Article A Rare Cause of Dysphagia: Giant Lipoma in Parapharyngeal Space(Lippincott Williams and Wilkins, 2020) Aslan, Mehmet; Ozer Ozturk, Ebru; Dogukan, Fatih MertBackground: Parapharyngeal area is one of the most complex areas of head and neck anatomy. Tumors of the parapharyngeal region are very rare among head and neck tumors. Lipomas also constitute a tiny proportion of parapharyngeal tumors so parapharyngeal area lipomas are very rare in the literature. Due to anatomical location, these tumors treatment is challenging. Clinical Report: A 20-year-old male patient admitted to our department with complaints of swallowing difficulty and a mass on the right side of the neck. Magnetic resonance imaging was performed, and it was reported as a 6 4.5 cm lipoma. He underwent excision of parapharyngeal area lipoma by a transoral approach. Conclusion: Parapharyngeal lipomas are very rare, and difficult to diagnose before they turn out symptomatic. Mardin Artuklu University / Mardin Artuklu ÜniversitesiIntroduction : Cyanide poisoning, whether it be accidental or intentional, remains a significant danger to adults and children, especially in societies where agriculture is a primary source of income. We examined the clinical follow-up, complications, and results of cyanide poisoning cases that occurred after eating the pits and seeds of plants containing cyanide glycosides, such as apricot kernels and almonds. Methods : Between 01/01/2017 and 01/08/2022, 14 children aged 1-18 years who were followed up with a prediagnosis of cyanide poisoning in our Paediatric Intensive Care Unit (PICU) were retrospectively analysed. Results : Eight of the patients followed with a preliminary diagnosis of cyanide poisoning were female and six were male. The most common admission month was July (42.8%) coinciding with the agricultural season. The most common symptoms at presentation were weakness and fatigue (n = 7). In the PICU, 4 patients presented lip cyanosis; 3, altered level of consciousness. Vomiting, seizure, headache, dizziness and palpitatons were less frequent. Four patients were treated with hydroxocobalamin (Cyanokit (R) ) as an antidote due to acidosis in their blood gases. All patients treated for cyanide poisoning were discharged. Conclusions : Cyanide poisoning should be considered in paediatric patients with suspicious findings, sudden loss of consciousness, increased anion gap acidosis and lactic acidosis. The history of eating the seeds of plants such as apricot and almonds should be investigated .Article Ameliorative Effects of Agomelatine Against Doxorubicin-Induced Hepatotoxicity(BMC, 2025) Savas, Hasan Basri; Sozen, Mehmet Enes; Cuce, Gokhan; Batur, TubaDrug-induced hepatotoxicity is a significant impediment to the use of doxorubicin, a commonly employed chemotherapeutic agent with established efficacy in cancer treatment. The present study aimed to determine the potential protective effects of agomelatine against doxorubicin hepatotoxicity in rat toxicity models. Thirty-two rats were divided into four groups: control (with saline administration), Doxo (with 40 mg/kg doxorubicin administration), Doxo + Ago20, and Doxo + Ago40 (with 20 and 40 mg/kg agomelatine administration and 40 mg/kg doxorubicin administration). On the day of 14 rats were sacrificed, samples were collected for comparison of immunohistochemical, hematological, and biochemical analysis. There were statistically significant differences between the study groups in terms of immunohistochemical, hematological, and biochemical parameters. Agomelatine administration reduced the TNF-alpha, and caspase-3, which increased by doxorubicin, and reversed levels of oxidative stress markers altered by doxorubicin (p < 0.05). Doxorubicin induces oxidative stress, apoptosis, and hepatotoxicity. Despite sustained research efforts, the outbreak remains uncontrolled, with affected patients still lacking proper treatment. This review is a valuable asset for researchers and practitioners aiming to delve into the yet unexplored potential of Anatolian flora in the fight against COVID-19 and other viral infections. Numerous medicinal plants in Anatolia, such as thyme, sage, cannabis, oregano, licorice root, and Origanum sp., contain bioactive compounds with proven antiviral properties that have been used in the region for centuries. It is now the second largest cause of death in the world. The present study concerned antioxidant, anticancer and anticholinesterase activities and protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in methanol extracts of flowers, fruits and seeds of Hypericum amblysepalum. Materials and Methods: Antioxidant properties including free radical scavenging activity and reducing power, and amounts of total phenolic compounds were evaluated using different