    Molecular phylogeny of plant pathogenic fungi based on start codon targeted (SCoT) polymorphism

    Background: A number of molecular marker systems have been developed to assess genetic diversity, carry out phylogenetic analysis, and diagnose and discriminate plant pathogenic fungi. The start codon targeted (SCoT) markers system is a novel approach used here to investigate intra and interspecific polymorphisms of phytopathogenic fungi. Materials and methods: This study assessed genetic variability between and within 96 isolates of ten fungal species associated with a variety of plant species using 36 SCoT primers. Results: The six primers generated 331 distinct and reproducible banding patterns, of which 322 were polymorphic (97.28%), resulting in 53.67 polymorphic bands per primer. All primers produced informative amplification profiles that distinguished all fungal species. With a resolving power of 10.65, SCoT primer 12 showed the highest polymorphism among species, followed by primer 33 and primer 29. Polymorphic loci (PPL), Nei's diversity index (h), and Shannon index (I) percentages were 6.25, 0.018, and 0.028, respectively. UPGMA analysis separated all isolates based on morphological classification and revealed significant genetic variation among fungal isolates at the intraspecific level. PCoA analysis strongly supported fungal species discrimination and genetic variation. The other parameters of evaluation proved that SCoT markers are at least as effective as other DNA markers. Conclusions: SCoT markers were effective in identifying plant pathogenic fungi and were a powerful tool for estimating genetic variation and population structure of different fungi species.

    The Strategic Relations Between the Mongols and Christians: The Factor of Diplomatic Missions

    In the 13th century, the Mongols had a significant impact on both the Muslim and Christian worlds. The rapid expansion of the Mongol Empire alarmed both communities, especially following the conquest of Baghdad in 1258 and the subsequent fall of the Abbasid Caliphate. For Christians, this created an opportunity to view the Mongols as potential allies against Muslims. Christian Europe regarded the Mongol campaigns against the Islamic world as developments that could align with their Crusader objectives. In this context, the Papacy and European kingdoms dispatched envoys to the Mongol leaders, seeking to establish diplomatic relations and secure the Mongols as allies against Islam. While the Mongols exhibited a tolerant attitude toward various religions, they leveraged these diplomatic missions to serve their own interests. Their interactions with Christians, intended to weaken the political and military power of Muslims, were strategic rather than a commitment to lasting alliances. The Mongols and Christians engaged in a diplomatic process where both sides aimed to maximize their benefits. These diplomatic missions facilitated a deeper mutual understanding between Mongol leaders and the Christian world. One notable example is the envoys sent by Pope Innocent IV to Guyuk Khan in 1245. While Christians hoped to form an alliance with the Mongols against Muslims, the Mongols aimed to strengthen their empire by fostering relations with the West. However, instead of creating a Crusader army against Islam, the Mongols pursued their expansionist policies.These diplomatic interactions also paved the way for religious exchanges. The Mongols' tolerance allowed Christian missionaries to operate within their territories. Franciscan and Dominican friars, in particular, gained access to Mongol courts and sought to spread Christianity. However, these missionary efforts remained secondary to the broader political and military objectives of the diplomatic missions. In this context, the diplomatic relations between the Mongols and Christians in the 13th century were shaped by the pursuit of strategic advantages against Muslims. Both sides hoped to gain military and political benefits from these interactions; however, these diplomatic exchanges did not result in a lasting alliance. The Mongols prioritized their own interests, while Christians, in the aftermath of the failed Crusades, continued to seek new allies. Diplomatic missions became a significant historical element reflecting the complex relationships between the Muslim, Christian, and Mongol worlds.

    The Modified Asymmetric Chondro-Perichondrial Island Graft in Type I Tympanoplasty: A Retrospective Analysis of 784 Patients

    OBJECTIVE: This study aimed to assess the efficacy of a novel modified asymmetric chondro-perichondrial island graft in tympanoplasty. The design features a longer anterior segment compared to the posterior segment, addressing limitations of symmetrical grafts. We investigated the impact of this modified graft on hearing improvement and graft success rates in patients undergoing tympanoplasty surgery. METHODS: This retrospective study evaluated 784 patients with chronic otitis media who underwent primary type I tympanoplasty with a modified asymmetric chondro-perichondrial island graft technique. Demographics and characteristics of all patients, preoperative location of the perforation, and Pure-Tone Audiometry (PTA) were assessed preoperatively, and graft success and postoperative hearing outcomes were evaluated at the 12-month follow-up. RESULTS: This study evaluated the efficacy of a modified asymmetric chondro-perichondrial island graft in tympanoplasty. In 784 patients, the mean 12-month postoperative Air-Bone Gap (ABG) improvement was 17.3 dB with a 99% graft success rate. Preoperative ABG significantly improved from 24 dB to 6.6 dB postoperatively (p < 0.001). Perforation location did not affect ABG improvement (p = 0.193) but did influence graft success rate (p < 0.001). No sensorineural hearing loss, retraction pockets, or cholesteatoma were observed postoperatively. CONCLUSION: Tympanoplasty offers a well-established surgical approach for restoring hearing function and preventing recurrent otorrhea. The asymmetric cartilage-perichondrium island graft design has emerged as a promising technique to achieve optimal functional and anatomical outcomes in tympanoplasty. LEVEL OF EVIDENCE: Level 4.

