Article Citation - WoS: 0Analysis of legionella and Some Chemicals in Water Samples in Mardin Province(Kahramanmaras Sutcu Imam Univ Rektorlugu, 2022) Dündar, Abdurrahman; Yildirim, Idris; Dundar, Nagihan Demir; Department of Medical Services and Techniques / Tıbbi Hizmetler ve Teknikleri BölümüIn this study microbiological and chemical analyzes were carried out on water and swab samples taken from two water wells, a water tank and taps of buildings belonging to a public institution in Mardin Province. In these specimens the presence of Legionella and its serogroups as well as the parameters such as chlorine, nitrate, nitrite concentration, pH level and electrical conductivity were analyzed. While Legionella growth was not observed in 25 water samples, L. pneumophila serogroup 1 was detected in 4 of 25 swab culture samples taken from the same places. The pH values of water samples ranged from 7.53 to 8.02 the lowest pH value was observed in well no 1 as 7.53 while the highest was observed in well no 2 as 8.02. The lowest electrical conductivity value was measured at well 2 as 376.44 mu S cm(-1) and the highest was measured as 446.57 mu S/cm from well 1. As a result of the analyzes made in our research nitrite was not detected in any of the water samples. Whilst the nitrate amount was found to be the lowest with 4.30 ppm in the well, the highest with 10.85 ppm in the well 2. The chlorine values in the well 1 and 2 from which water samples were taken and the main tank were measured as 2.21, 2.05 and 2.43 ppm respectively. These values were found to be in the range of 1.68-1.73 ppm in the samples taken from the building taps.Article Citation - WoS: 11Citation - Scopus: 11Anoxybacillus sp. SO B1-immobilized Amberlite XAD-16 was developed. The functional groups of Anoxybacillus sp. SO B1 immobilized on Amberlite XAD-16 were characterized in KBr tablets by Fourier transform infrared (FT-IR) spectrometry. Various parameters such as pH, amount of the adsorbent, eluent type and volume, and flow rate of the sample solution were studied. The optimum pH values of quantitative sorption for Cu(II) and Pb(II) were found to be pH 7.0 and 5.0 and Cu(II) and Pb(II) ions could be quantitatively eluted with 5.0ml of 1.0mol L-1 HCI and 10.0ml of 0.25mol L-1 HNO3, respectively. Recoveries of Cu(II) and Pb(II) were found to be 100.9 +/- 1.57% and 100.3 +/- 0.49% (N = 5), the limits of detection of Cu(II) and Pb(II) in the determination by FAAS (3s, N = 10) were found to be 0.8 and 1.6 mu g L-1, respectively. The proposed enrichment method was applied for metal ion determination from water samples such as two parts of Tigris River water in Diyarbakir and Elazig, Lake of Hazar in Elazig, and tap water in Diyarbakir. Tests used are as follows: reducing power, free radical scavenging, superoxide anion radical scavenging, total antioxidant activity, metal chelating activitiy, etc.; total phenolic content was determined. The percentage inhibition of P. ostreatus, P. eryngii, P. florida, and P. sajor-caju at 20 mg/mL concentration on peroxidation in a -carotenelinoleic acid system was 57.19, 60.68, 62.12, and 58.81%, respectively. The reducing power of P. eryngii was higher than the other samples, and its value was 0.86 at 10 mg/mL concentration. P. ostreatus and P. sajor-caju proved to be better at scavenging superoxide anion radicals than the P. eryngii and P. florida. The biological activities of the complexes were also tested. Compounds 2 and 5 were the best complexes at DPPH radical scavenging and reducing power activity at 73.27 % and 0.41 at 200 μg/mL, respectively. The highest antimicrobial activity exhibited by complex 3 as 14 mm inhibition zone against S. aureus. All of the complexes have cleaved the DNA from the double-strand and exhibited three bands on gel electrophoresis. Pleurotus eryngii, in particular, is widely recognized for its organoleptic quality and favorable healthy effects, being commercially produced