tests. Protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in extracts were determined by HPLC. Cytotoxic effects were determined using the MTT test with human cervix cancer (HeLa) and rat kidney epithelium cell (NRK-52E) lines. Acetyl and butyrylcholinesterase inhibitory activities were measured by by Ellman method. Results: Total phenolic content of H. amblysepalum seeds was found to be higher than in fruit and flower extracts. DPPH free radical scavenging activity of the obtained extracts gave satisfactory results versus butylated hydroxyanisole and butylated hydroxytoluene as controls. Reducing power activity was linearly proportional to the studied concentration range: 10-500 µg/mL LC50 values for H. amblysepalum seeds were 11.7 and 2.86 respectively for HeLa and NRK-52E cell lines. Butyryl-cholinesterase inhibitory activity was 76.9 ± 0.41 for seed extract and higher than with other extracts. Mardin Artuklu University / Mardin Artuklu ÜniversitesiIntroduction Peripheral artery disease (PAD) poses a growing clinical challenge due to an aging population, despite advances in treatment methods. Various scoring systems have emerged to predict high-risk patients, including the HALP (hemoglobin, albumin, lymphocyte, and platelet) score, known for predicting prognosis in cancers and stroke. This study assesses the HALP score's relation to lesion severity and long-term mortality in PAD patients. Methods We retrospectively analyzed 305 symptomatic PAD patients undergoing endovascular intervention. The following formula was used to calculate the HALP score: hemoglobin (g/L) x albumin (g/L) x lymphocyte count (/L) / platelet count (/L). Lesion severity was classified by TASC-II: TASC AB and TASC CD. Mortality data were obtained from hospital and social security records. Results The study involved 305 patients (mean age 64.4 +/- 11.8 years; 72.1% male), divided into survivors (208) and non-survivors (97). ROC analysis identified HALP score as the strongest predictor of long-term mortality (AUC: 0.736; 95% CI: 0.679-0.793; p < .001). HALP score (HR, 0.087; 95% CI, 0.025-1.300; p < .001), age (p < .001), DM (p = .007), and CRP (p = .013) independently predicted mortality. Kaplan-Meier analysis showed higher HALP scores linked to lower long-term mortality (Log-rank: 20.102, p < .001), with an average follow-up of 48 +/- 18 months. Conclusion The HALP score emerged as a robust predictor of PAD prognosis, surpassing individual components and other parameters. Lower HALP scores correlated with more severe lesions and reduced life expectancy.Article Attitudes of Health Sciences Students in Turkey Towards Individuals With Different Sexual Orientations(Womens Health & Action Research Centre, 2025) Deger, Vasfiye B.; Kacan, Havva; 09.01. Mardin Artuklu University / Mardin Artuklu ÜniversitesiStudy was planned in a descriptive design to examine how the empathic tendencies of students in the Faculty of Health Sciences affect their attitudes toward individuals who have different sexual orientations and was conducted with seven hundred and six students. The data collected online using the Student Information Form, Empathic Tendency Scale (ETS) and Attitudes Toward Lesbian and Gay Men Scale (ATLGS). Participants' mean scores were 62.98 +/- 18.79 on the ESS and 29.02 +/- 10.75 on the ATLGS. When the correlation between ETS and ATLGS scores was analyzed, a positive and moderate (p<0.05) correlation (r = 0.62) was found between ETS and ATLGS scores. These scores were moderate and the correlation between ETS and ATLGS scores was significant (F=438.585; p<0.05). Empathic disposition explained 38.3% of the total change in ATLGS level (R2=0.383) and increased ATLGS level (B=0.620). It was found that as students' empathic skills improved, their homophobic attitudes decreased.Article Automated Mucormycosis Diagnosis from Paranasal CT Using ResNet50 and ConvNeXt Small(MDPI, 2025) Toprak, Serdar Ferit; Dedeoglu, Serkan; Kozan, Gunay; Ayral, Muhammed; Can, Sermin; Turk, Omer; Akdag, MehmetPurpose: Mucormycosis is a life-threatening fungal infection, where rapid diagnosis is critical. We developed a deep learning approach using paranasal computed tomography (CT) images to test whether mucormycosis can be detected automatically, potentially aiding or expediting the diagnostic process that traditionally relies on biopsy. Methods: In this retrospective study, 794 CT images (from patients with mucormycosis, nasal polyps, or normal findings) were analyzed. Images were resized and augmented for training. Two transfer learning models (ResNet50 and ConvNeXt Small) were fine-tuned to classify images into the three categories. We employed a 70/30 train-test split (with five-fold cross-validation) and evaluated performance using accuracy, precision, recall, F1-score, and confusion matrices. Results: The ConvNeXt Small model achieved 100% accuracy on the test set (precision/recall/F1-score = 1.00 for all classes), while ResNet50 achieved 99.16% accuracy (precision approximate to 0.99, recall approximate to 0.99). Cross-validation yielded consistent results (ConvNeXt accuracy similar to 99% across folds), indicating no overfitting. An ablation study confirmed the benefit of transfer learning, as training ConvNeXt from scratch led to lower accuracy (similar to 85%) Conclusions: Our findings demonstrate that deep learning models can accurately and non-invasively detect mucormycosis from CT scans, potentially flagging suspected cases for prompt treatment. These models could serve as rapid screening tools to complement standard diagnostic methods (histopathology), although we emphasize that they are adjuncts and not replacements for biopsy. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThis study aimed to investigate the protective effect of quercetin against arsenic-induced oxidative damage, inflammation, and apoptosis in mouse BALB/c 3T3 fibroblast cells (NIH-3T3). Arsenic at different concentrations of 0.05 mu M (low), 0.5 mu M (medium), 10 mu M (high) doses were used to induce toxicity, while 120 mu m quercetin was used for treatment. MTT and LDH analyses were performed to determine the effect of arsenic and quercetin on cell viability, while oxidative stress markers and antioxidant enzyme activities were measured by spectrophotometric method. TNF-alpha and IL-1 beta levels were measured by the ELISA method, Autodock programs were used for molecular docking studies. In addition, computer-based analyses of quercetin and succimer molecules were performed using SwissADME web tools. TNF-alpha (PDB ID: 2AZ5), IL-1 beta (PDB ID: 1ITB), Caspase3 (PDB ID: 2XYG), Bax (PDB ID: 4S0O), SOD (PDB ID:1CBJ), GSH-Px (PDB ID: 1GP1) and Bcl-2 (PDB ID: 1G5M) crystal structures were obtained from the Protein Data Bank. Bax and Bcl-2 levels of apoptotic genes and mRNA expression levels of Caspase-3 activity were measured using the QRT-PCR technique. TUNEL staining was performed to determine DNA fragmentations, while DAPI staining was done to visualise nuclear modifications. Quercetin has been found to significantly reduce oxidative stress, inflammation, and apoptosis in cells and exert anti-apoptotic effects. Molecular docking studies revealed quercetin shows good binding affinity with molecules with SOD, GSH-Px, Bax, Bcl-2, Caspase-3, TNF-alpha and IL-1 beta structures, and has been observed to bind with Bax and Bcl-2 with molecular docking scores of -7.5 and - 7.7 kcal/mol, respectively. These findings are supported by results showing that quercetin is effective in anti-apoptotic and anti-inflammatory processes in arsenic-induced cells under in vitro conditions. Mardin Artuklu University / Mardin Artuklu ÜniversitesiThis study aimed to investigate the efficacy of manual therapy and exercise versus synchronized telerehabilitation with self-manual therapy and exercise in treating Subacromial Pain Syndrome (SAPS). Sixty individuals diagnosed with SPS, aged 18-50 years, were randomly assigned to home exercise (HE), manual therapy (MT), and telerehabilitation (TR) groups. Treatment protocols were administered over 8 weeks and included specific exercises and therapy interventions. Outcome measures included the Visual Pain Scale (VAS), shoulder range of motion (ROM) via goniometric measurements, Quick Disability Arm-Shoulder-Hand Problems Survey (Q-DASH), and patient satisfaction. Results revealed that both MT and TR groups exhibited reduced pain, increased ROM, lower Q-DASH scores, and higher patient satisfaction than the HE group. However, no significant differences were found between the MT and TR groups regarding pain levels, ROM, Q-DASH scores, or patient satisfaction. The study concludes that both telerehabilitation and manual therapy effectively alleviate pain and are well-received by patients with SPS. Additionally, manual therapy demonstrates superiority in enhancing functional levels compared to exercise-based interventions (Registration: NCT05200130).Article Comparison of the Transobturator Tape and Minisling Methods in the Treatment of Stress Urinary Incontinence(Iniestares, S.A., 2025) Sagir, Süleyman; Basgut, Ozlem; Tuncekin, Adem; Ergün, Müslüm; Turgut, OmerBackground: This study aims to evaluate and compare the transobturator tape and minisling surgical techniques in the treatment of stress urinary incontinence, focusing on their effectiveness and postoperative outcomes. Methods: This retrospective analysis included 113 patients who underwent midurethral sling surgery for stress urinary incontinence at Islahiye State Hospital between January 2016 and October 2022. The patients were divided into two groups: Those treated with transobturator