Presentation Acanthus dioscoridis L. var. dioscoridis’ in total yağ asidi, fosfolipit ve triaçilgliserol kompozisyonlarının belirlenmesi(2014) Keskin, Cumali; Kaçar, Semra; Kaçar, Semra; 21.02. Mardin Artuklu University / Mardin Artuklu ÜniversitesiÖzet: Yağ asitleri fosfolipitler, trigliseritler ve monogliseritler gibi lipitlerin temel bileşenleridir. Doymuş, tekli doymamış veya çoklu doymamış olabilirler. Acanthus dioscoridis’in total lipit, fosfolipit ve triaçilgliserol yağ asidi kompozisyonları gaz kromatografisi (GC) ile belirlendi. A. dioscoridis’in total lipit ve TLC yöntemiyle ayrılan fosfolipit ve triaçilgliserol fraksiyonlarında 9 farklı yağ asidi tespit edildi. A. dioscoridis’in toprak üstü kısımlarından ekstrakte edilen total lipit, fosfolipit ve triaçilgliserol fraksiyonlarından elde edilen metil esterlerinde; doymuş yağ asitleri içinde C14:0, C15:0, C16:0, C17:0 ve C18:0 tekli doymamış yağ asitleri içinde C16:1ω-7 ve C18:1 ω-9, çoklu doymamış yağ asitleri içinde C18:2ω-2 ve C18:3 ω-3 temel bileşenler olarak belirlenmiştir. Total lipit, fosfolipit ve triaçilgliserolde sırasıyla doymuş yağ asitlerinden majör bileşen olarak palmitik asit (C16:0), stearik asit (C18:0) ve miristik asit (C14:0) tespit edildi. Total doymamış yağ asitleri (∑SFA) (%42.66) en fazla fosfolipit fraksiyonunda saptandı. Oleik asit (C18:1n-9) (%26.33) en fazla triaçilgliserol fraksiyonunda tespit edildi. Palmitoleik asit (C16:1n-7) (%13.67) ve total MUFA (%40.00) oranı; triaçilgliserol fraksiyonunda diğerlerine oranla belirgin miktarda yüksek bulundu. Temel yağ asitlerinden linoleik asit (C18:2n-6) (%16.26) ve linolenik asit (C18:3n-3) (%35.25) yüksek oranda total lipitte saptandı. Total doymamış yağ asitleri (∑PUFA) en fazla total lipitte (%51.51) diğer taraftan en düşük triaçilgliserol fraksiyonunda (%35.45) saptandı. Mardin Artuklu University / Mardin Artuklu ÜniversitesiVerbascum thapsus (Mullein) is a medicinal plant used in folk medicine to treat various ailments. For this study, the biological functions of Verbascum thapsus (VT) methanol extract were determined in vitro. The plant's methanol extract was created through the maceration process. The phytochemical composition of plant extracts was investigated using liquid chromatography-electrospray ionization tandem mass spectrometry. The antioxidant capacity of the extract was determined using the 2,2-diphenyl-1-picrylhydrazil (DPPH radical) and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS radical). Cell lines Caco-2 (human colorectal adenocarcinoma cells), LNCaP (Lymph Node Carcinoma of Prostate), and HEK293 (Human embryonic kidney 293 cells) were used to model colon, prostate, and non-cancerous cells. The cytotoxic activity of the plant extract on the proliferation of these cells was determined by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) assay protocol. VT extract showed moderate DPPH and ABTS radical scavenging activities at 30 mg/ml concentration. With this, VT extract was determined to inhibit acetylcholinesterase (AChE) enzyme and had strong cytotoxic activity on cancerous cell lines. In addition, our findings clearly showed that the plant extract had greater cytotoxic activity on the viability of cancerous cells compared to non-cancerous (Human embryonic kidney cells; HEK293) cells. The current findings showed that V. thapsus might be a promising anti-cancer medication candidate for the treatment of human colorectal adenocarcinoma and colon cancer, as well as a good source antioxidans. Pleurotus eryngii, in particular, is widely recognized for its organoleptic quality and favorable healthy effects, being commercially produced in great extent. The present study is just for evaluating the antioxidant capacity (scavenging DPPH radicals, reducing power, Total antioxidant activity by β-carotene-linoleic acid metod, Chelating effect on ferrous ions, superoxide anion radical scavenging activity ), antimicrobial effects (by using the disc diffusion method) and insufficiently