Rats were divided into four groups as control, doxorubicin (DOX), DOX + ALA and ALA. A single dose of 15 mg/kg DOX was administered i.p to the DOX and DOX + ALA groups. 50 mg/kg ALA was given to the DOX + ALA and ALA groups by oral gavage every other day. After 28 days, rat testes and serum samples were collected and analysed. Administration of DOX alone caused a decrease in body and relative testicular weights, seminiferous tubule diameter and germinal epithelium thickness, Johnsen's score and serum testosterone levels. DOX treatment led to severe testicular damage such as tubular degeneration, and atrophic tubules. Also, the activities of superoxide dismutase and glutathione peroxidase were reduced, while the level of malondialdehyde was increased in the testis. The mRNA levels of apoptotic-related genes (CASP3, TP53, BAX, BCL2) and apoptotic index were increased, while mitofusin-2 decreased. DOX caused an increase in CASP3 and a decrease in mitofusin-2 immunoreactivities. Treatment with ALA markedly improved all of DOX-induced biochemical, histochemical and molecular alterations in rat testis. Consequently, ALA has a therapeutic role in ameliorating DOX-induced testicular damage in rats.Article Analysis of bioactive compounds using LC-ESI-MS/MS, cytotoxic, antimicrobial effects, and enzyme activities from Cyclotrichium origanifolium(Wiley Online Library, 2022) Aktepe, Necmettin; Keskin, Cumali; Baran, Ayşe; Baran, Mehmet Fırat; Keskin, Cumali; Taşkin, Abdullah; Yavuz, Ömer; Demirtaş, İbrahim; Oğuz, Ercan; Jahan, IsratCyclotrichium origanifolium is a medicinal plant belonging to the Lamiaceae family. In this study, phenolic content analysis, antimicrobial effects, and cytotoxic effects of extracts of C. origanifolium were investigated. In the extracts, phenolic compound analysis by the liquid chromatography–electrospray ionization– tandem mass spectrometry method, antimicrobial effect by the minimum inhibition concentration method, and cytotoxic effect on human dermal fibroblasts (HDF), glioblastoma cell (U87), ovarian adenocarcinoma cell (Skov-3), and human colorectal adenocarcinoma cell (CaCo-2) cancer cell lines were investigated. Cytotoxicity analyses were performed by the MTT method. In addition, the GST and AChE enzyme activities of the extracts were also measured. Around 18 compounds were detected in both the methanol and ethanol extract. It was found that the best antimicrobial effect on Gram-negative Pseudomonas aeruginosa was on methanol extract, while the ethanol extract was on Candida albicans fungus (respectively, 2.50mg/ml, 5.0 μg/ml). A 500μg/ml of methanol extract has been shown to have cytotoxic activity high effect on HDF cells. GST and AChE activity were found to decrease in a concentration-dependent manner.Article The antioxidant and anti-apoptotic potential of Pleurotus eryngii extract and its chitosan-loaded nanoparticles against doxorubicin-induced testicular toxicity in male rats(Wiley, 2021) Erdem Güzel, Elif; Acay, Hilal; Yıldırım, Ayfer; Acay, Hilal; Yıldırım, AyferThis study was conducted to evaluate the protective role of Pleurotus eryngii extract (PE) and Pleurotus eryngii extract-loaded chitosan nanoparticles (PE-CSNP) against doxorubicin (DOX)-induced testicular toxicity in rats. Male rats were divided into six groups: control (DMSO/ethanol), PE (200 mg/kg PE), PE-CSNP (30 mg/kg PECSNP), DOX (10 mg/kg DOX, a single dose, i.p), DOX+PE (10 mg/kg DOX+200 mg/ kg PE) and DOX+PE-CSNP (10 mg/kg DOX+30 mg/kg PE-CSNP). PE and PE-CSNP were administered by oral gavage every other day for 21 days. DOX-treated rats showed histopathological impairment compared with the control group. There was an increase in the apoptotic index, caspase 3 (CASP3), BCL2-associated X apoptosis regulator (BAX), dynamin-related protein 1 (DRP1) expression and total oxidative status (TOS) in the DOX group, while mitofusin-2 (MFN2), total antioxidative status (TAS) and serum testosterone levels of the DOX group reduced when compared with the other groups. PE and PE-CSNP treatments provided significant protection against DOX-induced oxidative stress by reducing TOS levels and increasing TAS levels. CASP3, BAX, apoptotic index and DRP1-MFN2 expressions were restored by PE and PE-CSNP. However, the PE-CSNP showed higher antioxidant and anti-apoptotic efficacy compared with PE. Thus, our results provide evidence that CSNP and PE could synergistically have a potent antioxidant and anti-apoptotic therapy against DOX-induced testicular damage in male rats.Article Antioxidant, Ache Inhibitory, and Anticancer Effects of Verbascum Thapsus Extract(C M B Assoc, 2023) Keskin, Cumali; Baran, Ayse; Valioglu, Ferzane; Teng, Lei; Atalar, Mehmet Nuri; Keskin, Cumali; Beilerli, AferinVerbascum thapsus (Mullein) is a medicinal plant used in folk medicine to treat various ailments. For this study, the biological functions of Verbascum thapsus (VT) methanol extract were determined in vitro. The plant's methanol extract was created through the maceration process. The