in great extent. The present study is just for evaluating the antioxidant capacity (scavenging DPPH radicals, reducing power, Total antioxidant activity by β-carotene-linoleic acid metod, Chelating effect on ferrous ions, superoxide anion radical scavenging activity ), antimicrobial effects (by using the disc diffusion method) and insufficiently studied cytotoxic activity (using PC-3 cell lines) of Pleurotus eryngii by using various solvent systems. For this reason; obtained extracts of the mushroom applying hexane, ethylacetate and methanol respectively were tested with in its biologic activities. As a result, The highest total antioxidant activity by β -carotene-linoleic acid metod was identified in methanol extract (63.62 mg /ml). The highest DPPH scavenging activity was obtained from ethyl acetate extract with 88.81 mg/ml. EDTA is used as standard in methal chelating activity. The highest metal chelating activity of Pleurotus eryngii identified in hexane (88,1 mg /ml) and ethylacetate (88.84 mg /ml) extracts. Additionally, in most of studied tests antioxidant activity of the mushroom increased with increasing concentration. On the other hand, the highest antimicrobial activity was obtained against Pseudomonas aeruginosa ATCC 27853 (11 mm) in ethyl acetate extract. However, the best antimicrobial activities were observed in hexane and methanol extracts for other tested microorganism. It was not determined any antimicrobial activity against Escherichia coli ATCC 25922. It has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Ethyl acetate extract showed significant inhibitory value at the concentrations of 530 µg/ml (86.20 %) against PC-3 cell lines. Using methanol extract, the maximun inhibition was observed at concentration of 680 µg/ml with value of 51.86 %. The adsorbents were characterized via Scanning Electron Microscopy (SEM), Energy Dispersive Spectrometer (SEM-EDS), and Fourier transform infrared spectroscopy (FT/IR). Fomes fomentarius presented a low sorption capacity contrary to the two other fungi (RB and AA). RB and AA were selected as potential adsorbents for BR18 and RBBR, respectively. The maximum BR18 removal efficiencies for 10, 25, and 50 mg/L were obtained after 60 min to be 90, 88, and 86%, respectively. The RBBR adsorption efficiencies were 96.4, 96, and 90% for dye concentrations of 10, 25, and 50 mg/L. The adsorption of BR18 onto the RB biomass followed the Freundlich isotherm, while Langmuir is the best isotherm for RBBR sorption’s elucidation onto AA fungus biomass. The removal of BR18 by BR biomass was found to follow the pseudo-second order. In contrast, the adsorption of RBBR onto the AA biomass followed Lagergren’s pseudo-first order. For both adsorbents, the adsorption was exothermic, feasible, and spontaneous in nature. Söz konusu makro mantarların etanol ekstraktlarının antioksidan potansiyellerinin tespiti amacıyla; DPPH serbest radikal yakalama, metal iyonlarını şelatlama, indirgeme gücü ve süperoksit anyon radikal süpürme testleri uygulanmıştır. Antimikrobiyal aktivitenin belirlenmesi amacıyla da disk difüzyon yöntemi çalışılmıştır. Süperoksit anyon radikal aktivitesinde tespit edilen en yüksek değer % 83,24 ile Morchella tomentosa en düşük değer ise % 73,05 ile Coprinus silviaticus'a ait bulundu. DPPH serbest radikalini söndürme aktivite tekniğinde en fazla etkinliği gösteren Amanita fulva etanolik ekstraktının 1.0, 2.0, 5.0, 10.0, 15.0, 20.0 mg/mL konsantrasyonlardaki etkinlik verilerinin sırasıyla %19.33, %24.46, %58.74, %77.75, %87.89, %88.40 olduğu tespit edilmiştir. Metal şelatlama etkinliğinde kullanılan ekstraktların ve pozitif kontrol olarak çalışılan standartların etkinlik sıralaması EDTA> Amanita fulva> Morchella tomentosa> Phallus impudicus> Coprinus silviaticus> BHT> BHA> Trolox şeklinde tespit edilmiştir. When we treated [Ru(arene)(μ-Cl)Cl]2 {arene:p-cymene,benzene} with chelating phosphinite ligands, we obtained neutral Ru(II)-complexes possessing