tape (Group 1, n = 70) and those treated with minisling (Group 2, n = 43). Data were collected from medical records and follow-up interviews. The variables assessed included demographics (age and body mass index), preop-erative and postoperative Urinary Incontinence Quality of Life Scale scores, and postoperative pain details. Operative details, such as procedure duration and hospital stay length, were recorded. Patients were also asked about postoperative symptoms, including groin pain and dyspareunia. Additionally, the duration of symptom-free months following surgery was documented. Binary logistic regression analysis was conducted to evaluate the effect of surgical methods on long-term well-being. Results: The transobturator tape and minisling techniques demonstrated similar improvements in postoperative Urinary Incontinence Quality of Life Scale scores (p = 0.213). However, the minisling procedure had significantly shorter operative time (29.2 min vs. 52.2 min, p < 0.001) and hospital stay (1.33 days vs. 1.96 days, p = 0.003) than transobturator tape. Postoperative complications were notably higher in the transobturator tape group, with significantly more patients reporting groin pain (28.6% vs. 4.7%, p = 0.002) and dyspareunia (20.0% vs. 0.0%, p = 0.002). Logistic regression analysis identified surgical method as the primary predictor of long-term well-being beyond one year (p = 0.040). Conclusions: While the transobturator tape and minisling surgeries are effective for treating stress urinary incontinence, the minisling method offers advantages, such as shorter operative time, reduced hospital stay, and fewer postoperative complica-tions, over the transobturator tape method. These findings suggest that the minisling approach may provide a more favorable patient experience, particularly due to reduced postoperative pain and quick recovery, than the transobturator tape method. © 2025 Elsevier B.V., All rights reserved.Article Cryogel-Immobilized Catalase as a Biocatalyst with Enhanced Stability Against Microplastics(MDPI, 2025) Erol, Kadir; Alkan, Mehmet Huseyin; Alacabey, IhsanCatalase is a pivotal antioxidant enzyme that decomposes hydrogen peroxide and reduces oxidative stress. However, its low thermal and operational stability limits applications in challenging environments, particularly those contaminated with emerging pollutants such as polystyrene-based microplastics (PS-MPs). In this study, cryogels composed of Poly(2-hydroxyethyl methacrylate-co-allyl glycidyl ether) [Poly(HEMA-co-AGE)] were synthesized and evaluated as immobilization matrices to enhance catalase stability. Cryogels containing varying AGE concentrations were characterized using FT-IR, SEM, TEM, TGA, and BET analyses. The formulation with 250 mu L AGE exhibited optimal physicochemical properties, including improved water retention, increased surface area, and high immobilization capacity (356.3 mg center dot g(-1)). Immobilized catalase maintained superior activity under PS-MP-induced stress across a range of concentrations (0-1.0 mg center dot mL(-1)), temperatures (4-60 degrees C), and exposure times (up to 5 h). Kinetic modeling revealed a significant improvement in substrate affinity, with Km decreasing from 54.9 to 17.1 mM, while Vmax decreased moderately. Long-term stability tests showed that immobilized catalase retained similar to 80% activity after 70 days at 4 degrees C and 55% after 15 reuse cycles. Desorption studies confirmed the reusability of the cryogel system. These findings suggest that Poly(HEMA-co-AGE) cryogels provide a robust and reusable platform for catalase stabilization, offering potential for applications such as wastewater treatment and biosensing in microplastic-contaminated systems.Article Does the Presence of Chronic Lymphocytic Thyroiditis Affect Diagnostic Value of Fine Needle Aspiration Biopsy in Bethesda Category III Nodules?(Dr. Mladen Stojanovic University Hospital, 2023) Pedük, Şevki; Koçer, BelmaThis study aimed to determine the relationship between the presence of Hashimo-to’s thyroiditis (HT) and malignancy rates with prognostic factors in thyroid nodules diagnosed as Bethesda category III, and to examine the effect of HT on diagnostic value of fine-needle aspiration biopsy (FNAB). Demographic information, preoperative examination, and final pathological evaluation of patients with Bethesda category III (AUS-FLUS) nodules who had been operated on in our department over the last 6 years were analyzed. Statistical analyses were performed using the Student’s t-test, Mann-Whitney U test and χ2-test and logistic regression analysis using SPSS version 22 software. The malignancy rate on final pathology of 159 patients was 24.5%. Malignancy rates were found to be higher in patients with HT coexistence (30.7% vs. 