phytochemical composition of plant extracts was investigated using liquid chromatography-electrospray ionization tandem mass spectrometry. The antioxidant capacity of the extract was determined using the 2,2-diphenyl-1-picrylhydrazil (DPPH radical) and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS radical). Cell lines Caco-2 (human colorectal adenocarcinoma cells), LNCaP (Lymph Node Carcinoma of Prostate), and HEK293 (Human embryonic kidney 293 cells) were used to model colon, prostate, and non-cancerous cells. The cytotoxic activity of the plant extract on the proliferation of these cells was determined by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) assay protocol. VT extract showed moderate DPPH and ABTS radical scavenging activities at 30 mg/ml concentration. With this, VT extract was determined to inhibit acetylcholinesterase (AChE) enzyme and had strong cytotoxic activity on cancerous cell lines. In addition, our findings clearly showed that the plant extract had greater cytotoxic activity on the viability of cancerous cells compared to non-cancerous (Human embryonic kidney cells; HEK293) cells. The current findings showed that V. thapsus might be a promising anti-cancer medication candidate for the treatment of human colorectal adenocarcinoma and colon cancer, as well as a good source antioxidans. We aimed to determine the degree of kidney function impairment by evaluating urine neutrophil gelatinase-associated lipocalin (NGAL) and kidney injury molecule-1 (KIM-1) levels. Materials and methods: in total, 15 obese, 26 overweight, and 26 control adolescents aged 10 to 16 years were enrolled into the study. Urine samples were evaluated for NGAL and KIM-1 levels using enzyme-linked immunosorbent assay kits. We investigated the association between obesity and related comorbidities with urinary NGAL and KIM-1 excretion. Results: no significant differences were noted between the obese, overweight, and control groups in urinary NGAL and KIM-1 excretion (p = 0.327 and p = 0.917, respectively). In the obese and overweight groups urinary NGAL levels were 50.39 [30.88-74.22] in females and 26.67 [23.24-45.59] in males (p = 0.013). Also, urinary NGAL levels were increased in obese and overweight adolescents with LDL dyslipidemia at 64.12 [30.98-114.32] as compared to those without LDL dyslipidemia: 39.51 [25.59.56.37] (p = 0.024). Furthermore, a correlation was observed between insulin and homeostasis model assessment of insulin resistance levels with the NGAL/creatinine ratio in the overweight group (r = 0.515; p = 0.008, and r = 0.483; p = 0.014, respectively). Such correlation was not found in the obese group. Conclusion: the effect of obesity on renal function could not be determined in children. A longer exposure may be required for obesity-induced disruption of renal function in children. Renal function may be disrupted by dyslipidemia in obese adolescents. Furthermore, obesity impaired renal function in female adolescents. The normalization of these urinary markers as related to urine creatinine should be discussed.Presentation Assessment Of The Antimicrobial, Antioxidant And Cytotoxic Activities Of The Wild Edible Mushroom Pleurotus Eryngii From Hakkari, Turkey(2017) Acay, Hilal; Dündar, Abdurrahman; Keskin, Cumali; Kaçar, SemraAbstract: Mushroom is an important source of natural compounds with acknowledged bioactivity. Pleurotus eryngii, in particular, is widely recognized for its organoleptic quality and favorable healthy effects, being commercially produced in great extent. The present study is just for evaluating the antioxidant capacity (scavenging DPPH radicals, reducing power, Total antioxidant activity by β-carotene-linoleic acid metod, Chelating effect on ferrous ions, superoxide anion radical scavenging activity ), antimicrobial effects (by using the disc diffusion method) and insufficiently studied cytotoxic activity (using PC-3 cell lines) of Pleurotus eryngii by using various solvent systems. For this reason; obtained extracts of the mushroom applying hexane, ethylacetate and methanol respectively were tested with in its biologic activities. As a result, The highest total antioxidant activity by β -carotene-linoleic acid metod was identified in methanol extract (63.62 mg /ml). The highest DPPH scavenging activity was obtained from ethyl acetate extract with 88.81 mg/ml. EDTA is used as standard in methal chelating activity. The highest metal chelating activity of Pleurotus eryngii identified in hexane (88,1 mg /ml) and ethylacetate (88.84 mg /ml) extracts. Additionally, in most of studied tests antioxidant activity of the mushroom increased with increasing concentration. On the other hand, the highest antimicrobial activity was obtained against Pseudomonas aeruginosa ATCC 27853 (11 mm) in ethyl acetate extract. However, the best antimicrobial activities were observed in hexane and methanol extracts for other tested microorganism. It was not determined any antimicrobial activity against Escherichia coli ATCC 25922. It has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Ethyl acetate extract showed significant inhibitory value at the concentrations of 530 µg/ml (86.20 %) against PC-3 cell lines. Using methanol extract, the maximun inhibition was observed at concentration of 680 µg/ml with value of 51.86 %. Overall, the mushroom used as a daily nutrient could be a source for new drug developments and a kind of treatment in cancer therapies, and also, organic extracts of Pleurotus eryngii may contain substances that stimulate bioactivityPresentation Atatürk Baraj Gölü’nden Dört Tatlısu Balığının Yağ Asidi İçeriklerinin Tespiti.