For this study, the biological functions of Verbascum thapsus (VT) methanol extract were determined in vitro. The plant's methanol extract was created through the maceration process. The phytochemical composition of plant extracts was investigated using liquid chromatography-electrospray ionization tandem mass spectrometry. The antioxidant capacity of the extract was determined using the 2,2-diphenyl-1-picrylhydrazil (DPPH radical) and 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS radical). Cell lines Caco-2 (human colorectal adenocarcinoma cells), LNCaP (Lymph Node Carcinoma of Prostate), and HEK293 (Human embryonic kidney 293 cells) were used to model colon, prostate, and non-cancerous cells. The cytotoxic activity of the plant extract on the proliferation of these cells was determined by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, a tetrazole) assay protocol. VT extract showed moderate DPPH and ABTS radical scavenging activities at 30 mg/ml concentration. With this, VT extract was determined to inhibit acetylcholinesterase (AChE) enzyme and had strong cytotoxic activity on cancerous cell lines. In addition, our findings clearly showed that the plant extract had greater cytotoxic activity on the viability of cancerous cells compared to non-cancerous (Human embryonic kidney cells; HEK293) cells. The current findings showed that V. thapsus might be a promising anti-cancer medication candidate for the treatment of human colorectal adenocarcinoma and colon cancer, as well as a good source antioxidans. In patients waiting for organ transplantation, increased waiting time can increase anxiety by creating negative automatic thoughts. The aim of this study is to examine the relationship between corneal transplant recipients' negative automatic thoughts and anxiety before organ transplantation. Material and methods. The population of this correlational descriptive study consisted of 108 patients waiting for cornea transplantation in the Eye Bank unit of a hospital in the south east region of Türkiye. The data were obtained from the patients presented to the ophthalmology clinic for transplantation between September and December 2022. A patient information form, the Negative Automatic Thoughts Scale, and the Trait Anxiety Scale were used to collect the data. Percentage distribution, arithmetic mean, logistic regression and correlation analysis were used in the data analysis. Results. It was determined that the mean score of the negative automatic thoughts of the transplant patients participating in the study was 100.69±47.83, and the trait anxiety mean score was 53.06±6.07. There was a positive relationship between negative automatic thoughts and trait anxiety at a moderate level, which was statistically significant (p<0.05, r=0.53). Conclusion. It was determined in the study that the patients' negative automatic thoughts and trait anxiety were high, and their anxiety increased as the negative automatic thoughts increased.Article Citation - WoS: 5Citation - Scopus: 4Biosynthesis of AgNPs by extract from waste leaves of Citrullus lanatus sp. Green synthesis is very important for biomedical applications as they show biocompatible properties. In this study, AgNPs were easily synthesized using the environmentally friendly green synthesis approach using agricultural waste parts of Citrullus lanatus sp. plant grown in Diyarbakır region. Characterization of synthesized AgNPs was made. Fourier Transform Infrared Spectroscopy (FTIR) analysis was used to evaluate the phytochemicals responsible for effective reduction in the formation of AgNPs. UV-visible spectrophotometer (UV-Vis.) Spectra were also used to determine the presence of AgNPs. X-Ray Diffraction Diffractometer (XRD), Scanning Electron Microscope (SEM), Transmission Electron Microscope, Zeta potential analyzes were performed to define the crystal structures, dimensions and surface charges of AgNPs, respectively. In these data, it was determined