, but chicken manure excelled in this respect. Thus, it has been concluded that chicken manure could be used as a suitable alternative to chemical fertilizer for chickpea culti vation in order to create a sustainable agricultural system, increase productivity and protect and improve soil properties.Article Effects of Humic Acid and Phosphorus Applications on Nutrient Composition of Lentil (Lens Culinaris Medic.)(Scibulcom Ltd., 2016) Togay, Y.; Dogan, Y.; Togay, N.; Doğan, Yusuf; 06.02. The highest root and shoot length was obtained after the application of 2.5 mg ZnO kg-1 NP. In the light of the results, ZnO nanoparticles are thought to be useful for seed development and agricultural applications.Article Evaluation Of Bread And Einkorn Wheat Under in Vıtro Drought stress(The Journal of Animal & Plant Sciences, 2017) Aslan, Didem; Aktaş , Hüsnü; Ordu, Bülent; Zencirci, NüsretThe purpose of this study was to investigate the resistance of bread and einkorn wheat genotypes under in vitro conditionsagainst drought stress during germination. Twelve bread and ten einkorn wheat genotypes were used as plant material and sevendrought stress levels were appliedbased ona three-replicate factorial restricted randomized block design in order to investigate their effects on germination rate (GR), germinating power (GP), coleoptile length (CL), shoot length (SL), root length (RL), shoot root length ratio (SRLR), root fresh weight (RFW), root dry weight (RDW), and root fresh dry weight ratio (RFDWR) during the year 2014-2015. PEG–6000 was used toevaluate the effect of drought stress under in vitro conditions on the wheat genotypes. The values of all traits were decreasedbytheincreased effect ofPEG levels (p≤ 0.05).The results of the variance analysisshowedthat thegenotypeshadsignificant statistical differences for theexaminedtraits under drought stress(p < 0.05).According to theresults of theGGE biplot analysis, of the total variation between the genotypes and traits investigated under drought stress (75.97%), PC1and PC2 represented51.51% and 24.47%, respectively. In addition, einkorn wheat populationswerelocated inthesector of GR, GP, and SRL,which means that these populations had agreater performance for these traits under drought stress conditions.Bread wheat and einkorn genotypes behaved differently for the traits under drought stress. It is considered that the results of the field and in vivo experiments for cold and drought stresswill contribute toproducingreliable suggestions.Article Citation - WoS: 2Citation - Scopus: 2Factors influencing adaptation of innovations in small ruminant production in the TRC3 Region in Turkey(Emirates Journal of Food and Agriculture, 2021) Acıbuca, Veysi; Bostan Budak, DilanThis study investigated the socio-economic factors affecting the adoption of innovation in small ruminant production in TRC3 region in Turkey. Stratified random sampling technique was used to select 366 small ruminant farmers from the region. Semi-structured questionnaire was used to collect main data for the study. Data collected were analyzed using descritive statistics, t-tests and multiple linear regression analysis. Results of the study showed that the farmers had a mean age of 45,8 years and 97,0% were male. Traditional production was dominant among the small ruminant producers and as the number of animal increases the income increases (p< 0,01). Out of twelve independent variables, eight of them were statistically significant on multiple linear regression analysis. The F-statistics was statistically significant at 1% level on communication behavior and animal health practices; 5% level on age, breeding reasons, milking way, produced products and fold type. The coefficient of multiple determination (R2) value was estimated to be 0,470 this implies that 47,0% of total variation in the output of small ruminant production was accounted for by the independent variables that were fitted into the model. The main problems of small ruminant production were high feed prices, high deaths of lamps/goat, insufficient use of pastures because of security problems and lack of shepherd due to young people unwillingness to live in rural areas