21.5%, p=0.20). Poor prognostic factors such as multifocality, number of metastatic lymph nodes (p=0.04), and extrathy-roidal extension were more common in patients with cancer in the pathology specimen who were in the non-HT group. It cannot be said that HT decreases diagnostic value of FNAB in lesions diagnosed with AUS-FLUS. Method: In accordance with the purpose of the study, volunteers were asked to consume 100 ml of the extract every morning for 29 days. Three tubes of blood samples were taken from the volunteers on the 15th and 29th days of the study. Biochemical and hematological analysis of the blood samples were performed and immunomodulatory effects through cytokines were examined. The values obtained from 3 tubes of blood obtained from volunteers before the use of mushroom extract were used as control. The chemical composition and beta-glucan content of 100 ml of mushroom water extract were also analyzed. Result: IL-4, IL-6, IL-10 and IL-13 could not be detected because the values were below the lowest standard value. TNF-alpha, IFN-gamma and IL-1 beta 15th and 29th day values decreased compared to the 1st day (control) values (p < 0.05). However, there was no significant difference observed between the 15th and 29th day. No abnormalities were observed in biochemical and hematological values. Also, the beta-glucan content of extract was found 38.12 mg/100 ml. Conclusion: The frequency range of kidney and liver function test results confirmed that P. osreatus is a reliable food source. Considering the cytokine values these results indicate that P. ostreatus water extract has an anti-inflammatory effect. As no significant difference was observed in 29 days of use, it is thought that 15 days of daily consumption of the extract may be sufficient for the anti-inflammatory effect to occur. Our study aimed to evaluate the relationship between HALP score and prognosis in non-ST-elevation myocardial infarction (NSTEMI) patients. Methods Between 1 January 2020 and 1 January 2022, 568 consecutive patients diagnosed with NSTEMI from a single center were included in the study retrospectively. The patients were divided into two equal groups according to the median HALP cutoff value of 44.05. Patients were followed for at least 1 year from the date of admission. Results The average age of the patients was 62.3 +/- 10.6 years and 43.7% were female. In-hospital and 1-year mortality were found to be significantly higher in the group with low HALP scores (6.0 vs. 2.1%, P = 0.019 and 22.5 vs. 9.9%, P < 0.001, respectively). In receiver operating characteristic curve analysis, a cutoff level of 34.6 of the HALP score predicted 1-year mortality with 71% sensitivity and 65% specificity (area under the curve: 0.707, 95% confidence interval: 0.651-0.762, P < 0.001). In Kaplan-Meier analysis, higher mortality rates were observed over time in the group with lower HALP scores (log-rank test=16.767, P < 0.001). In Cox regression analysis, the HALP score was found to be an independent predictor of 1-year mortality (odds ratio: 0.969, 95% confidence interval: 0.958-0.981, P < 0.001). The effective use of these signals, especially in disease detection, is very important in terms of both time and cost. Currently, in general, EEG studies are used in addition to conventional methods as well as deep learning networks that have recently achieved great success. The most important reason for this is that in conventional methods, increasing classification accuracy is based on too many human efforts as EEG is being processed, obtaining the features is the most important step. This stage is based on both the time-consuming and the investigation of many feature methods. Therefore, there is a need for methods that do not require human effort in this area and can learn the features themselves. Based on that, two-dimensional (2D) frequency-time scalograms were obtained in this study by applying Continuous Wavelet Transform to EEG records containing five different classes. Convolutional Neural Network structure was used to learn the properties of these scalogram images and the classification performance of the structure was compared with the studies in the literature. In order to compare the performance of the proposed method, the data set of the University of Bonn was used. The data set consists of five EEG records containing healthy and epilepsy disease which are labeled as A, B, C, D, and E. METHODS: A total of 24 female rats were randomized into three groups. Group 1 served as the control. Group 2 underwent an ovarian torsion/detorsion procedure. Group 3 underwent similar procedures as Group 2; additionally, 4 mg/kg of etomidate was administered intraperitoneally 30 minutes before ovarian detorsion. Blood samples were analyzed for lipid peroxidation, pro-inflammatory cytokine