that AgNPs showed maximum absorbance at 460 nm, spherical appearance, 21.27 crystal nano size and -30.05 mV zeta potential. Antimicrobial effects of AgNPs on gram positive Staphylococcus aureus (S. aureus) and Bacillus subtilis (B. subtilis) bacteria, gram negative Escherichia coli (E. coli) and Pseudomonas aeruginosa (P. aeruginosa) bacteria, as well as on fungus C. albicans pathogenic microorganisms It was analyzed by specifying the Minimum Inhibition Concentration (MIC) by microdilution.Article ÇOCUKLUK DÖNEMİNDE GÖRÜLEN DUYGUSAL VE DAVRANIŞSAL SORUNLARLA İLGİLİ TÜRKİYE’DE YAPILAN LİSANSÜSTÜ TEZLERİN İNCELENMESİ(İnönü Üniversitesi Sağlık Hizmetleri Meslek Yüksekokulu Dergisi, 2019) AYANOĞLU, MineBu çalışmanın amacı, duygusal ve davranışsal sorunlarla ilgili Türkiye’de yapılan lisansüstü tezlerin incelenmesidir. Araştırmada nicel araştırma yöntemlerinden betimsel tarama modeli kullanılmıştır. Araştırmanın örneklemini Yüksek Öğretim Kurumu Ulusal Tez Merkezi internet sitesinden ulaşılan 145 yüksek lisans, doktora ve tıpta uzmanlık tezleri oluşturmaktadır. Araştırmada veri toplama aracı olarak, araştırmacılar tarafından araştırmanın amacına uygun olarak hazırlanan veri toplama formu oluşturulmuştur. İncelenen tezler, veri toplama formunda yer alan sorular doğrultusunda incelenmiş, elde edilen verilerin frekans ve yüzdeleri analiz edilmiştir. Araştırma sonucunda duygusal ve davranışsal sorunları içeren en fazla çalışmanın tıpta uzmanlık tezi (%44.83) olduğu; yıllara göre bakıldığında en fazla 2000-2009 yılları arasında (%41) çalışma yapıldığı saptanmıştır. Çalışma gruplarına bakıldığında en fazla farklı yaş grubundaki çocuklar (%39.31) üzerinde çalışmaların yapıldığı; lisansüstü çalışmalarında en fazla amaçsal örnekleme tekniğinin (%58.62) kullanıldığı görülmüştür. Hastalık türü/çalışılan sorun türüne göre en fazla birden fazla bozukluk türünü aynı anda çalışılan (%50.34) lisansüstü tezlere rastlanmıştır. İncelenen lisansüstü tezlerin çoğunlukla tam metin (%79.31) olarak ulaşıldığı; anabilim dalları incelendiğinde duygusal ve davranışsal sorunlarla ilgili lisansüstü çalışmaların çoğunluğunun Çocuk Ruh Sağlığı/Çocuk Ergen Ruh Sağlığı ve Hastalıkları anabilim dalında (%30.34) yayınlandığı tespit edilmiştir. N-acetyl cysteine (NAC) is used as the APAP antidote. Cyclosporin A (CsA) is suppressed mitochondrial damage by binding cyclophilin, a mitochondrial pore transport component. The study aimed to evaluate the effects of NAC, CsA, and NAC+CsA treatments on APAP-induced hepatotoxicity in mice. Mice were randomly divided into five groups (n = 6). 400 mg/kg/ip/single dose APAP, 1200 mg/kg/i.p/single dose NAC and 50 mg/kg/i.p/single dose CsA were performed. Light and electron microscopic alterations were investigated in liver samples. Levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and liver glutathione (GSH) were analyzed. 3-nitrotyrosine and cytochrome c immunoreactivities were evaluated in liver tissue. Here, we found that APAP leads to histopathological and ultrastructural changes in mice liver. Also, APAP increased cytochrome c and 3-nitrotyrosine immunopositive staining. Besides, a significant decrease in liver GSH and an increase in serum AST and ALT levels were detected in the APAP group. Interestingly, NAC+CsA treatment improved histological alterations, cytochrome c, and 3-nitrotyrosine immunoreactivities and liver GSH, serum AST/ALT levels caused by APAP. We suggest that the combination of NAC and CsA reduces acetaminophen-induced hepatotoxicity in mice.Article Effect of Delivery Method on Breastfeeding Status in Primiparous Mothers Within the First 24 Hours(Springer Publishing Co, 2024) Yesil, Yesim; Oner, Serap; Celik, Gulbiye Dinc; Bay, Habibe; Eksioglu, Aysun; Turfan, Esin CeberObjective: This study aims to evaluate the effect of the delivery method on breastfeeding status of primiparous mothers during the first 24 hours. Methods: In this descriptive and analytical study, the sample