Mardin Artuklu University / Mardin Artuklu ÜniversitesiCyclotrichium origanifolium is a medicinal plant belonging to the Lamiaceae family. In this study, phenolic content analysis, antimicrobial effects, and cytotoxic effects of extracts of C. origanifolium were investigated. In the extracts, phenolic compound analysis by the liquid chromatography–electrospray ionization– tandem mass spectrometry method, antimicrobial effect by the minimum inhibition concentration method, and cytotoxic effect on human dermal fibroblasts (HDF), glioblastoma cell (U87), ovarian adenocarcinoma cell (Skov-3), and human colorectal adenocarcinoma cell (CaCo-2) cancer cell lines were investigated. Cytotoxicity analyses were performed by the MTT method. In addition, the GST and AChE enzyme activities of the extracts were also measured. Around 18 compounds were detected in both the methanol and ethanol extract. It was found that the best antimicrobial effect on Gram-negative Pseudomonas aeruginosa was on methanol extract, while the ethanol extract was on Candida albicans fungus (respectively, 2.50mg/ml, 5.0 μg/ml). A 500μg/ml of methanol extract has been shown to have cytotoxic activity high effect on HDF cells. Mardin Artuklu University / Mardin Artuklu ÜniversitesiIn this study, in vitro antioxidant, antimicrobial, anticholinesterase and phenolic profile of different solvent extracts of Scilla mesopotamica speta were determined in detail. In vitro antioxidant activities and total phenolic and flavonoid contents of plant extracts obtained with different solvents were tested in terms of 2.2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activities. The highest total phenolic and total flavonoid contents were determined in the ethyl acetate extract (62.24 mu g GAE/mg) and chloroform extract (87.72 mu g QE/mg) respectively. The highest DPPH radical scavenging activity was detected in ethyl acetate extracts. Antimicrobial and antifungal activities were investigated by MIC method. The inhibitory activities of the extracts on the acetyl cholinesterase enzyme were investigated. Liquid chromatography (LC) tandem mass spectrometry LC-MS/MS was used to determine the phenolic component content of extracts. Thirty-one different components were identified in the analyses and their amounts were measured. Practical applications Scilla mesopotamica speta is an endemic and medicinal plant. It was determined that the extracts of this plant had a very rich content in terms of phenolic compounds, especially caffeic and ferulic acids. However, this plant was remarkable for its antioxidant, anticholinesterase, and antimicrobial activities. Mardin Artuklu University / Mardin Artuklu ÜniversitesiPurpose The phenolic composition, antioxidant, antimicrobial activity, enzyme inhibition activity, and cytotoxic activity potentials of the plant Rumex acetosella L. (R. acetosella) were examined in this study. Materials and Methods: The chemical composition of R. acetosella methanol extract was identified by the LC-MS/MS method. The antioxidant activity was tested using beta-carotene/linoleic acid, DPPH free radical scavenging, ABTS cation radical scavenging, CUPRAC reducing power, and metal chelating activity methods. The cytotoxic activity was determined by the MTT assay using human ovarian adenocarcinoma (Skov-3), glioblastoma (U87), human dermal fibroblasts (HDF), and human colorectal adenocarcinoma (CaCo-2) cell lines. The antimicrobial activity of methanolic extracts was tested on gram-negative (Escherichia coli and Pseudomonas aeuriginosa) and gram-positive bacteria (Staphylococcus aureus and Bacillus subtilis) using the in vitro minimum inhibition concentration method (MIC). Enzyme inhibition activity of R. acetosella methanol extract was measured spectrophotometrically against acetylcholinesterase (AChE) and glutathione S-transferase (GST) enzymes. Results: The findings showed that the major components of the methanol extract content were luteolin-7-O-glucoside (1.599 m/L), polydatin (91,024 m/L), and shikimic acid (0.773 m/L). It was determined that the extract and standard antioxidant (a-tocopherol) results in DPPH center dot, and ABTS center dot + tests performed to determine the antioxidant activity were close to each other, and this value was more effective than the standard antioxidant (alpha-tocopherol) in the CUPRAC test. These results suggested that the plant's antioxidant potential was higher when compared with reference antioxidant compounds. It was determined that the methanol extract of R. acetosella had a weaker effect on the growth of the tested microorganisms than the antibiotics used as standard. The activity of the GST and AChE enzymes was found to be severely inhibited by the methanol extract of R. acetosella. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThe impact of oils rich in long chain monounsaturated fatty acids (LCMUFA) against hyperlipidemia and non-alcoholic fatty liver disease (NAFLD) has been inadequately described. In addition, the chemical solvents and the high temperature used in vegetable oils extraction process from seeds cause severe loss of many vital compounds. So the goal of this paper was to examine the effect of cold pressed Eruca sativa Mill seeds oil (ESSO), as a source of LCMUFA, on hyperlipidemia and NAFLD in Syrian hamster. The ESSO content of fatty acids was analyzed using chromatographic methods. Fifty two (52) healthy male golden Syrian hamsters used in this experiment were randomly divided into 4 groups (Completely Randomized Design). Negative control group, CHD group, positive control group and ESSO group. This experiment was achieved in two periods. The first period continued 4 weeks, in which hyperlipidemia and NAFLD were induced in CHD, positive control and ESSO groups through feeding on a hyperlipidemic diet. The second period also lasted 4 weeks, in which ESSO was orally gavaged at 2 g/kg of the body weight daily to animals of ESSO group. The levels of total cholesterol (TC), triglycerides, HDL-C and glucose and the activities of ALT, AST, ALP, LDH and CK were analysed in the serum. One way analysis of variance (ANOVA) followed by Duncan's multiple range test was used for statistical analysis. The consumption of hyperlipidemic diet for 4 weeks caused a significant raise (P<0.05) of triglycerides, glucose, ALT, AST, LDH, CK and a significant reduction (P<0.05) of the HDL-C/TC ratio, at the same time created lipid accumulation in liver cells in CHD, positive control and ESSO groups in comparison with negative control group at the end of the first period. These negative influences were alleviated in ESSO group by administration of ESSO at the end of second period. Mardin Artuklu University / Mardin Artuklu ÜniversitesiSilver has long been valued as a precious metal, and it is used to make ornaments, jewelry, high-value tableware, utensils, and currency coins. Human exposures to silver and silver compounds can occur oral, dermal, or by inhalation. In this study, we investigated genotoxic and oxidative effects of silver exposure among silver jewelry workers. DNA damage in peripheral mononuclear leukocytes was measured by using the comet assay. Serum total antioxidative status (TAS), total oxidative status (TOS), total thiol contents, and ceruloplasmin levels were measured by using colorimetric methods among silver jewelry workers. Moreover, oxidative stress index (OSI) was calculated. Results were compared with non-exposed healthy subjects. The mean values of mononuclear leukocyte DNA damage were significantly higher than control subjects (p < 0.001). Serum TOS, OSI, and ceruloplasmin levels were also found to be higher in silver particles exposed group than those of non-exposed group (p < 0.001, p < 0.001, p < 0.01, respectively). However, serum TAS levels and total thiol contents of silver exposed group were found significantly lower (p < 0.05, p < 0.001, respectively). This plant is found in the Mediterranean basin as a southern west part of Europe and North Afric forestes. Objective: Isolation of Isocarpine From Adenocarpus Complicatus as Anticandidal Agent. Methods: A new piperidine alkaloid Isocarpine was isolated from the aerial parts of Adenocarpus complicatus L., which is gross in Syria. The structure of the new compound was determined by UV, IR, EI-Ms, 1H-NMR, 13C-NMR, DEPT-135, DQF-COSY and HMQC. Results: The anticandidal activity of isocarpine was screened against 38 Candida strains and showed remarkable inhibitory effect compared with fluconazole as an antifungal reference drug. Mardin Artuklu University / Mardin Artuklu ÜniversitesiÖZET AMAÇ: Bu çalışmanın amacı Mardin kent merkezinde yaşayan 15 yaş üzerindeki kadınlarda