levels, and antioxidant enzyme activity RESULTS: Biochemical analysis of blood samples revealed reductions in pro-inflammatory cytokines, including interleukin-1 Beta (IL1 beta ), interleukin-6 (IL-6), and tumor necrosis factor -alpha (TNF- alpha ), in Group 3 compared to Group 2 (p=0.005, p=0.016, and p<0.001, respectively). Additionally, a decrease in malondialdehyde (MDA) levels was observed in Group 3 compared to Group 2 (p<0.001). In contrast, activities of antioxidant enzymes, including superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), were significantly increased in Group 3 compared to Group 2 (p=0.031 and p=0.001, respectively). Furthermore, Group 3 demonstrated notable reductions in histopathological scores for follicular degeneration, vascular occlusion, bleeding, and inflammation compared to Group 2 (p<0.001, p<0.001, p<0.001, and p=0.001, respectively). This study aimed to evaluate prenatal comfort, sleep, and quality of life in pregnant women with cholestasis. Methods: This cross-sectional study was implemented between November 2022 and June 2023 at Mardin Training and Research Hospital with 150 pregnant women who received a diagnosis of pregnancy-induced intrahepatic cholestasis and agreed to participate. The following tools were utilized to collect data: A personal information form exploring socio-demographic and obstetric characteristics of participants, the Prenatal Comfort Scale (PCS), the Pittsburgh Sleep Quality Index (PSQI), and the World Health Organization Quality of Life-Brief Form (WHOQOL-BREF). Results: The mean age of participants was 27.79 +/- 6.33 years. The mean PCS and PSQI scores were 61.20 +/- 5.84 and 9.52 +/- 3.02, respectively. The mean scores of "physical health, psychological health, social relationships, and environmental health" sub-dimensions in WHOQOL-BREF were 10.63 +/- 2.18, 10.48 +/- 2.10, 11.31 +/- 3.28, and 11.27 +/- 2.10, respectively. A significant difference was found for PSQI regarding hospitalization status and change in sleep quality variables (p = 0.025 and p = 0.035, respectively). Conclusions: Cholestasis of pregnancy creates problems such as pruritus, body image changes, hospitalization, and poor sleep quality in women. This study showed that pregnant women with cholestasis had low levels of sleep quality and quality of life, implying that cholestasis affects their sleep quality, prenatal comfort levels, and quality of life in general. Method: This is a cross-sectional study. The population of the study consisted of Syrian pregnant women who applied to Mardin Training and Research Hospital between August 15 and September 16, 2023. A total of 146 Syrian pregnant women who met the inclusion criteria participated in the study. The sociodemographic information form and the Prenatal Care Satisfaction Scale were used as data collection tools. Results: The rate of those who received prenatal care from a midwife/nurse is 80.1% and those who received less than 4 prenatal care was 89.7%. The most common reason for not receiving adequate prenatal care was lack of information with a rate of 39.7%. The mean score of the PCSS was 73.39 +/- 14.78. Conclusion: The study findings indicate that lack of information is one of the major barriers to healthcare access for migrant pregnant women. The present study aimed to evaluate the clinical characteristics, molecular genetic aetiology, and long-term outcomes of a large nationwide cohort of children with VDDR1A from Turkey. Methods: In this multi-centre retrospective cross-sectional study, we collected clinical characteristics, laboratory features, molecular genetic analysis results, and long-term follow-up of a nationwide cohort of patients with VDDR1A using a web-based research network, CEDD-NET, for paediatric endocrinology research. Results: In total, 118 patients (57 F, 61 M) with VDDR1A were recruited. The median age of the diagnosis was 1.7 years (0.2-18.3 years). The most common presenting complaints were skeletal deformity (n = 61), short stature (n = 45), and delay in walking (n = 42). The most common mutation was a splice-donor-site mutation (c.195+2T>G) (n = 42), followed by a 7-bp duplication 1319-1325dupCCCACCC (Phe443Profs∗24) (n = 25), and two missense mutations p.K192E (c.574A>G) (n = 17) and c.1474C>T (p.R492W) (n = 12). The novel c.195+2T>C and c.1215_1215+2delTGTinsCGA splice-site and c.1144C>A missense variants were firstly described in our cohort. Conclusion: The most common four mutations accounted for the underlying aetiology of VDDR1A in approximately 81% of the cohort, indicating Turkey may serve as a mutational hotspot or exhibit a founder effect for these variants. Our large cohort s results suggested no clear and clinically meaningful phenotype-genotype relationship in VDDR1A. © 2025 Elsevier B.V., All rights reserved.