consisted of 126 primiparous mothers with vaginal (n = 54) and cesarean (n = 72) deliveries. The research data were collected in a hospital through a personal information form and LATCH. Breastfeeding was assessed at the 6th, 12th, and 24th hours within the first 24 -hour period after the delivery. Results: A total of 83.3% of mothers with vaginal delivery started out breastfeeding in the first hour, while the rate was 43.1% for mothers with cesarean delivery. A total of 94.4% of mothers who had vaginal birth gave their babies colostrum, whereas the rate was 80.6% for mothers who had a cesarean birth. While there were no statistically significant differences in Applications 1 and 3 between the LATCH scores of mothers with vaginal and cesarean deliveries, Application 2 produced a significant difference (p < .05). Conclusions: Delivery method influenced the breastfeeding behavior and had an impact particularly on initiation of breastfeeding in the first hour.Article The Effect of N-acetylcysteine on Amylase, Electrolytes, Vitamins and Nitrosative Stress Levels in Rats Treated with Maras Powder(Eastern Journal of Medicine, 2020) Unsal, Velid; Belge Kurutaş, ErgülThe aim of this study is to investigate the effects of N-acetylcysteine (NAC) on amylase, electrolytes, vitamins and nitrosative stress levels in the plasma of rats treated with smokeless tobacco "maras powder". Our study consisted of three groups. Control (n = 10), the group using maras powder (n = 10), maras powder+ NAC group. To the Maras powder group, 200 mg maras powder was placed in the sublingual area under general anesthesia. It was waited for 15 minutes for Maras powder to be absorbed through mucosa. This procedure was repeated once a day and for 7 days. To the NAC group, 200 mg of Maras powder was given as in the Maras powder group and NAC was injected intraperitoneally with a dose of 100 mg / kg / day. On the 8th day, the levels of amylase, vitamins (A, C and E), electrolytes (Na+, K+, Cl- ) and, as the biomarkers of nitrosative stress, nitric oxide (NO) and nitrotyrosine (3-NTx) in the plasma of all three groups were measured by the methods of ELISA. It was determined that amylase, 3-NTx, NO, electrolyte levels increased in the group using Maras powder compared to the control and NAC groups, but vitamin levels decreased. (p <0.05). It was detected that the levels of amylase, 3-NTx, NO, electrolyte in the group treated with NAC decreased compared to the group using Maras powder, whereas vitamin levels significantly increased and approached to the values of the control group. Smokeless tobacco 'Maras powder' increases nitrosative stress and distorts the levels of vitamin A, E, C and electrolyte in the plasma. NAC has a positive function in reducing the damage caused by Maras powder.Article Citation - WoS: 4Citation - Scopus: 6Effect of seasons on the fatty acid profile of total lipid, phospholipid and triacylglycerol in Mastacembelus mastacembelus (Atatürk Dam Lake, Turkey).(2018) Kaçar, Semra; Başhan, Mehmet; Oymak, AhmetThe present study investigated the lipid content and FA profiles of total lipids, phospholipids (PL) and triacylglycerols (TAG) of Mastacembelus mastacembelus from the Atatürk Dam Lake (Turkey). The results showed that the total lipid content of the dorsal muscle varied seasonally from 0.50% to 3.59%. Wide ranges of saturated fatty acids (SFAs) (36.21 to 50.52%) and polyunsaturated fatty acids (PUFAs) (25.22 to 42.02%) were found in the PL fraction. However, higher monounsaturated fatty acids (MUFAs) (37.63 to 45.07%) and SFA (34.71 to 38.82%) were found in TAG (triacylglycerol). The ratios of ω-3 to ω-6 PUFA ranged from 0.65 to 1.32 and 1.07 to 3.48 in PL (phospholipid) and TAG fraction, respectively. The results also showed that the major components were palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1ω-9), palmitoleic acid (C16:1ω-7), docosahexaenoic acid (DHA, C22:6ω-3), arachidonic acid (AA, C20:4ω-6) and