studied cytotoxic activity (using PC-3 cell lines) of Pleurotus eryngii by using various solvent systems. For this reason; obtained extracts of the mushroom applying hexane, ethylacetate and methanol respectively were tested with in its biologic activities. As a result, The highest total antioxidant activity by β -carotene-linoleic acid metod was identified in methanol extract (63.62 mg /ml). The highest DPPH scavenging activity was obtained from ethyl acetate extract with 88.81 mg/ml. EDTA is used as standard in methal chelating activity. The highest metal chelating activity of Pleurotus eryngii identified in hexane (88,1 mg /ml) and ethylacetate (88.84 mg /ml) extracts. Additionally, in most of studied tests antioxidant activity of the mushroom increased with increasing concentration. On the other hand, the highest antimicrobial activity was obtained against Pseudomonas aeruginosa ATCC 27853 (11 mm) in ethyl acetate extract. However, the best antimicrobial activities were observed in hexane and methanol extracts for other tested microorganism. It was not determined any antimicrobial activity against Escherichia coli ATCC 25922. It has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Ethyl acetate extract showed significant inhibitory value at the concentrations of 530 µg/ml (86.20 %) against PC-3 cell lines. Using methanol extract, the maximun inhibition was observed at concentration of 680 µg/ml with value of 51.86 %. Mardin Artuklu University / Mardin Artuklu ÜniversitesiRecently, green inhibitors are replacing classical inhibitors in the acid cleaning industry due to very low cost, environmentally friendly and none toxic. In this study, Cyclotrichium niveum is used as a potential green corrosion inhibitor to investigate the corrosion behavior of mild steel (MS) in chloride solution using electrochemical, morphological, structural and quantum chemical methods. Fifty-three flavonoids are detected in methanol extract of cyclotrichium niveum by LC/ESI-MS/MS. While icorr values decreased, Rp and inhibition efficiencies values derived from electrochemical impedance spectroscopy (EIS) and linear polarization resistance (LPR) increased in increasing inhibitor concentrations. Maximum inhibition efficiency is found to be 97.3%, that is the almost highest value, when compared to previous studies and plant extract is classified as a mixed type inhibitor with respect to the potentiodynamic polarization (PDP). The model that best fits the experimental values is the Langmuir isotherm. The temperature effect is also studied in the range of 25–55 ○C. Higher activation energy is obtained in the presence of inhibitor, suggesting that it acts as an efficient inhibitor by forming a physical barrier to the charge and mass transfer reaction, leading to reduction in corrosion rate. Scanning electron and atomic force microscopies showed the more uniform, crack and pits free structure, confirming that there is a protective film over the MS surface for inhibited solution. XPS analysis showed the existence of C, O, and Fe atoms on the MS surface. Quantum chemical calculation manifested the adsorption mechanism associated with the electronic structure of the molecules. Mardin Artuklu University / Mardin Artuklu ÜniversitesiSynthesizing nanoparticles (NPs) using wild edible fungi with environmentally friendly synthesis methods is more preferred because of the advantages it provides. The fact that its synthesis is easy, economical, non-toxic and has a wide range of uses increases the interest in this subject. The aim of this study was to determine the antioxidant and cytotoxic activity of biomolecular synthesized Pleurotus eryngii silver nanoparticles (PE-AgNPs) against human prostate carcinoma (PC-3), human cervix (HeLa) and breast carcinoma (MCF-7) cells. PE-AgNPs showed significant cytotoxic activity against HeLa, PC-3, MCF-7 cell lines, and also dimethyl sulfoxide solvents of PE-AgNPs applied for their metal chelating activity, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity and antioxidant activity using the β-carotene linoleate model system. Biosynthetic PE-AgNPs were found to inhibit the proliferation of PC-3, HeLa and MCF-7 cells with IC50 values of 2.185, 46.594 and 6.169 µg / ml, respectively, during a 24-hour incubation period. With the parallel of increasing concentration (1, 2, 5, 10 mg/mL) the activities were also increased at all the tests studied. At 10 mg/ml antioxidant activities were 82%, 85% and 77% for chelation of ferrous ions reducing power, DPPH scavenging and β-carotene linoleate tests respectively. The results show that PE-AgNPs may contribute to the development of a suitable anticancer drug that can lead to a new development of nanoparticles for cancer treatment. The major FAs of TL, TG, PL in both species were Cl6:0 (palmitic açid, PA), Cl8:1 n-9 (oleic acid, OLA) and Cl8:2 n-6 (linoleic acid, LA). In both species, total PUFA amounts were found to be higher than total MUFA and total SFA in TL, TG and PL fractions. The efficient production of the fatty acids especially linolenic and oleic acids which are majorly needed in building blocks of dietary human has confirmed these species as good source of nutrition. On the other hand, insufficiently studied cytotoxic activity (using pC-3 cell lines) of these mushrooms were investigated by using various solvent systems Ethyl acetate extract of Russula delica and, Pleurltus ostreatus showed significant inhibitory value at the concentrations of 520-530 µg/ml (99,45%- 92,82%) against PC-3 cell lines with IC50; 274,53-297,77 µg /mL respectively. Methanol extracts did not show any cytotoxic activity. lt has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Mardin Artuklu University / Mardin Artuklu ÜniversitesiWith the increase of antibiotic resistance, which is present at a worrying rate, research on the use of newly developed nanoparticles as an antimicrobial agent with green biotechnology has intensified. The study aimed to investigate the antimicrobial effects of chitosan nanoparticles (CSNP) synthesized using Pleurotus eryngii extract (PE). Characterization of P. eryngii-loaded chitosan nanoparticles (PE-CSNPs) was performed with Fourier transform infrared spectrophotometer, X-ray diffraction, Field-emission scanning electron microscopy, Brunauer-Emmett-Teller, Differential scanning calorimetry, and zeta potential techniques. The FE-SEM images showed that the surface morphology of nanoparticles is similar to CS, but has more porosity network and smaller dimensions structure. The average particle size of spherical PE-CSNPs was obtained as 330.1 nm. The specific surface area and average pore diameter of the synthesized nanoparticles were found as 3.99 m2g-1 and 2.25 nm, respectively. X-ray diffraction determines the presence of an amorphous peak at 2θ = 21.2° results from CS and PE. PE-CSNPs synthesized using P. eryngii extract showed strong antimicrobial activity against Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Candida albicans as 0.0156, 0.0625, 0.0625 and 0.0312 mg ml-1, respectively. Astragalus species contained palmitic (C 16:0) (20.13%-53.8%), linoleic (C 18:2ω- 6) (13.25%-41.06%), oleic (C 18:1ω-9) (5.78%-25.7%), linolenic (C 18:3ω-3) (6.1%-22.89%), and stearic acid (C 18:0) (5.16%-13.1%) in the roots and linolenic (8.9%-51.42%), palmitic (20.16%-44.88%), linoleic (7.34%-27.57%), oleic (2.45%-17.91%), and stearic acid (4.28%-8.31%) in the shoots as major components. In this study the fatty acid composition of Astragalus species showed uniform fatty acid patterns. Mardin Artuklu University / Mardin Artuklu ÜniversitesiIn this study, 25-25 milk samples (25 colostral and 25 mature) collected at different lactational stages were used to analyse the fatty acid composition of breast milk. A gas-chromatographic method was used to perform and analyse the transmethylation of total milk lipid extracts. The milk samples contained 20 different fatty acids. Palmitic acid (C16:0), stearic acid (C18:0), myristic acid (C14:0), oleic acid (C18:1n-9), and linoleic acid (LA, C18:2n-6) were the major components of total lipid, phospholipid (PL), and triacylglycerol (TAG) fractions. Colostrum had a lower percentage of polyunsaturated fatty acids (PUFAs), a higher percentage of saturated fatty acids (SFAs), and a lower level of eicosapentaenoic acid (EPA, C20:5 n-3) and docosahexaenoic acid (DHA, C22:6 n-3) in total lipid than mature milk. In this article, the spectroscopic (ultraviolet-visible (UV-vis), electron-dispersing X-ray (EDX), and Fourier transform infrared (FT-IR)), microscopic (field emission scanning electron microscope (FE-SEM), transmission electron microscope (TEM), and atomic force microscope (AFM)), structural (X-ray diffractometer (XRD) and zetasizer), and analytic (thermogravimetric/differential thermal analyzer (TGA-DTA)) characterization of the silver nanoparticles (AgNPs) produced from Papaver rhoeas (PR) L. leaf extract are presented. PR-AgNPs are generally spherical and have a maximum surface plasmon resonance of 464.03 nm. The dimensions of the manufactured nanomaterial are in the range of 1.47-7.31 nm. PR-AgNPs have high thermal stability and a zeta potential of 36.1 mV. The minimum inhibitory concentration (MIC) values (mg L-1) of PR-AgNPs on Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans are 1.50, 0.75, 3.00, 6.00, and 0.37, respectively. In the study, the cytotoxic and proliferative effects of PR-AgNPs using the MTT (3-(4,5-dimethylthiazol-2-yl)-diphenyltetrazolium bromide) method on various cancer cell lines (CACO-2 (human colon adenocarcinoma cell), MCF-7 (human breast cancer cell), T98-G (glioblastoma multiforme cell), and healthy HUVEC (human umbilical vein endothelial cell)) cell lines are presented. After 24 and 48 h of the application, the half-maximum inhibitory concentration (IC50) values (mu g mL(-1)) of PR-AgNPs on HUVEC, CACO-2, MCF-7, and T98-G lines are 2.365 and 2.380; 2.526 and 2.521; 3.274 and 3.318; 3.472 and 3.526, respectively. Glutathione-S-transferase enzymes play a critical role in the detoxification of xenobiotics in living systems. This study aimed to examine the interaction of methylene blue with human erythrocyte glutathione-S-transferase purified in one step. Human erythrocyte glutathione-S-transferase was purified with approximately 750-fold purification and 30% efficiency by glutathione agarose affinity chromatography. The results showed that the enzyme was inhibited by methylene blue with an IC50 value of 1.40 mmol/L. The Ki constant of methylene blue was 1.17 mmol/L. The Lineweaver–Burk graph of the methylene blue showed that the type of inhibition was compatible with mixed type inhibition. A new third spot was also detected by thin-layer chromatography. Fatty acid compositions oftotal lipid, phospholipid and triacylglycerol fractions of Pleurotuseryngii were investigated by Gas chromatography. Totally 13 differentfatty acids constituents were identified. Antioxidant propertiesincluding total antioxidant activity, free radical scavenging activity,reducing power activity, metal chelating activity, and superoxideanion radical scavenging activity were evaluated using different tests.The highest total antioxidant, DPPH scavenging, metal chelating, andsuperoxide anion radical scavenging activities were determined inethyl acetate extracts. The potential of cytotoxic activity on PC-3(prostate cancer) cell lines was found to depend on the concentrationand the type of the tested extracts fractions. It was determined thathexane, ethyl acetate, and methanol extracts of Pleurotus eryngii haveinhibition effect on the growth of tested microorganisms. Mardin Artuklu University / Mardin Artuklu ÜniversitesiOlive oil has been consumed with admiration for many years in different cuisines of the world, especially in Mediterranean countries, due to its chemical profile and unique aroma flavor components. Turkey is among the leading countries in the world in terms of annual average olive oil production and