(2012) Kaçar, Semra; Başhan, Mehmet; Oymak, AhmetBu çalışmada Eylül 2008 tarihinde Atatürk Baraj Gölü'nden toplanan Capoeta trutta, Liza abu, Cyprinion macrostomum ve Achanthobrama marmid'in kas dokusu yağ asidi kompozisyonu araştırılmıştır. In particular, some components in certain foods can directly affect human health by showing biological activity (bioactive) in the body. In this context, extra virgin olive oil is a food that has recently gained importance due to its positive effects on health. In this study, phenolic compound profiles (oleuropein, hydroxytyrosol, tyrosol, catechin, pinoresinol, luteolin, apigenin, caffeic acid, vanillic acid, t-ferulic acid, and p-coumaric acid), total phenolic content (TFC) and total antioxidant capacity (TAC) of 5 different extra virgin olive oils obtained from different olive varieties (Domat, Uslu, Arbequina, Taggiasca, Koroneiki) were investigated. The amounts of the individual phenolic compound in the oil samples were determined by a high-pressure liquid chromatography system (HPLC), while TFC and TAC were determined by using spectrophotometric methods. Hydroxytyrosol, as the major phenolic compound, was found at the highest content (129.375 mg/kg) in Koroneiki olive oil, whereas its lowest (14.485 mg/kg) content was found in Domat olive oil (P<0.05). Koroneiki olive oil also showed the highest content for tyrosol, while Uslu and Arbequina olive oils had the lowest contents for this compound (P<0.05). For pinoresinol, the maximum contents were found in Domat (16,817 mg/kg) and Koroneiki (15,160 mg/kg) samples, whereas the Uslu oils (5,260 mg/kg) exhibited the minimum content. Phenolic acids were found at minor levels in all samples. According to the spectrophotometric results, although the TFC level in Koroneiki olive oil was the highest (294.822 mg gallic acid equivalent (GAE)/kg), this value was not statistically different from the values in Domat (259.950 mg GAE/kg) and Uslu (284.267 mg GAE/kg) oils (P>0.05). Similarly, the difference between the TFC of Arbequina (208.962 mg GAE/kg) and Taggiasca (219.526 mg GAE/kg) oils were not significant (P>0.05). The results of TAC demonstrated that Koroneiki oil showed the highest (110.396 mg trolox equivalent (TE)/kg) value for antioksidan aktivity, while Taggiasca oil showed the lowest (58.465 mg TE/kg) value (P<0.05). The results of this study show that the investigated bioactive properties can be used as a distinguishing feature for olive oil from different olive varieties.Article Biochemical components, enzyme inhibitory, antioxidant and antimicrobial activities in endemic plant Scilla mesopotamica speta(Wiley, 2021) Aktepe, Necmettin; Keskin, Cumali; Baran, Ayşe; Baran, Mehmet Fırat; Akmeşe, Şükrü; Akmeşe, ŞükrüIn this study, in vitro antioxidant, antimicrobial, anticholinesterase and phenolic profile of different solvent extracts of Scilla mesopotamica speta were determined in detail. In vitro antioxidant activities and total phenolic and flavonoid contents of plant extracts obtained with different solvents were tested in terms of 2.2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activities. The highest total phenolic and total flavonoid contents were determined in the ethyl acetate extract (62.24 mu g GAE/mg) and chloroform extract (87.72 mu g QE/mg) respectively. The highest DPPH radical scavenging activity was detected in ethyl acetate extracts. Antimicrobial and antifungal activities were investigated by MIC method. The inhibitory activities of the extracts on the acetyl cholinesterase enzyme were investigated. Liquid chromatography (LC) tandem mass spectrometry LC-MS/MS was used to determine the phenolic component content of extracts. Thirty-one different components were identified in the analyses and their amounts were measured. Practical applications Scilla mesopotamica speta is an endemic and medicinal plant. It was determined that the extracts of this plant had a very rich content in terms of phenolic compounds, especially caffeic and ferulic acids. However, this plant was remarkable for its antioxidant, anticholinesterase, and antimicrobial activities. Considering the strong antioxidant, antimicrobial, and enzyme inhibition activities of the Scilla mesopotamica speta it can be suggested as a source of anticancer, antimicrobial, and antiviral drugs.Article Biosynthesis of AgNPs by extract from waste leaves of Citrullus lanatus sp.