docosapentaenoic acid (C22:5ω-3) in total lipid, C16:0, C18:1ω-9, AA and DHA in PL, C16:0, C16:1ω-7, C18:1ω-9, linoleic acid (LA, C18:2ω-6), and DHA in TAG extracted from the muscle of M. mastacembelus in all seasons.Article Citation - WoS: 2Citation - Scopus: 3Effects of Concomitant Use of N-Acetylcysteine and Cyclosporine a on Acetaminophen-Induced Acute Kidney Injury in Mice(Agricultural Research Communication Centre, 2024) Kocamueftueoglu, Gonca Ozan; Tektemur, Nalan Kaya; Guzel, Elif Erdem; Tektemur, Ahmet; Ozan, Ibrahim EnverBackground: Acetaminophen (APAP), a commonly used analgesic, causes acute kidney injury (AKI) in overdose although it is rare. Mitochondrial dysfunction plays a major role in the pathophysiology of renal damage, although the exact molecular mechanism is unknown. This study aimed to evaluate the potential therapeutic effect of cyclosporin A (CsA), a mitochondrial membrane permeability Methods: Male BALB/c mice were divided into Control, APAP, APAP+NAC, APAP+CsA and APAP+NAC+CsA groups (n=6). A single dose of APAP (400 mg/kg) was administered intraperitoneally. All other treatments (1200 mg/kg NAC, 50 mg/kg CsA) were performed intraperitoneally 3 h after APAP administration. All animals were decapitated and blood samples and kidney tissue samples were collected for evaluation. Serum creatinine (Cr) and blood urea nitrogen (BUN) levels were measured. The kidney tissue 8-hydroxydeoxyguanosine (8-OHdG), cytochrome c (Cytc) and 3-nitrotyrosine (3-NT) levels and cytochrome c (Cytc) expressions were determined. Result: Increased Cr and BUN levels, histopathological examinations and expressions of 8-OHdG, 3-NT and Cytc were detected in the APAP group. Combined NAC+CsA treatment sufficiently reversed oxidative stress, serum Cr and BUN levels and histopathological alterations induced by APAP. Moreover, cytc levels and renal tubular injury were remarkably reduced by combined drug treatment compared to the APAP+NAC group. These data suggest that the therapeutic effect of combined NAC+CsA treatment in mice with APAP-induced nephrotoxicity can be related to the combination of the antioxidant effect of NAC and the mitochondrial MPTP inhibitor effect of CsA.Article Citation - WoS: 2ERUCA SATIVA MILL SEEDS OIL ALLEVIATES HYPERLIPIDEMIA AND NON-ALCOHOLIC FATTY LIVER DISEASE IN SYRIAN HAMSTER(2022) Alhilal, M; Sulaiman, Y A M; Subuh, A M; Habra, N; Alhilal, SThe impact of oils rich in long chain monounsaturated fatty acids (LCMUFA) against hyperlipidemia and non-alcoholic fatty liver disease (NAFLD) has been inadequately described. In addition, the chemical solvents and the high temperature used in vegetable oils extraction process from seeds cause severe loss of many vital compounds. So the goal of this paper was to examine the effect of cold pressed Eruca sativa Mill seeds oil (ESSO), as a source of LCMUFA, on hyperlipidemia and NAFLD in Syrian hamster. The ESSO content of fatty acids was analyzed using chromatographic methods. Fifty two (52) healthy male golden Syrian hamsters used in this experiment were randomly divided into 4 groups (Completely Randomized Design). Negative control group, CHD group, positive control group and ESSO group. This experiment was achieved in two periods. The first period continued 4 weeks, in which hyperlipidemia and NAFLD were induced in CHD, positive control and ESSO groups through feeding on a hyperlipidemic diet. The second period also lasted 4 weeks, in which ESSO was orally gavaged at 2 g/kg of the body weight daily to animals of ESSO group. The levels of total cholesterol (TC), triglycerides, HDL-C and glucose and the activities of ALT, AST, ALP, LDH and CK were analysed in the serum. One way analysis of variance (ANOVA) followed by Duncan's multiple range test was used for statistical analysis. The consumption of hyperlipidemic diet for 4 weeks caused a significant raise (P<0.05) of triglycerides, glucose, ALT, AST, LDH, CK and a significant reduction (P<0.05) of