the general formula [Ru(arene)phosphiniteCl2]. The structure of the ligands and complexes was confirmed using analytical and spectroscopic techniques. The quantum chemical calculations were carried out for the ruthenium complexes at the DFT/CAM-B3LYP level of theory in gas phase. The phosphinite complexes were subjected to cyclic voltammetry studies in order to determine the energies of HOMO and LUMO levels and to estimate their electrochemical and some electronic properties. Organic complex-based memory substrates were immobilized using TiO2-modified ITO electrodes, and the memory functions of phosphinite-based organic complexes were verified by chronoamperometry (CA) and open-circuit potential amperometry (OCPA). In the present study, the antioxidant potentials of ruthenium-based p-cymene and benzene complexes through DPPH radical scavenging, metal chelating, and reducing power activities were also determined. In addition, DNA binding abilities and antimicrobial activities of these complexes against pathogenic bacteria were studied. Serdar; Kılınç, Ersin; Dundar, Abdurrahman; Kilinc, Ersin; Department of Medical Services and Techniques / Tıbbi Hizmetler ve Teknikleri BölümüThe potential biosorption abilities of apple shell (AS), orange peel (OP), banana peel (BP), and millet waste (MW) as a bio-waste material to remove 2,4-dichlorophenoxy acetic acid (2,4-D), 2,4-dichlorophenoxy propanoic acid (2,4-DP), and 2,4-dichlorophenoxy butyric acid (2,4-DB) from aqueous solution were investigated in batch condition. Optimum biosorption conditions were determined as a function of contact time, pH of the solution, amount of biomass, and initial pesticide concentrations. The concentrations of the pesticides in the remaining solutions were simultaneously determined by high performance thin layer chromatography. Langmuir and Freundlich models were applied to describe the biosorption isotherm of the pesticides by agricultural wastes. The experimental adsorption data were fitted to Langmuir adsorption models (r(2)>0.99). It was found that adsorption of 2,4-DP was higher for all biosorbent. The highest maximum adsorption capacities of 2,4-DP were found as 40.08, 22.71, 33.26, and 45.45mg/g, respectively, for AS, OP, BP, and MW. Maximum adsorption capacity was obtained for 2,4-D as OP>MW>BP>AS, 2,4-DP as MW>AS>BP>OP, and 2,4-DB as OP>AS>BP>MW. From the results, it can be said that MW was an effective biosorbent for removal of 2,4-DP and OP was an effective biosorbent for removal of 2,4-D and 2,4-DB through the studied pesticides. Optimum biosorption conditions were determined as 60min of contact time, 100mg of biomass, AS at pH 6.0, OP at pH 6.0, MW at pH 7.0, and BP at pH 7.0. FT-IR was employed to understand the surface properties of biosorbents. According to the results, agricultural wastes have high adsorption capacity.Master Thesis COVID-19 tanısı konan hastaların cinsiyet ve yaş faktörlerinin bazı biyokimyasal değerlerle korelâsyonunun araştırılması(Mardin Artuklu Üniversitesi, 2022) Dündar, Abdurrahman; Dündar, Abdurrahman; Department of Medical Services and Techniques / Tıbbi Hizmetler ve Teknikleri Bölümü2019 yılının son döneminde Wuhan şehrinde ortaya çıkan Covid-19 hastalığı çok hızlı bir şekilde dünyaya yayılış göstermiş ve kısa bir süre sonra ülkemizde görülmeye başladı. Covid-19 ile ilgili önemli çalışmalar yapılmakta bu virüs ve benzeri virüs kökenli hastalıkların tespiti ve erken teşhisi ile tedavi amaçlı Crp, Lym, Ferritin, Kan Gazı ve D-dimer gibi bazı biyokimyasal değerlerin düzeylerinin hastalık şiddeti ve diğer biyokimyasal değerler ile ilişkisini değerlendirilmesi ve bu biyokimyasal değerlerin yaş, cinsiyet üzerindeki etkilerini değerlendirmeyi amaçladık. Yaptığımız çalışmada, hasta dosyalarının retrospektif incelenmesine dayalı kesitsel ve tanımlayıcı bir çalışmadır. 