It is now the second largest cause of death in the world. The present study concerned antioxidant, anticancer and anticholinesterase activities and protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in methanol extracts of flowers, fruits and seeds of Hypericum amblysepalum. Materials and Methods: Antioxidant properties including free radical scavenging activity and reducing power, and amounts of total phenolic compounds were evaluated using different tests. Protocatechuic, catechin, caffeic acid, syringic acid, p-coumaric acid and o-coumaric concentrations in extracts were determined by HPLC. Cytotoxic effects were determined using the MTT test with human cervix cancer (HeLa) and rat kidney epithelium cell (NRK-52E) lines. Acetyl and butyrylcholinesterase inhibitory activities were measured by by Ellman method. Results: Total phenolic content of H. amblysepalum seeds was found to be higher than in fruit and flower extracts. DPPH free radical scavenging activity of the obtained extracts gave satisfactory results versus butylated hydroxyanisole and butylated hydroxytoluene as controls. Reducing power activity was linearly proportional to the studied concentration range: 10-500 µg/mL LC50 values for H. amblysepalum seeds were 11.7 and 2.86 respectively for HeLa and NRK-52E cell lines. Butyryl-cholinesterase inhibitory activity was 76.9 ± 0.41 for seed extract and higher than with other extracts. Conclusions: The present results suggested that H. amblysepalum could be a potential candidate anti-cancer drug for the treatment of human cervical cancer, and good source of natural antioxidants.Article Citation - WoS: 22Citation - Scopus: 25Biosorption of chlorophenoxy acid herbicides from aqueous solution by using low-cost agricultural wastes(TAYLOR & FRANCIS INC, 2015) Dündar, Abdurrahman; Celik, K. Serdar; Ozdemir, Sadin; Dundar, Abdurrahman; Kilinc, Ersin; Department of Medical Services and Techniques / Tıbbi Hizmetler ve Teknikleri BölümüThe potential biosorption abilities of apple shell (AS), orange peel (OP), banana peel (BP), and millet waste (MW) as a bio-waste material to remove 2,4-dichlorophenoxy acetic acid (2,4-D), 2,4-dichlorophenoxy propanoic acid (2,4-DP), and 2,4-dichlorophenoxy butyric acid (2,4-DB) from aqueous solution were investigated in batch condition. Optimum biosorption conditions were determined as a function of contact time, pH of the solution, amount of biomass, and initial pesticide concentrations. The concentrations of the pesticides in the remaining solutions were simultaneously determined by high performance thin layer chromatography. Langmuir and Freundlich models were applied to describe the biosorption isotherm of the pesticides by agricultural wastes. The experimental adsorption data were fitted to Langmuir adsorption models (r(2)>0.99). It was found that adsorption of 2,4-DP was higher for all biosorbent. The highest maximum adsorption capacities of 2,4-DP were found as 40.08, 22.71, 33.26, and 45.45mg/g, respectively, for AS, OP, BP, and MW. Maximum adsorption capacity was obtained for 2,4-D as OP>MW>BP>AS, 2,4-DP as MW>AS>BP>OP, and 2,4-DB as OP>AS>BP>MW. From the results, it can be said that MW was an effective biosorbent for removal of 2,4-DP and OP was an effective biosorbent for removal of 2,4-D and 2,4-DB through the studied pesticides. Optimum biosorption conditions were determined as 60min of contact time, 100mg of biomass, AS at pH 6.0, OP at pH 6.0, MW at pH 7.0, and BP at pH 7.0. FT-IR was employed to understand the surface properties of biosorbents. The essential oil components determined by GC/MS instrument. The antimicrobial activity determined by disc diffusion method against the gram negative bacteria, namely Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, the gram positive bacteria namely Staphylococcus aureus ATCC 25923, Streptococcus pyogenes ATCC19615 and Candida albicans ATCC10231 as the yeast. The antioxidant activities of essential oils were carried out by DPPH free radical scavenging activity method. It was determined that the main components of the anise essential oil were trans-anethole (52.94%) followed by iso-anethole (13.89%), caryophllene oxide (8.55%) and caryophyllene (29.4%); the main components of the cumin essential oil were β-pinene (15.77%), α-terpinene (15:52%), 1-Phenyl-1-butanol (15:13%), cumic aldehyde (12.74%) respectively. Anise essential oil showed weak antimicrobial activity and cumin essential oil showed moderate antimicrobial activity against test microorganisms. Cumin essential oil showed strong antimicrobial activity against C. albicans with 22±0.9 mm inhibition zone diameter. P. aeruginosa showed resistance to both essential oils. The antioxidant activity of cumin essential oil (75.60%) was observed closer to ascorbic acid (78.75%), higher than BHA (50.45%) and BHT (23.54%). Although anise essential oil exhibited low activity, it has the similar activity with BHT. To increase the efficiency, we univariately optimized the pH, buffer volume, CAMNP mass, eluent concentration and volume, mixing type and time, temperature, and interaction time. The study identified the limits of detection and quantification values for CAMNP-SPE-SQT-FAAS under the specified optimum conditions to be 12.30 and 41.00 mu g L-1, respectively. The newly developed method improved the detection power of traditional FAAS by 13.25 times. The amounts of protocatechuic acid, catechin, caffeic acid and syringic acid in methanol extracts were determined by HPLC. Total phenolic content of H. retusum seed extract was found more than fruit and flower extracts. The DPPH free radical scavenging activity of flower and seed methanol extracts showed close activity versus BHT as control. Among three extracts of H. retusum only flower methanol extract was exhibited considerable cytotoxic activities against to HeLa and NRK-52E cell lines. Moreover, seed methanol extract showed both acetyl and butyrl-cholinesterase inhibitory activity. The highest anti-genotoxic effects were seen 25 and 50 μg/mL concentrations. In this study, the extracts showed a strong antioxidant and anti-genotoxic effect. The seed extract was more efficient- than extracts of fruit and flowers. Our results suggest that the antioxidant and anti-genotoxic effects of extracts depend on their phenolic contents. The features of the azo Schiff bases were assigned from microanalytical, spectroscopic (IR, UV/Vis., H-1- and C-13 NMR, API-ES mass), magnetic, and molar conductivity measurements at room temperature as well as thermal analysis. The electronic absorption spectroscopy and magnetic susceptibility measurements of the complexes indicate square pyramidal arrangement for Pd-II and octahedral environment for all the other complexes. The azo Schiff base HL acts as a monobasic tridentate ligand, which commonly coordinates through the oxygen atoms of the phenol OH and the pyrimidine one group, and the nitrogen atom of the azomethine group. The thermal behaviors of the ligand and its metal complexes were studied using thermogravimetric analysis (TGA) and differential thermal analysis (DTA). The metal complexes proved to be more thermally stable than the ligand; they decomposed at 10-30 degrees C higher temperatures. Antioxidant properties of the ligand and its metal complexes (DPPH free radical scavenging, ferrous chelating and reducing power activities) were tested. Activated carbon obtained from pumpkin seed waste functionalized with ZnCl2 was used as an absorbent. Pumpkin seed hydrochar was characterized by FT-IR, SEM, TGA-DTA, BET, and XPS. In the experimental stages, the adsorption equilibrium time was determined as 45 minutes, the adsorbent dosage was 0.8 g and the optimum pH was 6.0. After this step, the adsorption parameters of Langmuir, Freundlich, Temkin, and Dubinin-Radushkevich isotherms were investigated. It has been pointed out that the adsorption process fits better with the Freundlich isotherm model, and the adhesion occurs in a multilayered manner and on a heterogeneous surface. Freundlich and Dubinin-Radushkevich isotherms support that the bonding mechanism is realized by physical interactions. When the kinetic data were evaluated, adsorption mechanism was found to be compatible with the pseudo-second-order kinetic model. The thermodynamic parameters of adsorption indicate that the system is endothermic, and the adsorption of reactive blue 19 on activated