the HDL-C/TC ratio, at the same time created lipid accumulation in liver cells in CHD, positive control and ESSO groups in comparison with negative control group at the end of the first period. These negative influences were alleviated in ESSO group by administration of ESSO at the end of second period. This study was conducted with 1234 nurses and midwives. A multiple regression model was created to see and predict the effect on individuals' total innovative scores and self-efficacy scores. The total innovative and the self-efficacy score are predicted with multiple regression analyses. It was observed that the variable that most affected both the total innovative score and the self-efficacy score of the individuals was the education level of the individuals. The fact that midwives and nurses have a certain level of innovative and self-confidence is important for the society to receive better and faster health services. In this study, it was observed that the education level was important for the development of innovative and self-confidence in both groups.Article Citation - WoS: 20Citation - Scopus: 24Experimental and theoretical investigation of adsorption and inhibition properties of 2-Amino-1,3,5-triazine-4,6-dithiol against corrosion in hydrochloric acid solution on mild steel(Elsevier, 2023) Arslanhan, Selim; Yıldız, Reşit; Döner, AliIn this work, 2-Amino-1,3,5-triazine-4,6-dithiol (2-ATD) as novel and high efficiency corrosion inhibitor has been investigated for mild steel (MS) corrosion in 0.5 M HCl solution using electrochemical methods, scanning electron microscopy (SEM), energy disperse X-ray spectroscopy (EDX), atomic force microscopy (AFM) and quantum chemical calculation methods. Potentiodynamic polarization (PDP) curves indicate that 2-ATD is mixed type inhibitor, corrosion inhibition efficiency increased with increasing inhibitor concentration and reached its value of 96.5%. Evolution of exposure time versus corrosion behavior of 2-ATD is examined in corrosive medium. While corrosion potential (Ecorr) shifted more negative values, polarization resistances (Rp) decreased after 120 h exposure time due to the corrosion process. H2 volume is measured in uninhibited and inhibited solutions (10 mM 2-ATD) after 120 h exposure time. Very low volume (3.6 mL cm-2) of H2 is obtained on MS electrode in inhibited solution after 120 h of exposure, indicating that 2-ATD covers the entire surface against aggressive attack and retards the both anodic dissolution of MS and cathodic hydrogen evolution reactions. The adsorption process proposal is the Langmuir isotherm which is most suitable. Adsorption and thermodynamic parameters show that 2-ATD has a strong adsorption effect onto MS surface and includes mixed adsorption style (physical and chemical). Corrosion current density increases with increasing temperature and high activation energy (Ea) proves the strong adsorption of 2-ATD on the MS surface. Anti-corrosion mechanism of 2-ATD is described more detail with the potential of zero charge method. SEM, EDX and AFM analysis support the obtained results of electrochemical methods and confirm the existence of protective layer and strong adsorption of 2-ATD on the MS surface. Chronoamperometry test shows that current densities are almost constant whole experiment in the presence of organic film. Finally, quantum chemical calculation method of 2-ATD in blank solution is performed to investigate the active sites for possible attachment with MS surface.Article Factors Affecting the Burden and Quality of Life of Caregivers for Gynecological Cancer Patients(Archives of Health Science and Research, 2022) Yaşar, Beril Nisa; Terzioğlu, FüsunObjective: The aim of this study is to evaluate the care burden and quality of life of caregivers of patients with gynecologic cancer. Material and Methods: This descriptive study was conducted with 251 individuals who provided care to patients with gynecologic cancer in