01 Haziran 2020 ile 30 Mayıs 2021 tarihleri arasında Kızıltepe devlet hastanesine başvuran ve hastalık tanısı konulan 12 aylık süreçte hastaların Crp, Lym, Ferritin, Kan Gazı ve D-dimer gibi bazı biyokimyasal değerleri ve aynı zamanda bu değerlerin diğer biyokimyasal değerler arasındaki ilişki ile bu değerlerin yaş ve cinsiyete olan etkileri incelendi ve analizleri yapıldı. Araştırma da elde edilen veriler SPSS for Windows 25.0 programı kullanılarak analiz edilmiştir. Verileri değerlendirilirken tanımlayıcı istatistiksel metotları (sayı, yüzde, ortalama, standart sapma) kullanılmıştır. Çalışmamız da inceleyeceğimiz tüm parametreleri eksiksiz olan 01 Haziran 2020 ile 30 Mayıs 2021 tarihleri arasında Kızıltepe devlet hastanesine başvuran ve COVID-19 tanısı koyulan 1812 hasta dahil edildi. Bu hastalardan %51,9'u (n: 941) kadın % 48,1 (n: 871) erkektir. Ve yaş ortalaması 56,58±19,89 olarak hesaplanmıştır. Yaptığımız çalışmaya göre Crp ve Ferritin değerleri erkeklerde daha yüksek, Lenfosit düzeyleri ise kadınlar daha yüksek olduğu saptandı. Ayrıca Crp, Ferritin ve D.Dimer değerleri ile yaş arasında pozitif yönde, Lenfosit ile yaş arasında ise negatif yönde anlamlı bir ilişki saptandı. Biyokimyasal değerlerin hastalığın klinik seyrinde önemli bir biyolojik belirteçler olduğu literatürde ki araştırmalar ile uyumludur. Covid-19 ile ilgili çok sayıda araştırma yapılmış ve literatüre kazandırılmıştır ve yeni çalışmalarda yapılmaktadır. Bizde biyokimyasal değerlerin Covid-19'un klinik seyrinde biyolojik belirteç olarak kullanılması ayrıca bu değerlerin yaş ve cinsiyet üzerindeki ilişkisinin tespitinin hastalığın seyrinde önemli olduğundan bu konu ile ilgili literatüre katkı sağlamayı amaçladık. Hastalığı daha ağır geçiren erkeklerde Crp ve Ferritin değerleri analiz sonuçlarına göre anlamlı yüksek olduğu görüldü. Lenfosit değerinin ise erkeklerde daha düşük olduğu saptandı. Phenolic acid composition of mushrooms was also analysed. L. deliciosus and F. fomentarius were generally showed the highest activities at antioxidant test systems (metal chelating, superoxide anion radical scavenging, total antioxidant, 2,2-diphenyl-1-picrylhydrazyl radical scavenging and reducing power activity tests). The highest activities at antimicrobial activity displayed by A. arvensis and as 18 ± 0.8 against to Staphylococcus aureus. The best IC50 values of mushroom methanol extracts at anticancer activities on HeLa and NRK-52E were 7.09 and 18.23 mg/mL exhibited by C. micaceus and A. campestris, respectively. The highest butyrylcholinesterase activity exhibited by L. deliciosus. Total amount of phenolic acids were found as 1,224.70 mg/kg at L. deliciosus. © 2016 The Author(s). The major FAs of TL, TG, PL in both species were Cl6:0 (palmitic açid, PA), Cl8:1 n-9 (oleic acid, OLA) and Cl8:2 n-6 (linoleic acid, LA). In both species, total PUFA amounts were found to be higher than total MUFA and total SFA in TL, TG and PL fractions. The efficient production of the fatty acids especially linolenic and oleic acids which are majorly needed in building blocks of dietary human has confirmed these species as good source of nutrition. On the other hand, insufficiently studied cytotoxic activity (using pC-3 cell lines) of these mushrooms were investigated by using various solvent systems Ethyl acetate extract of Russula delica and, Pleurltus ostreatus showed significant inhibitory value at the concentrations of 520-530 µg/ml (99,45%- 92,82%) against PC-3 cell lines with IC50; 274,53-297,77 µg /mL respectively. Methanol extracts did not show any cytotoxic activity. lt has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. The newly prepared compounds have been characterized by several spectroscopic techniques. All compounds are evaluated for their antioxidant and antibacterial potential. For the antioxidant studies, three tests are applied; DPPH (2,2-diphenyl-1-picrylhydrazylradical) scavenging, metal chelating and reducing power activity. Compound 4 exhibits the best DPPH scavenging activity as 35.2% at 100 mg/L concentration. The metal chelating activities of