carbon is a spontaneous process.Article Citation - WoS: 10Citation - Scopus: 10Role of 2.4 Ghz Radiofrequency Radiation Emitted From Wi-Fi on Some Mirna and Faty Acids Composition in Brain(Taylor & Francis inc, 2022) Kızmaz, Veysi; Akdag, Mehmet Zulkuf; Bashan, Mehmet; Kizmaz, Veysi; Erdal, Nurten; Erdal, Mehmet Emin; Yegin, Korkut; Department of Medical Services and Techniques / Tıbbi Hizmetler ve Teknikleri BölümüThe purpose of this study is to investigate the effects of 2.4 GHz Wi-Fi exposure, which is continuously used in the internet connection by mobile phones, computers and other wireless equipment, on microRNA and membrane and depot fatty acid composition of brain cells. Sixteen Wistar Albino rats were divided equally into two groups such as sham and exposure. The rats in the experimental group (n = 8) were exposed to 2.4 GHz RFR emitted from a Wi-Fi generator for 24 h/day for one year. The animals in the control group (n = 8) were kept under the same conditions as the experimental group, but the Wi-Fi generator was turned off. At the end of the study, rats were sacrificed and brains were removed to analyze miRNA expression and membrane and depot fatty acids of brain cells. We analyzed the situation of ten different miRNA expressions and nineteen fatty acid patterns in this study. We observed that long-term and excessive exposure of 2.4 GHz Wi-Fi radiation increased rno-miR-181a-5p, phosphatidylserine (PS) and triacylglycerol (TAG) in the brain. In conclusion, 2.4 GHz Wi-Fi exposure has the potential to alter rno-miR-181a-5p expression and the fatty acid percentage of some membrane lipids such as phospholipid (PL), phosphatidylserine (PS) and triacylglycerol (TAG), which are depot fats in the brain. La(III), Th(IV), U(IV) and Ce(III) were subjected to solid-phase extraction procedure. We observed that La(III) was selectively preconcentrated, while other ions remained in solution at pH 6.0. 5.0mL of 1.0molL(-1) HCl was used to elaute La(III) from column. 250mg of C. versicolor loaded on 1000mg of XAD-4 was optimised as solid-phase matrix. Concentrations of ions in solutions were determined by inductively coupled plasma- optical emission spectrometry (ICP-OES). The calibration plot after preconcentration was linear in the range from 1.0 to 50.0ngmL(-1) for La(III). Limit of detection was found as 0.27ngmL(-1) for La(III) by SPE method. Relative standard deviation was found lower than 6.7% for 1.0ngmL(-1) of La(III) solution (n=10). The sensitivity of ICP-OES was improved by a factor of 46.8. Metallophthalocyanine complexes ( 4 { 7 ) were obtained by cyclotetramerization of 7-tert-butyldibenzo [b,e] [1,4] dioxine-2,3-dicarbonitrile. All compounds were characterized by elemental analysis and other spectroscopic methods (IR, UV/Vis, and 1 H NMR). Phthalocyanine compounds remained nonaggregated in tetrahydrofuran at the studied concentration ranges. Metallophthalocyanines ( 4 { 7 ) were tested for their antioxidant activities. The antioxidant activity processes included evaluation of radical-scavenging activity, chelat- ing activity, and reducing power. These compounds were compared to standard antioxidant ascorbic acid. Silver nanoparticles have been successfully synthesized by environmentally friendly and economical methods. Characterization of synthesized AgNPs used UV-visible spectroscopy (UV-Vis.), thermogravimetric and differential thermal analysis (TGA-DTA), X-ray diffraction spectroscopy (XRD), scanning electron microscopy (SEM), electron diffraction X-ray (EDX), Fourier transform infrared spectroscopy (FT-IR). XRD results demonstrated that the crystal structure of AgNPs was cubic and had a size of 27.30 nm. AgNPs were found to be spherical in SEM images. The element composition was also revealed by EDX Antimicrobial activity of silver nanoparticles; Gram-negative Escherichia coli was tested on gram positive Staphylococcus aureus and Candida albicans yeast. The concentration values of 0.0552, 0.1535 and 0.0383 mg L-1 were determined as MIC (minimum inhibitory concentration), respectively. It was evaluated that the commercial antibiotics used were more effective at low concentrations compared to colistin, vancomycin and fluconazole.