gynecologic oncology services of a university hospital and a state oncology hospital. The data were collected by a semi-structured questionnaire form, Caregiver Burden Inventory (CBI), and Quality of Life Family Version. Results: The mean age of the participants was 39.76 ± 14.20; 79.3% of them were female, 65.3% were married, 28.7% were high school graduates, 57.4% were employed and 56.2% were unemployed before starting care, 91.2% had health insurance, 58.6% perceived their income as moderate, and 57% had relatives of the patient receiving care. Care burden was statistically significant in terms of gender, marital status, educational level, and degree of affinity with the patient (P < .005). The correlation between caregivers’ quality of life, gender, health insurance, income status, and the degree of affinity with the patient was statistically significant (P < .005). (almond tree) growing in the Mardin region. Characterization of AgNPs obtained by biosynthesis: Fourier Transform Infrared Spectroscopy (FTIR), X-Ray Diffractometer (XRD), UV-visible spectrophotometer (UV-Vis.), Scanning Electron Microscope (SEM), Transmission Electron Microscope(TEM), Zeta potential and Zeta size analysis was done through data. It was determined that AgNPs have maximum absorbance at 443 nm wavelength, they exhibit 14.67 crystal nano size, -19.9 mV zeta potential in spherical appearance. Astragalus species contained palmitic (C 16:0) (20.13%-53.8%), linoleic (C 18:2 omega-6) (13.25%-41.06%), oleic (C 18:1 omega-9) (5.78%-25.7%), linolenic (C 18:3 omega-3) (6.1%-22.89%), and stearic acid (C 18:0) (5.16%-13.1%) in the roots and linolenic (8.9%-51.42%), palmitic (20.16%-44.88%), linoleic (7.34%-27.57%), oleic (2.45%-17.91%), and stearic acid (4.28%-8.31%) in the shoots as major components. In this study the fatty acid composition of Astragalus species showed uniform fatty acid patterns. Palmitic and stearic acids were the major saturated and linoleic and linolenic acids were the major unsaturated fatty acids in the roots and shoots.Article Citation - WoS: 14Citation - Scopus: 16Green synthesis of gold nanoparticles from prunus cerasifera pissardii nigra leaf and their antimicrobial activities on some food pathogens(Progress in Nutrition, 2021) Hatipoğlu, AbdulkerimIn this study, a new and easy method for the biosynthesis of gold nanoparticles (AuNPs) with Prunus cerasifera pissardii nigra (PC) leaf extract as a reducing and stabilizing agent was presented. The nanoparticles were demonstrated a characteristic peak at the maximum wavelength of 535 nm with colour change as a result of the ultraviolet (UV)-visible spectrophotometer analysis data. Emission Scanning Electron Microscopy (FE-SEM), Transmission Electron Microscopy (TEM) and energy-dispersive X-ray spectroscopy (EDX) analyzes revealed that the crystal size of the synthesized AuNPs was below 20 nm and the morphological structure was mostly spherical. The size of the crystal structures of AuNPs was calculated as 17.94 nm from the X-ray diffraction (XRD) analysis data. Fourier Transform Infrared (FT-IR) Spectroscopy results confirmed the involvement of various biomolecules in the reduction and stabilization of PC-AuNPs. The zeta potential of the synthesized nanomaterial was measured as -27 mV. The average size of AuNPs was determined as 103.8 nm with Zetasizer. It was determined that AuNPs have strong inhibitory activity against Escherichia coli, Staphylococcus aureus, Bacillus subtillis and Pseudomonas aeruginosa and Candida albicans.Article Hastaların ve Hemşirelerin Algıladığı Bakım Davranışları ve Bakım Memnuniyeti(2023) Akça,Derya; Tanrıverdi, Seher; Pehlivan,SedaAmaç: Hemşirelerin, kendi bakım davranışlarını değerlendirmesi ve aynı bakım davranışlarının hastalar tarafından nasıl değerlendirildiğini anlaması oldukça önemlidir. Bu araştırma, hastaların ve hemşirelerin algıladığı bakım davranışları ve bakım memnuniyeti arasındaki ilişkinin belirlenmesi amacıyla yapılmıştır. Yöntem: Tanımlayıcı olarak yapılan bu araştırma, iç hastalıkları ve cerrahi kliniklerinde çalışan 119 hemşire ve bakım verdikleri 100 hasta ile gerçekleştirilmiştir. Veriler, hastalar ve hemşireler için ayrı veri toplama formları kullanılarak yüz yüze görüşme yöntemi ile toplanmıştır. Veri toplama formları; bireyin tanıtıcı özellikleri formu, Vizüel Analog Skala ve Bakım Davranışları Değerlendirme Ölçeği-Kısa Formundan oluşmuştur. Bulgular: Algılanan bakım davranışları toplam puan ortalaması, hastalarda ve hemşirelerde sırasıyla; 88,50 (SS=23,14) ve 111,44 (SS=16,52) olarak bulunmuştur. Bakım memnuniyeti puan ortalaması, hastalarda ve hemşirelerde sırasıyla; 6,66 (SS=2,07) ve 7,42 (SS=2,12) olup hemşirelerin algıladığı bakım davranışları (p<0,001) ve memnuniyet düzeyleri (p<0,01) hastalara göre istatistiksel olarak anlamlı bir şekilde yüksek saptanmıştır. Hastaların ve hemşirelerin algıladığı bakım davranışları puanı ile memnuniyet puanı arasında istatistiksel olarak olumlu yönde ilişki bulunmuştur (sırasıyla 0,591; 0,316; p<0,01). Sonuç: Hastaların ve hemşirelerin algıladığı bakım davranışları ve bakım memnuniyetinin orta düzeyin üzerinde olduğu ve algılanan bakım davranış düzeyi arttıkça bakım memnuniyetinin arttığı sonucuna ulaşılmıştır. İş yükü, çalışanlar ve örgütler bakımdan çok önemli bir değişkendir. AMAÇ: Bu araştırmanın amacı, bir devlet hastanesinde çalışan sağlık çalışanlarının çalışma ortamlarına ilişkin iş yükü algı düzeylerini ortaya koymaktır. YÖNTEM: Araştırmada veri toplama aracı olarak, sağlık çalışanlarının çalışma ortamlarına ilişkin algılarını belirlemeye yönelik “Bireysel İş Yükü Algı Ölçeği” ve “Kişisel Bilgi Formu” kullanılmıştır. Bu ölçek Cox tarafından 2003 yılında geliştirilmiş ve Cox ve arkadaşları tarafından 2006 yılında geçerlilik ve güvenirliği test edilmiştir. Saygılı tarafından 2008 yılında Türkçeye uyarlanmıştır. Araştırmanın evrenini araştırma izni alınabilen devlet hastanesinde görev yapan tüm sağlık personeli (421 kişi) oluştururken, örneklemini bu hastanede çalışan tüm sağlık çalışanlarından araştırmaya katılımda gönüllü olan 220 kişi oluşturmuştur. Yüz doksan beş sağlık çalışanından kullanılabilir veri toplanmıştır. Araştırmanın verileri Ekim- Kasım 2016 tarihleri arasında toplanmıştır. Araştırma verileri Istatistical Package for Social Science for Windows 16.0 programı kullanılarak sayı, yüzdelik ve ortalama-standart sapma, bağımsız değişkenler ile ölçek alt boyutları arasındaki ilişkilerinin değerlendirilmesinde ise bağımsız gruplarda t- Student testi analizleri kullanarak incelenmiştir. BULGULAR: Bu araştırmada, ölçeğin toplamından alınan puan ortalaması 103,7±22,07’dir. Ölçeğin alt boyutlarından alınan puan ortalamaları ise; yönetim desteği alt boyutu puan ortalaması 3,63±0,66, ortalama meslektaş desteği alt boyutu puan ortalaması 3,73±0,71, birim desteği alt boyutu puan ortalaması 2,77±0,58, çalışma ortamı algısı alt boyutu puan ortalaması 3,52±0,84, ve mevcut işi sürdürme niyeti alt boyutu puan ortalaması 1,04±0,30 olarak bulunmuştur. Glutathione-S-transferase enzymes play a critical role in the detoxification of xenobiotics in living systems. This study aimed to examine the interaction of methylene blue with human erythrocyte glutathione-S-transferase purified in one step. Human erythrocyte glutathione-S-transferase was purified with approximately 750-fold purification and 30% efficiency by glutathione agarose affinity chromatography. The results showed that the enzyme was inhibited by methylene blue with an IC50 value of 1.40 mmol/L. The Ki constant of methylene blue was 1.17 mmol/L. The Lineweaver–Burk graph of the methylene blue showed that the type of inhibition was compatible with mixed type inhibition. A new third spot was also detected by thin-layer chromatography. Furthermore, the cytotoxicity of methylene blue on human erythrocytes was evaluated and it was found that the haemolysis per cent of methylene blue on erythrocytes was approximately 14%.