sigara içiciliği sıklığı ve etkileyen faktörleri saptamaktır. YÖNTEM: Araştırma kesitseldir. Araştırma evrenini Mardin kent merkezindeki dört sağlık ocağı ve bir AÇS dispanseri bölgesinde yaşayan 15 yaş üstü 21 890 kadın oluşturmuştur. En az örnek hacmi Epi Info 2000 programında beklenen sıklık %22,1, kabul edilebilir %25 alınarak 759 bulunmuştur. Nisan –Mayıs 2005 tarihlerinde, Mardin Sağlık Yüksekokulu öğrencileri tarafından, her sağlık kurumundan rasgele seçilen 12 sokaktaki ilk 25 kadın alınarak toplam 1500 kadın ile yüz yüze görüşülmüştür. Eksik veri nedeniyle 29 anket değerlendirilmeye alınmamıştır. Sigara içme durumu hiç içmeyenler, içip bırakanlar ve halen içenler olarak sorgulanmıştır. Çapraz tablolarda halen sigara içtiğini söyleyenler sigara içenler olarak alınmış ve diğerleriyle (hiç içmeyenler ve bırakanlar) ile karşılaştırılmıştır. Veriler bilgisayar ortamında değerlendirilmiş, istatistiksel olarak yüzde, ki kare testi kullanılmıştır. BULGULAR: Kadınların %22,9’unun sigara içicisi, %3,9’ununn sigarayı bırakmış olduğu saptanmıştır. Sigara içiciliği en yüksek 25–34 yaş grubunda (%31,8) en düşük 55 yaş ve üzeri grupta (%10,0) bulunmuştur (p<0,01). Sigara içme sıklığı en yüksek “yüksek okul mezunu” grupta (%34,3) en düşük “okuryazar olmayan” grupta (%16,0) çıkmıştır (p<0,01). Sigara içiciliği eşinden ayrılmış kadınlarda %30,4 iken, evlilerde %24,4 ve bekârlarda %22,4 bulunmuştur (p<0,05). Sigara içiciliği çalışan kadınlarda (%40,2) çalışmayan yada ev hanımı olan kadınlardan (%21,7) daha yüksek çıkmıştır (p<0,001). SONUÇ: Mardin’de kadınlarda sigara içme prevalansı yüksektir ve önemli bir halk sağlığı surunudur. Genç yaştaki, eğitim düzeyi daha yüksek, eşinden ayrılmış ve çalışan kadınlar risk gruplarıdır. In this study, we designed a novel series of 1,3,4-thiadiazoles through the reaction of 2-(4-methyl-2-(2-(1-phenylethylidene)hydrazineyl)thiazole-5-carbonyl)-N-phenylhydrazine-1-carbothioamide (3) with the proper hydrazonoyl halides. Using the MTT assay, the newly synthesized thiadiazoles' growth-inhibitory potential against the liver cancer cell line HepG2-1 was assessed. In comparison to the standard drug doxorubicin (IC50 = 0.72 ± 0.52 µM), the results showed that two compounds, 16b and 21 (IC50 = 0.69 ± 0.41 and 1.82 ± 0.94 µM, respectively) had promising anticancer activity. The structural activity relationship (SAR) was investigated. In addition, molecular docking analysis onto quinone oxidoreductase2 (NQO2) receptor (PDB: 4ZVM) was investigated against the potent compounds to examine the reliability of the in vitro results. In this study, the in vivo genotoxic and antigenotoxic effects of methanol and water extract and phenolic content of Hypericum perforatum flower, fruit, and seed methanol extracts were analyzed. HPLC was used to evaluate the quantities of 3,4-Dihydroxybenzoic acid, syringic acid, hydroxycinnamic acid, O-coumaric acid, caffeic acid, and catechin in the methanol extracts. The alkaline comet test was used to assess the DNA damage and protective effects of H. perforatum flower fruit, seed methanol, and water extract on human mononuclear leukocytes. The amounts of catechin and caffeic acid in seed methanol extract were found as quite high when compared to other extracts. The highest protective effects were seen at 10 and 50μg/ml concentrations of seed methanol extract. The optimum doses of fruit, flower, and seed extracts obtained from H. perforatum neutralized the genotoxic effect. This effect is stronger in seed methanol extract than other extracts. Structures of new compounds and complexes were determined by 1D-NMR: 1H NMR, 13C NMR, DEPT-135, 2D-NMR: COSY, HMQC, HSQC, HMBC, IR, and MS spectroscopy. The synthesized compounds were evaluated against a wide range of microorganisms, including Gram-positive and Gram-negative bacteria as well as fungal strains. Acyclic nucleoside was prepared for the first time, the expected structure of the final new compound 3 was determined based on IR, NMR, and mass spectroscopy, with safe and mild reaction conditions. The synthesized acyclic nucleoside has a potent and efficient antimicrobial activity compared to reference drugs particularly as an antibacterial agent, and can be used as an alternative to the commonly used antibiotics after performing the necessary biological research for its validation.