consumption. In recent epidemiological studies, low mortality rates in coronary heart diseases, some of cancer types (prostate, breast and colon cancer) and some chronic diseases have been associated with the intake of antioxidant containing vegetables and fruits. In particular, it is stated that olive oil, which is widely used in diets in some Mediterranean countries, has an important role in the relatively longer life expectancy. The beneficial effects of olive oil on health are attributed to its high oleic acid content and high vitamin and non-vitamin antioxidant components. In this study, quality studies were carried out on Edremit (Balıkesir), Memecik (Aydın) and Halhalı (Mardin) olive oils. The fatty acid compositions by Gas Chromatography-Flame Ionization Detector (GC FID), the volatile component contents by Gas Chromatography-Mass Spectrometry (GC-MS), the phenolic compounds by High-Performance Liquid Chromatography (HPLC) of olive oils was determined. Total phenolic content and DPPH (2,2-Diphenyl-1-picrylhydrazil) radical capture activity of olive oils were read with Ultra Violet (UV) spectrophotometer device. In addition to these, quality parameters (peroxide value, acidity level, specific absorption values and sensory properties) were also determined. The analysis results for the quality criteria of the samples showed that all three olive oils were in the Extra Virgin Olive Oil class according to the Turkish Food Codex-Communique on Olive Oil and Olive Pomace Oil. Among the major phenolic compounds, the highest values were found in Halhali olive oil with hydroxytyrosol 6.57 mg/kg and oleuropein 7.40 mg/kg, and the lowest values were found in Memecik olive oil with 0.75 mg/kg and 0.87 mg/kg, respectively. It was calculated by using the IC50 formula that the total phenolic contents were between 348.31-483.09 mg.GAE/kg in three olive oils. Dpph radical removal activities were found, that the lowest in Memecik olive oil (29.35 mg.TE/kg) and the highest in Halhali olive oil (269.26 mg.TE/kg), by calculating with absorbance values. As the main fatty acid, oleic acid levels ranged from 65.86% (Halhali) to 71.30% (Edremit). This research demonstrates a new, simple, cost-effective, and environmentally friendly method for synthesizing chitosan bionanocomposite (CBNC) from Morchella esculenta (L) Pers-extract (MEE). The antifungal property of the synthesized Morchella esculenta (L) Pers-chitosan bionanocomposite (MCBNC) against some plant pathogens was also evaluated. FTIR, XRD, FE-SEM, DSC, TGA, and BET were used to characterize the synthesized MCBNC. Mushroom-based chitosan nanoparticles were evaluated for antifungal activity against some fungal pathogens, including Neoscytalidium novaehollandiae, N. dimidiatum, Alternaria alternata, Verticillium dahliae, Bipolaris sorokiniana, and Colletotrichum sp. The findings obtained clearly showed that chitosan nanoparticles have antifungal activity. The results suggest that the chitosan nanoparticle can be used in the field to protect various crops from phytopathogens. Novelty impact statement Chitosan bionanocomposite (MCBNC) synthesis was performed for the first time using the wild mushroom Morchella esculenta, which has strong bioactive properties. It was observed that the bionanomaterial, which was characterized by FTIR, XRD, FE-SEM, DSC, TGA, and BET analyses, has high antifungal activity against plant pathogens such as Alternaria alternata and Bipolaris sorokiniana. Possibilities to evaluate these synthesized nanoparticles (RD-AuNPs) as bioactive substances were investigated. Characterization of synthesized RD-AuNPs were characterized via UV-vis, XRD, FTIR, EDX. In a spherical view, RD-AuNPs with a crystal size of 34.76 nm were synthesized. As a result, fungal systems used for nanomaterial biosynthesis as an effective alternative to chemical synthesis can be used in different biotechnological and medical applications. RD-AuNPs produced by green synthesis can be evaluated in this context.