    Sağlık Bilimlerinde Manevi İyi Oluş: Türkiye'deki Nicel Tezlerin Sistematik Analizi

    Giriş: Manevi iyi oluş, sağlık bilimleri alanında giderek daha fazla ilgi gören bir konu haline gelmiştir. Türkiye’de bu alanda yapılan lisansüstü nicel tezlerin incelenmesi, alanın mevcut durumunu anlamak ve gelecekteki araştırmalara yön vermek açısından önemlidir. Bu çalışmanın amacı; sağlık bilimleri alanında, manevi iyi oluş konusundaki lisansüstü nicel tezleri belirlenen değişkenler açısından analiz etmektir. Yöntem:Yükseköğretim Kurulu Ulusal Tez Merkezi veri tabanında, 2014-2024 yılları arasında yapılan tezler kullanılmış“spiritual well-being”, “spiritüel iyi oluş”, “spiritüel iyilik hali”, “manevi iyi oluş”, “manevi iyilik”, “manevi iyi olma” ve “manevi iyilik hali” anahtar kelimeleri kullanılarak taranmıştır. Önceden belirlenmiş kriterlere göre 50 tez değerlendirmeye alınmıştır. Manevi iyi oluş konusunu içeren tez çalışmaları; yayımlandıkları yıllar, akademik düzeyleri, araştırma türleri, veri toplama yöntemleri, katılımcı sayısı ve grupları, çalışılan değişkenler ve tezlerin kalite değerlendirmeleri gibi çeşitli değişkenler açısından detaylı bir şekilde analiz yapılmıştır. Bulgular: Son yıllarda manevi iyi oluş konusundaki lisansüstü nicel tezlerin sayısında belirgin bir artış olduğu gözlemlenmiştir. 2024 yılında yayımlanan 16 tez ile en yüksek sayıya ulaşılmıştır. Çalışmaların çoğunlukla yüz yüze veri toplama yöntemiyle yapıldığı ve tanımlayıcı araştırmaların öne çıktığı belirlenmiştir. Katılımcı sayısı bakımından 151-300 kişilik gruplarla yapılan çalışmalar ağırlıktadır. Katılımcı grupları incelendiğinde, en fazla onkoloji hastaları ve kanser tanısı almış bireylerin yer aldığı görülmüştür. Manevi iyi oluş ile çalışılan değişkenlerin büyük çoğunluğunun “psikolojik ve duygusal” faktörler olduğu saptanmıştır. Sonuç: Sağlık bilimleri alanında manevi iyi oluş üzerine yapılan lisansüstü tezlerin sayısında artış olduğu görülmektedir. Bununla birlikte analiz edilen tezlerde manevi iyi oluşun daha çok psikolojik faktörler ile ilişkili olarak ele alındığı ve deneysel araştırmaların sınırlı olduğu ortaya konmuştur. Bu durum; alanda geliştirilmesi gereken noktaları ve mevcut eksiklikleri göstermektedir. Dolayısıyla bu çalışma, gelecekte yapılacak akademik çalışmalar için önemli bir yol haritası sunmaktadır.

    Although it is claimed that the concept of existence has no definition, many different definitions and interpretations have been made by philosophers who have pondered this field throughout history. While most ancient philosophers and scientists were interested in the problem of what existence is and what it is, the essence (origin) of existence was also a matter of curiosity. According to some, the first being or the essence of being was air, water, and fire. According to the others, they are concepts such as ‘One, logos, idea, substance, cogito, perception, monad, object, self, soul, will’ and power. While the problem of knowledge of existence (ontology) goes through many philosophical stages and focuses on it as a research topic, it can be seen that in later stages this focus shifts to what knowledge is and what it is (epistemology). The method of reaching true (absolute) knowledge has varied from age to age. While scientific and transmitted knowledge was popular in ancient times to reach true knowledge, positive science was used as a reference in reaching absolute knowledge in modern and post-modern ages. This scientific method, which centers on experiment and observation, has been accompanied by technical technological developments. While mass media, and in this context, cinema/television based on visuality, gained great momentum after the 19th century, the first impression created by the train of the Lumiere brothers coming towards people was replaced by panic and fear. Therefore, the truth between visuality and reality has been questioned. What is intended to be done in this study is to try to understand the relationship between existence, image, reality, and effect.

    Confusing Factors in the Tear Break-Up Time Test: The Influence of Contact on the Ocular Surface: Cross-Sectional Study

    Objective: The purpose of the present paper was to examine the changes in the break-up time (BUT) value of the staining procedure in the fluorescein BUT test. Material and Methods: The natural BUT values of the participants were determined with a non-invasive BUT (NI-BUT) test. Then, the changes in the tears were re-evaluated with the topographic break-up test method after the staining procedure was simulated with strips that mimicked fluorescein staining but did not contain fluorescein. The BUT test performed after the staining simulation was named sham-hybrid BUT test (SHBUT). Between the two tests, the first BUT value (BUT1st), the second BUT value (BUT2nd), the third BUT value (BUT3rd), the fourth BUT value (BUT4th), the fifth BUT value (BUT5th), and the average value of the first 3 break-ups (BUTA3) were compared. Results: Although the mean BUT1st value in NI-BUT test was found to be 4.4±2.2 seconds, it was 9.4±5.1 seconds in the SH-BUT test (p=0.000). Mean values of BUT2nd, BUT3rd, BUT4th, BUT5th and BUTA3 parameters in NI-BUT and SH-BUT tests were found to be 5.2±2.3 and 10.6±4.9 (p=0.000); 5.9±3.1 and 11.9±4.7 (p=0.000); 6.4±3.1 and 12.5±4.4 (p=0.000); 6.8±1 and 12.9±4.3 (p=0.000) and 5.2±2.4 and 10.6±4.8 seconds (p=0.000) respectively. All parameters were found to be longer in the SH-BUT test at statistically significant levels. Conclusion: The staining procedure for the fluorescein BUT test causes a prolongation of the BUT test. It is important to consider the changes in BUT because of these effects when performing the fluorescein BUT test.