compounds 3 and 4 are 69.7% and 56.4%, respectively. Reducing power activities of compounds 3 and 4 are higher than alpha-tocopherol which is used as positive control. Method: In accordance with the purpose of the study, volunteers were asked to consume 100 ml of the extract every morning for 29 days. Three tubes of blood samples were taken from the volunteers on the 15th and 29th days of the study. Biochemical and hematological analysis of the blood samples were performed and immunomodulatory effects through cytokines were examined. The values obtained from 3 tubes of blood obtained from volunteers before the use of mushroom extract were used as control. The chemical composition and beta-glucan content of 100 ml of mushroom water extract were also analyzed. Result: IL-4, IL-6, IL-10 and IL-13 could not be detected because the values were below the lowest standard value. TNF-alpha, IFN-gamma and IL-1 beta 15th and 29th day values decreased compared to the 1st day (control) values (p < 0.05). However, there was no significant difference observed between the 15th and 29th day. No abnormalities were observed in biochemical and hematological values. Also, the beta-glucan content of extract was found 38.12 mg/100 ml. Conclusion: The frequency range of kidney and liver function test results confirmed that P. osreatus is a reliable food source. Considering the cytokine values these results indicate that P. ostreatus water extract has an anti-inflammatory effect. As no significant difference was observed in 29 days of use, it is thought that 15 days of daily consumption of the extract may be sufficient for the anti-inflammatory effect to occur. The azo Schiff base HL acts as a monobasic tridentate ligand, which commonly coordinates through the oxygen atoms of the phenol OH and the pyrimidine one group, and the nitrogen atom of the azomethine group. The thermal behaviors of the ligand and its metal complexes were studied using thermogravimetric analysis (TGA) and differential thermal analysis (DTA). The metal complexes proved to be more thermally stable than the ligand; they decomposed at 10-30 degrees C higher temperatures. Antioxidant properties of the ligand and its metal complexes (DPPH free radical scavenging, ferrous chelating and reducing power activities) were tested. Mantarların immünomodülatör etkileri çoğunlukla in- vitro ve in- vivo (deney hayvanlarında ve hasta bireylerde) olarak çalışılmış olmasına rağmen, özellikle ülkemizde ve dünyada gönüllü sağlıklı bireyler üzerinde yapılan çalışmalar birkaç adetle sınırlı olup, kapsamları da çok dardır. Bu nedenle yapılan çalışmada P. ostraetus'un su ekstraktları sağlıklı gönüllüler tarafından tüketildi. Mantar ekstraktının tüketimi öncesi ve sırasında gönüllülerden alınan kan örneklerinin biyokimyasal ve hematolojik analizleri ve sitokinler aracılığıyla immünomodülatör etkileri analiz edildi. Çalışmadan elde edilen bulgular incelendğinde IL-4, IL-6, IL-10 ve IL-13 değerleri en düşük standart değerin altında olduğundan tespit edilemediği görülmektedir.TNF-α, IFN-γ ve IL-1β 15. ve 29. gün değerleri 1. gün (kontrol) değerlerine göre azaldı (P< 0.05).Ancak 15. ve 29. gün arasında anlamlı bir fark gözlenmedi. Sonuç olarak biyokimyasal ve hematolojik değerlerde herhangi bir anormallik gözlenmedi ve böbrek ve karaciğer fonksiyon testi sonuçlarının sıklık aralığı P. osreatus'un güvenilir bir besin kaynağı olduğunu doğruladı. We highlight advantages and major challenges of the techniques and discuss future perspectives of both batch and column-based methods. Particular attention is paid to the preconcentration of metal ions using resin-immobilized microbacteria for solid phase extractions. We also discuss detection methods including UV-vis spectrophotometry, FAAS, ICP-OES and ICP-MS. Analytical figures of merit are compared, and examples are given for the application to a variety of samples including food, beverages, alloys, water, soil, and geological samples.
